    The development of episodic foresight in preschoolers

    How does the ability to think and plan into the future develop? Previous studies suggest that the ability to think about and act upon the future, a process referred to as episodic foresight, emerges between the ages of 3 and 4 (Atance, 2008). However, it is unclear what underlying processes change during the development of episodic foresight. We hypothesize that episodic foresight consists of two separate processes: 1) the formation and maintenance of goals, and 2) the construction of simulated scenarios, each of which can be made more or less difficult based on the task at hand. We report an experiment that tested the emergence of these processes. The experiment focused primarily on the effect of the number of features that must be constructed and held in memory (4 or 6) and the goals that must be maintained (Subgoal then Final). The results indicate that fouryear- old children are able to envision the future to successfully accomplish future goals, but are subject to working memory demands when there are more features in the future environment. However, three-year-olds are only able to attribute goals when the feature demands are low. When there are fewer features to construct, three-year-olds maintain only the final goal, ignoring the subgoal.     ADP-induced TXA2 generation requires the costimulation of P2Y1, P2Y12, and the GPIIb/IIIa receptors. Signaling events downstream of the P2Y receptors that contribute to ADP-induced TXA2 generation have not been clearly delineated. In this study, we have investigated the role of G-protein–gated inwardly rectifying potassium channels (GIRKs), a recently identified functional effector for the P2Y12 receptor, in the regulation of ADP-induced TXA2 generation. At 10-μM concentrations, the 2 structurally distinct GIRK channel blockers, SCH23390 and U50488H, caused complete inhibition of ADP-induced cPLA2 phosphorylation and TXA2 generation, without affecting the conversion of AA to TXA2 or ADP-induced primary platelet aggregation in aspirin-treated platelets. In addition, Src family kinase selective inhibitors abolished 2MeSADP-mediated cPLA2 phosphorylation and TXA2 generation. Furthermore, these GIRK channel blockers completely blocked Gi-mediated Src kinase activation, suggesting that GIRK channels are upstream of Src family tyrosine kinase activation. In weaver mouse platelets, which have dysfunctional GIRK2 subunits, ADP-induced TXA2 generation was impaired. However, we did not observe any defect in 2MeSADP-induced platelet functional responses in GIRK2-null mouse platelets, suggesting that functional channels composed of other GIRK subunits contribute to ADP-induced TXA2 generation, via the regulation of the Src and cPLA2 activity

    Memory-related hippocampal activation in the sleeping toddler

    Nonhuman research has implicated developmental processes within the hippocampus in the emergence and early development of episodic memory, but methodological challenges have hindered assessments of this possibility in humans. Here, we delivered a previously learned song and a novel song to 2-year-old toddlers during natural nocturnal sleep and, using functional magnetic resonance imaging, found that hippocampal activation was stronger for the learned song compared with the novel song. This was true regardless of whether the song was presented intact or backwards. Toddlers who remembered where and in the presence of which toy character they heard the song exhibited stronger hippocampal activation for the song. The results establish that hippocampal activation in toddlers reflects past experiences, persists despite some alteration of the stimulus, and is associated with behavior. This research sheds light on early hippocampal and memory functioning and offers an approach to interrogate the neural substrates of early memory
