Autoantibodies against type I IFNs in patients with critical influenza pneumonia

In an international cohort of 279 patients with hypoxemic influenza pneumonia, we identified 13 patients (4.6%) with autoantibodies neutralizing IFN-alpha and/or -omega, which were previously reported to underlie 15% cases of life-threatening COVID-19 pneumonia and one third of severe adverse reactions to live-attenuated yellow fever vaccine. Autoantibodies neutralizing type I interferons (IFNs) can underlie critical COVID-19 pneumonia and yellow fever vaccine disease. We report here on 13 patients harboring autoantibodies neutralizing IFN-alpha 2 alone (five patients) or with IFN-omega (eight patients) from a cohort of 279 patients (4.7%) aged 6-73 yr with critical influenza pneumonia. Nine and four patients had antibodies neutralizing high and low concentrations, respectively, of IFN-alpha 2, and six and two patients had antibodies neutralizing high and low concentrations, respectively, of IFN-omega. The patients' autoantibodies increased influenza A virus replication in both A549 cells and reconstituted human airway epithelia. The prevalence of these antibodies was significantly higher than that in the general population for patients 70 yr of age (3.1 vs. 4.4%, P = 0.68). The risk of critical influenza was highest in patients with antibodies neutralizing high concentrations of both IFN-alpha 2 and IFN-omega (OR = 11.7, P = 1.3 x 10(-5)), especially those <70 yr old (OR = 139.9, P = 3.1 x 10(-10)). We also identified 10 patients in additional influenza patient cohorts. Autoantibodies neutralizing type I IFNs account for similar to 5% of cases of life-threatening influenza pneumonia in patients <70 yr old

National audienceQu’est-ce que la vieillesse ? Comment vieillit-on ?

International audienceOne of the corollaries of the open access movement (OA) has been the setting upof open access journals, with the occurrence, for some of them, of publication fees (ArticleProcessing Charges) that authors must pay to publish. The aim of this study is twofold: — tocharacterize the articles produced by the researchers of the higher education and researchinstitutions of Orleans (France) in 2016 in relation to the OA (presence on the open archiveHAL, characteristics of the journals in which these articles have been published, APC theytheoretically require. . .); — to record the actual APCs paid by these matching institutions. Themain results of this study show that the hybrid model is becoming more and more popular amongpublishers (for 83.6% of the 923 articles studied, the authors had the possibility to choose thehybrid option). Regarding that hybrid option, even if it was offered to authors, it was onlychosen in 10% of cases, the number of articles published under this option accounted for 41.9%of articles published in open access. The APC expenses borne by the Orleans community arespread over only 23 articles (out of 923 published), and the actual amount spent (39,261 D ) is inthe end rather limited compared to the total amount spent for the acquisition of documentaryresources by institutions of comparable size to those of the Orleans community. The resultspresented in this study offer a useful comparison with previous works, for a better understandingof the complex problematics of OA and its resulting APCs.© 2019 Elsevier Masson SAS. All rights reserved.

Associations Between Frailty and Inflammation, Physical, and Psycho-Social Health in Older Adults: A Systematic Review

    No full text
International audienceFrailty is a complex geriatric syndrome with multifactorial associated mechanisms that need to be examined more deeply to help reverse the adverse health-related outcomes. Specific inflammatory and physical health markers have been associated with the onset of frailty, but the associations between these factors and psycho-social health outcomes seem less studied. This systematic review aimed to identify, in the same study design, the potential associations between frailty and markers of inflammation, and physical or psycho-social health. A literature search was performed from inception until March 2021 using Medline, Psycinfo, and EMBASE. Three raters evaluated the articles and selected 22 studies, using inclusion and exclusion criteria ( n = 17,373; 91.6% from community-dwelling samples). Regarding biomarkers, 95% of the included studies showed significant links between inflammation [especially the higher levels of C-reactive protein (CRP) and interleukin-6 (IL-6)], and frailty status. Approximately 86% of the included studies showed strong links between physical health decline (such as lower levels of hemoglobin, presence of comorbidities, or lower physical performance), and frailty status. At most, 13 studies among the 22 included ones evaluated psycho-social variables and mixed results were observed regarding the relationships with frailty. Results are discussed in terms of questioning the medical perception of global health, centering mostly on the physical dimension. Therefore, the development of future research studies involving a more exhaustive view of frailty and global (bio-psycho-social) health is strongly encouraged

Stereoselective Diversity-Oriented Solution and Solid-Phase Synthesis of Tetrahydroquinoline-Based Polycyclic Derivatives

    No full text
A diversity-oriented solution and solid-phase synthesis of tetrahydroquinoline-based tricyclic derivatives has been achieved from enantiomerically pure, natural product-like bicyclic scaffold. The solution synthesis of enantiopure bicyclic scaffold was developed by asymmetric hetero Michael reaction. Our approach for the synthesis of polycyclic derivatives utilized regio- and stereoselective hetero Michael reaction and ringclosing metathesis as key steps in solution and on solid phase.NRC publication: Ye
