    Skin-derived dendritic cells acquire and degrade the scrapie agent following in vitro exposure

    The accumulation of the scrapie agent in lymphoid tissues following inoculation via the skin is critical for efficient neuroinvasion, but how the agent is initially transported from the skin to the draining lymph node is not known. Langerhans cells (LCs) are specialized antigen-presenting cells that continually sample their microenvironment within the epidermis and transport captured antigens to draining lymph nodes. We considered LCs probable candidates to acquire and transport the scrapie agent after inoculation via the skin. XS106 cells are dendritic cells (DCs) isolated from mouse epidermis with characteristics of mature LC cells. To investigate the potential interaction of LCs with the scrapie agent XS106 cells were exposed to the scrapie agent in vitro. We show that XS106 cells rapidly acquire the scrapie agent following in vitro exposure. In addition, XS106 cells partially degrade the scrapie agent following extended cultivation. These data suggest that LCs might acquire and degrade the scrapie agent after inoculation via the skin, but data from additional experiments demonstrate that this ability could be lost in the presence of lipopolysaccharide or other immunostimulatory molecules. Our studies also imply that LCs would not undergo maturation following uptake of the scrapie agent in the skin, as the expression of surface antigens associated with LC maturation were unaltered following exposure. In conclusion, although LCs or DCs have the potential to acquire the scrapie agent within the epidermis our data suggest it is unlikely that they become activated and stimulated to transport the agent to the draining lymph node

    Consistency and specificity in burglars who commit prolific residential burglary: Testing the core assumptions underpinning behavioural crime linkage

    PURPOSE: Behavioural crime linkage is underpinned by two assumptions: (a) that offenders exhibit some degree of consistency in the way they commit offences (their modus operandi [MO]); and, (b) that offenders can be differentiated on the basis of their offence behaviour. The majority of existing studies sample at most three crimes from an offender's series of detected crimes and do not examine whether patterns differ across offenders. Here, we examine patterns observed across the entire detected series of each sampled offender, and assess how homogeneous patterns are across offenders. METHODS: Using a non-parametric resampling approach, we analyse the entire crime series of 153 prolific burglars to determine if they exhibit consistency and specificity in the way they commit offences. RESULTS: Findings suggest that offenders exhibit consistency in the way they commit offences. With respect to specificity, our results suggest that patterns are not homogeneous across offenders or the type of MO considered - some offenders exhibit more specificity than do others, and offenders are more distinctive for some aspects of their MO (particularly spatial choices) than they are for others. CONCLUSIONS: The findings provide support for the underlying principles of crime linkage, but suggest that some aspects of an offender's MO either conform to a common preference, or are perhaps more influenced by situational factors than stable scripted preferences. That some offenders fail to demonstrate sufficient specificity for accurate linkage suggests that identifying which crimes are likely to be the work of offenders who display more specificity a priori constitutes one challenge for future research of this kind

    Fire Ant Decapitating Fly Cooperative Release Programs (1994–2008): Two Pseudacteon Species, P. tricuspis and P. curvatus, Rapidly Expand Across Imported Fire Ant Populations in the Southeastern United States

    Natural enemies of the imported fire ants, Solenopsis invicta Buren S. richteri Forel (Hymenoptera: Formicidae), and their hybrid, include a suite of more than 20 fire ant decapitating phorid flies from South America in the genus Pseudacteon. Over the past 12 years, many researchers and associates have cooperated in introducing several species as classical or self-sustaining biological control agents in the United States. As a result, two species of flies, Pseudacteon tricuspis Borgmeier and P. curvatus Borgmeier (Diptera: Phoridae), are well established across large areas of the southeastern United States. Whereas many researchers have published local and state information about the establishment and spread of these flies, here distribution data from both published and unpublished sources has been compiled for the entire United States with the goal of presenting confirmed and probable distributions as of the fall of 2008. Documented rates of expansion were also used to predict the distribution of these flies three years later in the fall of 2011. In the fall of 2008, eleven years after the first successful release, we estimate that P. tricuspis covered about 50% of the fire ant quarantined area and that it will occur in almost 65% of the quarantine area by 2011. Complete coverage of the fire ant quarantined area will be delayed or limited by this species' slow rate of spread and frequent failure to establish in more northerly portions of the fire ant range and also, perhaps, by its preference for red imported fire ants (S. invicta). Eight years after the first successful release of P. curvatus, two biotypes of this species (one biotype occurring predominantly in the black and hybrid imported fire ants and the other occurring in red imported fire ants) covered almost 60% of the fire ant quarantined area. We estimate these two biotypes will cover almost 90% of the quarantine area by 2011 and 100% by 2012 or 2013. Strategic selection of several distributional gaps for future releases will accelerate complete coverage of quarantine areas. However, some gaps may be best used for the release of additional species of decapitating flies because establishment rates may be higher in areas without competing species

    Polycipiviridae: a proposed new family of polycistronic picorna-like RNA viruses

    Solenopsis invicta virus 2 is a single-stranded positive-sense picorna-like RNA virus with an unusual genome structure. The monopartite genome of approximately 11 kb contains four open reading frames in its 5′ one third, three of which encode proteins with homology to picornavirus-like jelly-roll fold capsid proteins. These are followed by an intergenic region, and then a single long open reading frame that covers the 3′ two thirds of the genome. The polypeptide translation of this 3′ open reading frame contains motifs characteristic of picornavirus-like helicase, protease and RNA-dependent RNA polymerase domains. Inspection of public transcriptome shotgun assembly sequences revealed five related apparently nearly complete virus genomes isolated from ant species and one from a dipteran insect. By high-throughput sequencing and in silico assembly of RNA isolated from Solenopsis invicta and four other ant species, followed by targeted Sanger sequencing, we obtained nearly complete genomes for four further viruses in the group. Four further sequences were obtained from a recent large-scale invertebrate virus study. The 15 sequences are highly divergent (pairwise amino acid identities as low as 17% in the non-structural polyprotein), but possess the same overall polycistronic genome structure distinct from all other characterized picorna-like viruses. Consequently we propose the formation of a new virus family, Polycipiviridae, to classify this clade of arthropod-infecting polycistronic picorna-like viruses. We further propose that this family be divided into three genera: Chipolycivirus (2 species), Hupolycivirus (2 species), and Sopolycivirus (11 species), with members of the latter infecting ants in at least three different subfamilies.This work was supported by a Wellcome Trust grant [106207] and a European Research Council (ERC) European Union's Horizon 2020 research and innovation programme grant [646891] to A.E.F

    Cytogenetic analysis of three species of Pseudacteon (Diptera, Phoridae) parasitoids of the fire ants using standard and molecular techniques

    Pseudacteon flies, parasitoids of worker ants, are being intensively studied as potentially effective agents in the biological control of the invasive pest fire ant genus Solenopsis (Hymenoptera: Formicidae). This is the first attempt to describe the karyotype of P. curvatus Borgmeier, P. nocens Borgmeier and P. tricuspis Borgmeier. The three species possess 2n = 6; chromosomes I and II were metacentric in the three species, but chromosome pair III was subtelocentric in P. curvatus and P. tricuspis, and telocentric in P. nocens. All three species possess a C positive band in chromosome II, lack C positive heterochromatin on chromosome I, and are mostly differentiated with respect to chromosome III. P. curvatus and P. tricuspis possess a C positive band, but at different locations, whereas this band is absent in P. nocens. Heterochromatic bands are neither AT nor GC rich as revealed by fluorescent banding. In situ hybridization with an 18S rDNA probe revealed a signal on chromosome II in a similar location to the C positive band in the three species. The apparent lack of morphologically distinct sex chromosomes is consistent with proposals of environmental sex determination in the genus. Small differences detected in chromosome length and morphology suggests that chromosomes have been highly conserved during the evolutionary radiation of Pseudacteon.     Characterizing Interdisciplinarity of Researchers and Research Topics Using Web Search Engines

    Researchers' networks have been subject to active modeling and analysis. Earlier literature mostly focused on citation or co-authorship networks reconstructed from annotated scientific publication databases, which have several limitations. Recently, general-purpose web search engines have also been utilized to collect information about social networks. Here we reconstructed, using web search engines, a network representing the relatedness of researchers to their peers as well as to various research topics. Relatedness between researchers and research topics was characterized by visibility boost-increase of a researcher's visibility by focusing on a particular topic. It was observed that researchers who had high visibility boosts by the same research topic tended to be close to each other in their network. We calculated correlations between visibility boosts by research topics and researchers' interdisciplinarity at individual level (diversity of topics related to the researcher) and at social level (his/her centrality in the researchers' network). We found that visibility boosts by certain research topics were positively correlated with researchers' individual-level interdisciplinarity despite their negative correlations with the general popularity of researchers. It was also found that visibility boosts by network-related topics had positive correlations with researchers' social-level interdisciplinarity. Research topics' correlations with researchers' individual- and social-level interdisciplinarities were found to be nearly independent from each other. These findings suggest that the notion of "interdisciplinarity" of a researcher should be understood as a multi-dimensional concept that should be evaluated using multiple assessment means.Comment: 20 pages, 7 figures.     The Nest Architecture of Three Species of North Florida Aphaenogaster Ants

    The architecture of the subterranean nests of Aphaenogaster floridana Smith (Hymenoptera: Formicidae), A. treatae Forel and A. ashmeadi (Emery), was studied from plaster, wax, or metal casts. After structural features were quantified from digital images, the entombed ants were retrieved from the plaster by dissolution or wax casts by melting and counted. Nests of all three species were rather simple, small and vertical, with horizontal chambers connected by vertical shafts. Shafts descending to lower chambers tended to arise from chamber edges, whereas those connecting to a chamber above tended to arise from chamber centers. A. floridana had the largest nests and colonies, and multiple shafts commonly connected upper chambers, a feature lacking in the other two species. In A. floridana nests a higher proportion of chamber area and greater spacing between chambers occurred in the deeper parts of the nest, regardless of nest size. The other two species showed no vertical differentiation of any size-free measure at any nest size. In all three species, nest size increased more slowly than the worker population, so crowding was greater in large colonies than in small, in contrast to the situation in three other ant species for which data were available.     Plasma cholesterol levels and brain development in preterm newborns.

    BackgroundTo assess whether postnatal plasma cholesterol levels are associated with microstructural and macrostructural regional brain development in preterm newborns.MethodsSixty preterm newborns (born 24-32 weeks gestational age) were assessed using MRI studies soon after birth and again at term-equivalent age. Blood samples were obtained within 7 days of each MRI scan to analyze for plasma cholesterol and lathosterol (a marker of endogenous cholesterol synthesis) levels. Outcomes were assessed at 3 years using the Bayley Scales of Infant Development, Third Edition.ResultsEarly plasma lathosterol levels were associated with increased axial and radial diffusivities and increased volume of the subcortical white matter. Early plasma cholesterol levels were associated with increased volume of the cerebellum. Early plasma lathosterol levels were associated with a 2-point decrease in motor scores at 3 years.ConclusionsHigher early endogenous cholesterol synthesis is associated with worse microstructural measures and larger volumes in the subcortical white matter that may signify regional edema and worse motor outcomes. Higher early cholesterol is associated with improved cerebellar volumes. Further work is needed to better understand how the balance of cholesterol supply and endogenous synthesis impacts preterm brain development, especially if these may be modifiable factors to improve outcomes

    The Seasonal Natural History of the Ant, Dolichoderus mariae, in Northern Florida

    Dolichoderus mariae Forel, (Hymenoptera: Formicidae) is an uncommon, monomorphic but locally abundant, reddish-brown ant of peculiar nesting habits, whose range includes most of the eastern USA. In north Florida the ant excavates soil under wiregrass clumps or other plants with fibrous roots to form a single, large, shallow, conical or ovoid chamber broadly open to the surface around the plant base. Colonies are highly polygyne and, during the warm season, inhabit multiple nests connected only by above ground trails, over which nests exchange workers. Although monomorphic, worker size may differ significantly between colonies. The colony cycle is dominated by strong seasonal polydomy. From one or two over-wintering nests, the colonies expanded to occupy up to 60 nests by late summer, then retract once more to one or two nests by mid-winter. The worker-to-queen ratio changed greatly during this cycle, with over two thousand workers per queen during fall and winter, dropping to a low of about 300 during midsummer. Most of these summer queens probably die during the fall. Colonies reoccupy roughly the same area year to year even though they contract down to one or two nests in winter. Observation of fights in the contact zone between colonies suggested that the colonies are territorial. The ants subsist by tending aphids and scale insects for honeydew and scavenging for dead insects within their territories
