    Functional specialization of the yeast Rho1 GTP exchange factors

    Rho GTPases are regulated in complex spatiotemporal patterns that may be dependent, in part at least, on the multiplicity of their GTP exchange factors (GEFs). Here, we examine the extent of and basis for functional specialization of the Rom2 and Tus1 GEFs that activate the yeast Rho1 GTPase, the ortholog of mammalian RhoA. First, we find that these GEFs selectively activate different Rho1-effector branches. Second, the synthetic genetic networks around ROM2 and TUS1 confirm very different global in vivo roles for these GEFs. Third, the GEFs are not functionally interchangeable: Tus1 cannot replace the essential role of Rom2, even when overexpressed. Fourth, we find that Rom2 and Tus1 localize differently: Rom2 to the growing bud surface and to the bud neck at cytokinesis; Tus1 only to the bud neck but in a distinct pattern. Finally, we find that these GEFs are dependent on different protein co-factors: Rom2 function and localization is largely dependent on Ack1, a SEL1 domain containing protein; Tus1 function and localization is largely dependent on the Tus1-interacting protein Ypl066w (which we name Rgl1). We have revealed a surprising level of diversity among the Rho1 GEFs that contributes another level of complexity to the spatiotemporal control of Rho1

    Individual correlates of podoconiosis in areas of varying endemicity: a case-control study

    BACKGROUND Podoconiosis is a non-filarial form of elephantiasis resulting in lymphedema of the lower legs. Previous studies have suggested that podoconiosis arises from the interplay of individual and environmental factors. Here, our aim was to understand the individual-level correlates of podoconiosis by comparing 460 podoconiosis-affected individuals and 707 unaffected controls. METHODS/PRINCIPAL FINDINGS This was a case-control study carried out in six kebeles (the lowest governmental administrative unit) in northern Ethiopia. Each kebele was classified into one of three endemicity levels: 'low' (prevalence 5%). A total of 142 (30.7%) households had two or more cases of podoconiosis. Compared to controls, the majority of the cases, especially women, were less educated (OR = 1.7, 95% CI = 1.3 to 2.2), were unmarried (OR = 3.4, 95% CI = 2.6-4.6) and had lower income (t = -4.4, p<0.0001). On average, cases started wearing shoes ten years later than controls. Among cases, age of first wearing shoes was positively correlated with age of onset of podoconiosis (r = 0.6, t = 12.5, p<0.0001). Among all study participants average duration of shoe wearing was less than 30 years. Between both cases and controls, people in 'high' and 'medium' endemicity kebeles were less likely than people in 'low' endemicity areas to 'ever' have owned shoes (OR = 0.5, 95% CI = 0.4-0.7). CONCLUSIONS Late use of shoes, usually after the onset of podoconiosis, and inequalities in education, income and marriage were found among cases, particularly among females. There were clustering of cases within households, thus interventions against podoconiosis will benefit from household-targeted case tracing. Most importantly, we identified a secular increase in shoe-wearing over recent years, which may give opportunities to promote shoe-wearing without increasing stigma among those at high risk of podoconiosis

    Artifacts at 4.5 and 8.0 um in Short Wavelength Spectra from the Infrared Space Observatory

    Spectra from the Short Wavelength Spectrometer (SWS) on ISO exhibit artifacts at 4.5 and 8 um. These artifacts appear in spectra from a recent data release, OLP 10.0, as spurious broad emission features in the spectra of stars earlier than ~F0, such as alpha CMa. Comparison of absolutely calibrated spectra of standard stars to corresponding spectra from the SWS reveals that these artifacts result from an underestimation of the strength of the CO and SiO molecular bands in the spectra of sources used as calibrators by the SWS. Although OLP 10.0 was intended to be the final data release, these findings have led to an additional release addressing this issue, OLP 10.1, which corrects the artifacts.Comment: 14 pages, AASTex, including 5 figures.     Boeing 747 aircraft with large external pod for transporting outsize cargo

    The effect on structural arrangement, system weight, and range performance of the cargo pod payload carrying capability was determined to include either the bridge launcher or a spacelab module on a Boeing 747 aircraft. Modifications to the carrier aircraft and the installation time required to attach the external pod to the 747 were minimized. Results indicate that the increase in pod size was minimal, and that the basic 747 structure was adequate to safely absorb the load induced by ground or air operation while transporting either payload

    A Mid-Infrared Galaxy Atlas (MIGA)

    A mid-infrared atlas of part of the Galactic plane (75<l<148,b=±675^\circ < l < 148^\circ, b = \pm6^\circ) has been constructed using HIRES processed infrared data to provide a mid-infrared data set for the Canadian Galactic Plane Survey (CGPS). The addition of this data set to the CGPS will enable the study of the emission from the smallest components of interstellar dust at an angular resolution comparable to that of the radio, millimetre, and far-infrared data in the CGPS. The Mid-Infrared Galaxy Atlas (MIGA) is a mid-infrared (12 μ\mum and 25 μ\mum) counterpart to the far-infrared IRAS Galaxy Atlas (IGA), and consists of resolution enhanced (0.5\sim 0.5' resolution) HIRES images along with ancillary maps. This paper describes the processing and characteristics of the atlas, the cross-beam simulation technique used to obtain high-resolution ratio maps, and future plans to extend both the IGA and MIGA.Comment: 38 pages (including 15 tables), 13 figures (8 dithered GIF and 5 EPS).
