Import 02/11/2016Import 20/01/2016Import 11/02/2016Náplní mé diplomové práce je zpracování dokumentace pro provedení stavby polyfunkčního domu v Ostravě. Objekt je navržen s ohledem na urbanistickou a architektonickou koncepci oblasti. Hlavním cílem bylo navrhnout moderní budovu, která svým provozem uspokojí potřeby obyvatel.
V objektu je umístěno wellness centrum, kadeřnické a nehtové studio (1.NP), komerční prostory (2.NP), a 3 komfortní byty (3.NP). Na řešeném pozemku je navržena i plocha pro parkování. 1.NP a 2.NP je navrženo v souladu s technickými požadavky zabezpečujícími bezbariérové užívání stavby.
Součástí diplomové práce jsou i tepelně technické posudky vybraných konstrukcí a energetický štítek obálky budovy, vyhodnocené dle ČSN 730540-2 (2011).The content of this thesis is preparing the documentation for construction of a multifunctional building in Ostrava. The building is designed with the urban and architectural concept area. The main objective was to design a modern building that their operations meet the needs of the population.
The facility is located wellness center, hairdressing and nail salon (1.NP), commercial premises (2.NP), and 3 comfortable flats (3.NP). To solve the plot is designed and space for parking. 1.NP and 2.NP is designed in accordance with the technical requirements of securing barrier-free use of the building.

Super-enhancer-based identification of a BATF3/IL-2R-module reveals vulnerabilities in anaplastic large cell lymphoma.
Anaplastic large cell lymphoma (ALCL), an aggressive CD30-positive T-cell lymphoma, comprises systemic anaplastic lymphoma kinase (ALK)-positive, and ALK-negative, primary cutaneous and breast implant-associated ALCL. Prognosis of some ALCL subgroups is still unsatisfactory, and already in second line effective treatment options are lacking. To identify genes defining ALCL cell state and dependencies, we here characterize super-enhancer regions by genome-wide H3K27ac ChIP-seq. In addition to known ALCL key regulators, the AP-1-member BATF3 and IL-2 receptor (IL2R)-components are among the top hits. Specific and high-level IL2R expression in ALCL correlates with BATF3 expression. Confirming a regulatory link, IL-2R-expression decreases following BATF3 knockout, and BATF3 is recruited to IL2R regulatory regions. Functionally, IL-2, IL-15 and Neo-2/15, a hyper-stable IL-2/IL-15 mimic, accelerate ALCL growth and activate STAT1, STAT5 and ERK1/2. In line, strong IL-2Rα-expression in ALCL patients is linked to more aggressive clinical presentation. Finally, an IL-2Rα-targeting antibody-drug conjugate efficiently kills ALCL cells in vitro and in vivo. Our results highlight the importance of the BATF3/IL-2R-module for ALCL biology and identify IL-2Rα-targeting as a promising treatment strategy for ALCL

In processing the oldest museum collections and their origins, we come up against wider
issues of acquisition sources and the usability of the documentation that has been preserved.
The history of the Náprstek Museum‘s acquisitions stretches back to the times of the
museum‘s founder, Vojta Náprstek (1826–1894), who, with his wife Josefa (1838–1907)
began to put together the collections of the travellers and collectors in his group of friends.
He also initiated their collections to a large degree. The museum‘s existing documentation,
both the oldest, kept by Josefa Náprstková herself, and the subsequent documentation, has
tried to cover all the museum‘s collections, but in the search for connections between an
object and its originator we often come up against a number of obstacles. This contribution
examines these difficulties, in connection with the existing documentation and acquisition
sources. A typical collection in this respect is that of Otakar Feistmantel (1848–1891),
a geologist and paleontologist who spent eight years in India (1875–1883). The collection
forms the basis of the Náprstek Museum‘s Indian collection and is being processed under
the National Museum‘s ”Figures of Czech Science and Culture“ academic research project

Import 05/08/2014Tématem bakalářské práce jsou technologická opatření stavby pro provádění etapy betonářských prací hrubé spodní stavby v zimním období. Provádění těchto stavebních prací v daném období přináší vyšší rizika spojená s nutností přísného dodržování předepsaných technologických postupů a opatření.
Cílem této bakalářské práce je stanovit přesný technologický postup pro provádění betonářských prací hrubé spodní stavby na daném objektu v zimním období včetně návrhu strojní sestavy a zajištění kvalitativních požadavků výsledné konstrukce. Výsledkem je porovnání finanční a ekonomické náročnosti provádění vybrané etapy v zimním období oproti provedení prací v letním období.The topic of the thesis are technological measures for the implementation of the construction phase of concreting gross substructures in winter. The implementation of these works will be shown brings increased risks associated with the need for strict compliance with the prescribed processes and measures.
The aim of this work is to establish a precise technique for implementing concrete works coarse substructures on the object during the winter season, including a proposal mechanical assembly and ensure quality requirements of the final design.
