Audit 2017: how democratic is local government in Northern Ireland?
Local authorities play key roles in the devolved government of Northern Ireland, as expressions of communities that were in the past highly polarised on religious and political lines. They are also the only other source of elected legitimacy to the Northern Ireland Assembly and Executive, and can act as checks and balances on the domestic concentration of power. Natural ventilation in double-skin façade design for office buildings in hot and humid climate
This research seeks to find a design solution for reducing the energy usage in high-rise office buildings in Singapore. There are numerous methods and techniques that could be employed to achieve the purpose of designing energy efficient buildings. The Thesis explores the viability of double-skin façades (DSF) to provide natural ventilation as an energy efficient solution for office buildings in hot and humid environment by using computational fluid dynamic (CFD) simulations and case study methodologies.
CFD simulations were used to examine various types of DSF used in office buildings and the behaviour of airflow and thermal transfer through the DSF; the internal thermal comfort levels of each office spaces were analyzed and compared; and an optimization methodology was developed to explore the best DSF configuration to be used in high-rise office buildings in the tropics. The correlation between the façade configurations, the thermal comfort parameters, and the internal office space energy consumption through the DSF is studied and presented.
The research outcome of the Thesis has found that significant energy saving is possible if natural ventilation strategies could be exploited with the use of DSF. A prototype DSF configuration which will be best suited for the tropical environment in terms of its energy efficiency through cross ventilation strategy is proposed in this Thesis. A series of comprehensive and user-friendly nomograms for design optimization in selecting the most appropriate double-skin façade configurations with considerations of various orientations for the use in high-rise office buildings in the tropics were also presented
Referendums on EU matters
This study was commissioned by the European Parliament’s Policy Department for Citizens’ Rights and Constitutional Affairs at the request of the Committee on Constitutional Affairs of the European Parliament. It analyses the political and legal dynamics behind referendums on EU-related matters. It argues that we have entered a period of increasing political uncertainty with regard to the European project and that this new political configuration will both affect and be affected by the politics of EU-related referendums. Such referendums have long been a risky endeavour and this has been accentuated in the wake of the Great Recession with its negative ramifications for public opinion in the European Union. It is clear that referendums on EU matters are here to stay and will continue to be central to the EU’s future as they are deployed to determine the number of Member States within the EU, its geographical reach, its constitutional evolution and adherence to EU policies. Only now they have become an even riskier endeavour
Microplastics in agriculture – a potential novel mechanism for the delivery of human pathogens onto crops
Mulching with plastic sheeting, the use of plastic carriers in seed coatings, and irrigation with wastewater or contaminated surface water have resulted in plastics, and microplastics, becoming ubiquitous in agricultural soils. Once in the environment, plastic surfaces quickly become colonised by microbial biofilm comprised of a diverse microbial community. This so-called ‘plastisphere’ community can also include human pathogens, particularly if the plastic has been exposed to faecal contamination (e.g., from wastewater or organic manures and livestock faeces). The plastisphere is hypothesised to facilitate the survival and dissemination of pathogens, and therefore plastics in agricultural systems could play a significant role in transferring human pathogens to crops, particularly as microplastics adhering to ready to eat crops are difficult to remove by washing. In this paper we critically discuss the pathways for human pathogens associated with microplastics to interact with crop leaves and roots, and the potential for the transfer, adherence, and uptake of human pathogens from the plastisphere to plants. Globally, the concentration of plastics in agricultural soils are increasing, therefore, quantifying the potential for the plastisphere to transfer human pathogens into the food chain needs to be treated as a priority
A Positive Feedback Synapse from Retinal Horizontal Cells to Cone Photoreceptors
Cone photoreceptors and horizontal cells (HCs) have a reciprocal synapse that
underlies lateral inhibition and establishes the antagonistic center-surround
organization of the visual system. Cones transmit to HCs through an excitatory
synapse and HCs feed back to cones through an inhibitory synapse. Here we report
that HCs also transmit to cone terminals a positive feedback signal that
elevates intracellular Ca2+ and accelerates neurotransmitter
release. Positive and negative feedback are both initiated by AMPA receptors on
HCs, but positive feedback appears to be mediated by a change in HC
Ca2+, whereas negative feedback is mediated by a change in
HC membrane potential. Local uncaging of AMPA receptor agonists suggests that
positive feedback is spatially constrained to active HC-cone synapses, whereas
the negative feedback signal spreads through HCs to affect release from
surrounding cones. By locally offsetting the effects of negative feedback,
positive feedback may amplify photoreceptor synaptic release without sacrificing
HC-mediated contrast enhancement
Height, selected genetic markers and prostate cancer risk:Results from the PRACTICAL consortium
Background: Evidence on height and prostate cancer risk is mixed, however, recent studies with large data sets support a
possible role for its association with the risk of aggressive prostate cancer.
Methods: We analysed data from the PRACTICAL consortium consisting of 6207 prostate cancer cases and 6016 controls and a
subset of high grade cases (2480 cases). We explored height, polymorphisms in genes related to growth processes as main effects
and their possible interactions.
Results: The results suggest that height is associated with high-grade prostate cancer risk. Men with height 4180cm are at a 22%
increased risk as compared to men with height o173cm (OR 1.22, 95% CI 1.01–1.48). Genetic variants in the growth pathway gene
showed an association with prostate cancer risk. The aggregate scores of the selected variants identified a significantly increased
risk of overall prostate cancer and high-grade prostate cancer by 13% and 15%, respectively, in the highest score group as
compared to lowest score group.
Conclusions: There was no evidence of gene-environment interaction between height and the selected candidate SNPs. Our
findings suggest a role of height in high-grade prostate cancer. The effect of genetic variants in the genes related to growth is
seen in all cases and high-grade prostate cancer. There is no interaction between these two exposures.</p
Multiple novel prostate cancer susceptibility signals identified by fine-mapping of known risk loci among Europeans
Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have
fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in
25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16
regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of
correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP,
while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in
Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium
(LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region.
Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant
enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the
refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa,
an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of
PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent
signals within the same regio
Effectiveness of a national quality improvement programme to improve survival after emergency abdominal surgery (EPOCH): a stepped-wedge cluster-randomised trial
Background: Emergency abdominal surgery is associated with poor patient outcomes. We studied the effectiveness of a national quality improvement (QI) programme to implement a care pathway to improve survival for these patients. Methods: We did a stepped-wedge cluster-randomised trial of patients aged 40 years or older undergoing emergency open major abdominal surgery. Eligible UK National Health Service (NHS) hospitals (those that had an emergency general surgical service, a substantial volume of emergency abdominal surgery cases, and contributed data to the National Emergency Laparotomy Audit) were organised into 15 geographical clusters and commenced the QI programme in a random order, based on a computer-generated random sequence, over an 85-week period with one geographical cluster commencing the intervention every 5 weeks from the second to the 16th time period. Patients were masked to the study group, but it was not possible to mask hospital staff or investigators. The primary outcome measure was mortality within 90 days of surgery. Analyses were done on an intention-to-treat basis. This study is registered with the ISRCTN registry, number ISRCTN80682973. Findings: Treatment took place between March 3, 2014, and Oct 19, 2015. 22 754 patients were assessed for elegibility. Of 15 873 eligible patients from 93 NHS hospitals, primary outcome data were analysed for 8482 patients in the usual care group and 7374 in the QI group. Eight patients in the usual care group and nine patients in the QI group were not included in the analysis because of missing primary outcome data. The primary outcome of 90-day mortality occurred in 1210 (16%) patients in the QI group compared with 1393 (16%) patients in the usual care group (HR 1·11, 0·96–1·28). Interpretation: No survival benefit was observed from this QI programme to implement a care pathway for patients undergoing emergency abdominal surgery. Future QI programmes should ensure that teams have both the time and resources needed to improve patient care.
