    The Microtubule-Targeting Agent Pretubulysin Impairs the Inflammatory Response in Endothelial Cells by a JNK-Dependent Deregulation of the Histone Acetyltransferase Brd4

    The anti-inflammatory effects of depolymerizing microtubule-targeting agents on leukocytes are known for a long time, but the potential involvement of the vascular endothelium and the underlying mechanistic basis is still largely unclear. Using the recently synthesized depolymerizing microtubule-targeting agent pretubulysin, we investigated the antiinflammatory potential of pretubulysin and other microtubule-targeting agents with respect to the TNF-induced leukocyte adhesion cascade in endothelial cells, to improve our understanding of the underlying biomolecular background. We found that treatment with pretubulysin reduces inflammation in vivo and in vitro via inhibition of the TNF-induced adhesion of leukocytes to the vascular endothelium by down-regulation of the pro-inflammatory cell adhesion molecules ICAM-1 and VCAM-1 in a JNK-dependent manner. The underlying mechanism includes JNK-induced deregulation and degradation of the histone acetyltransferase Bromodomaincontaining protein 4. This study shows that depolymerizing microtubule-targeting agents, in addition to their established effects on leukocytes, also significantly decrease the inflammatory activation of vascular endothelial cells. These effects are not based on altered pro-inflammatory signaling cascades, but require deregulation of the capability of cells to enter constructive transcription for some genes, setting a baseline for further research on the prominent antiinflammatory effects of depolymerizing microtubule-targeting agents

    Head and Neck Squamous-Cell cancer and its association with polymorphic enzymes of xenobiotic metabolism and repair

    Tobacco smoking, alcohol drinking, and occupational exposures to polycyclic aromatic hydrocarbons are the major proven risk factors for human head and neck squamous-cell cancer ( HNSCC). Major research focus on gene-environment interactions concerning HNSCC has been on genes encoding enzymes of metabolism for tobacco smoke constituents and repair enzymes. To investigate the role of genetically determined individual predispositions in enzymes of xenobiotic metabolism and in repair enzymes under the exogenous risk factor tobacco smoke in the carcinogenesis of HNSCC, we conducted a case-control study on 312 cases and 300 noncancer controls. We focused on the impact of 22 sequence variations in CYP1A1, CYP1B1, CYP2E1, ERCC2/XPD, GSTM1, GSTP1, GSTT1, NAT2, NQO1, and XRCC1. To assess relevant main and interactive effects of polymorphic genes on the susceptibility to HNSCC we used statistical models such as logic regression and a Bayesian version of logic regression. In subgroup analysis of nonsmokers, main effects in ERCC2 ( Lys751Gln) C/C genotype and combined ERCC2 ( Arg156Arg) C/A and A/A genotypes were predominant. When stratifying for smokers, the data revealed main effects on combined CYP1B1 ( Leu432Val) C/G and G/G genotypes, followed by CYP1B1 ( Leu432Val) G/G genotype and CYP2E1 ( -70G>T) G/T genotype. When fitting logistic regression models including relevant main effects and interactions in smokers, we found relevant associations of CYP1B1 ( Leu432Val) C/G genotype and CYP2E1 ( -70G>T) G/T genotype ( OR, 10.84; 95% CI, 1.64-71.53) as well as CYP1B1 ( Leu432Val) G/G genotype and GSTM1 null/null genotype ( OR, 11.79; 95% CI, 2.18-63.77) with HNSCC. The findings underline the relevance of genotypes of polymorphic CYP1B1 combined with exposures to tobacco smoke

    Integrity is a basic element of sporting competitions and a central category of sport economics. While contributions based on the New Institutional Economics ask for extensive regulations to ensure a perfect integrity, practitioners ask for a more moderate regulatory level. To solve this disaccord we follow the question of a theoretical background for regulatory interventions, in order to analyse their level of efficiency. Based on commodity theory, and focusing on subtractability and exclusion, we show that integrity has the characteristics of a common-pool resource. Thus dynamic perception replaces the notion of integrity as a binary construct and consumers are incorporated as endogenous stakeholders. Thereby we can solve the mentioned disaccord. Furthermore, based on these findings we examine both rules and modes of competitions and institutional arrangements of sport governing bodies as categories of maintenance and consumption of the common-pool resource
