    Time for Foucault? Reflections on the Roman Self from Seneca to Augustine

    The essay approaches the idea of the self as this was most often formulated in antiquity from Heraclitus to Augustine—not as the object of self-fashioning and self-care, but as an irresolvable problem that was a productive if disconcerting source of inquiry. The self is less cultivated than it is “unbounded,” less wedded to regimes of truth and discovery than it is exposed, precariously, to crises of identity and coherence in the face of a constantly changing and unfathomable world. The self on this view of it does not conform to the accounts that are given by Foucault, Hadot, or Gill. Readings of Marcus Aurelius, Seneca, and Augustine are used to support this first attempt at an alternative picture of the self in antiquity

    I magine you are the warden of a prison or the administrator of a jail, you enter the facility, and the heavy metal door clangs shut behind you. A riot breaks out and several members of your staff are taken hostage. Fires are set in various locations. The rioting inmates, members of a gang, attack other inmates who want no part in the rebellion and are only concerned for their safety. The decisions you as a leader make are literally matters of life or death, not only for yourself, but also for your staff and those incarcerated. The dynamics of power and guarding against potential corruption are also a constant concern. The leader of an incarceration facility must account for and respond to public opinion and political demands, as well as the individual needs and group dynamics of corrections officers and inmates. Some of the desired outcomes for leaders in a confinement setting are a reduction in recidivism, security of the facility, prevention of escape, and efforts at rehabilitation. While some would argue that depending upon the reason for incarceration, the inmates do not deserve much more consideration than would an animal in a zoo, others-leaders-in this situation strive for much more. Accomplished leaders in a confinement setting seek to develop a culture that creates and sustains the psychological health and well-being of the corrections officers and provides inmates opportunities to develop skills and their potential. After all, they are in the people business. Such an approach would conceivably result in an institution that functions based on the strengths of the corrections officers and inmates, thereby allowing them the greatest opportunity for self-development, physical and psychological security, and indirectly decreased chances of recidivism.     Inhibition of HMG CoA reductase reveals an unexpected role for cholesterol during PGC migration in the mouse

    <p>Abstract</p> <p>Background</p> <p>Primordial germ cells (PGCs) are the embryonic precursors of the sperm and eggs. Environmental or genetic defects that alter PGC development can impair fertility or cause formation of germ cell tumors.</p> <p>Results</p> <p>We demonstrate a novel role for cholesterol during germ cell migration in mice. Cholesterol was measured in living tissue dissected from mouse embryos and was found to accumulate within the developing gonads as germ cells migrate to colonize these structures. Cholesterol synthesis was blocked in culture by inhibiting the activity of HMG CoA reductase (HMGCR) resulting in germ cell survival and migration defects. These defects were rescued by co-addition of isoprenoids and cholesterol, but neither compound alone was sufficient. In contrast, loss of the last or penultimate enzyme in cholesterol biosynthesis did not alter PGC numbers or position in vivo. However embryos that lack these enzymes do not exhibit cholesterol defects at the stage at which PGCs are migrating. This demonstrates that during gestation, the cholesterol required for PGC migration can be supplied maternally.</p> <p>Conclusion</p> <p>In the mouse, cholesterol is required for PGC survival and motility. It may act cell-autonomously by regulating clustering of growth factor receptors within PGCs or non cell-autonomously by controlling release of growth factors required for PGC guidance and survival.</p

    Colonic epithelial cathelicidin (LL-37) expression intensity is associated with progression of colorectal cancer and presence of CD8+ T cell infiltrate

    Colorectal cancer (CRC) remains a leading cause of cancer mortality. Here, we define the colonic epithelial expression of cathelicidin (LL-37) in CRC. Cathelicidin exerts pleotropic effects including anti-microbial and immunoregulatory functions. Genetic knockout of cathelicidin led to increased size and number of colorectal tumours in the azoxymethane-induced murine model of CRC. We aimed to translate this to human disease. The expression of LL-37 in a large (n = 650) fully characterised cohort of treatment-naĂŻve primary human colorectal tumours and 50 matched normal mucosa samples with associated clinical and pathological data (patient age, gender, tumour site, tumour stage [UICC], presence or absence of extra-mural vascular invasion, tumour differentiation, mismatch repair protein status, and survival to 18 years) was assessed by immunohistochemistry. The biological consequences of LL-37 expression on the epithelial barrier and immune cell phenotype were assessed using targeted quantitative PCR gene expression of epithelial permeability (CLDN2, CLDN4, OCLN, CDH1, and TJP1) and cytokine (IL-1ÎČ, IL-18, IL-33, IL-10, IL-22, and IL-27) genes in a human colon organoid model, and CD3+ , CD4+ , and CD8+ lymphocyte phenotyping by immunohistochemistry, respectively. Our data reveal that loss of cathelicidin is associated with human CRC progression, with a switch in expression intensity an early feature of CRC. LL-37 expression intensity is associated with CD8+ T cell infiltrate, influenced by tumour characteristics including mismatch repair protein status. There was no effect on epithelial barrier gene expression. These data offer novel insights into the contribution of LL-37 to the pathogenesis of CRC and as a therapeutic molecule

    The importance of reflection to marketing educators is increasingly recognized. However, there is a lack of empirical research which considers reflection within the context of both the marketing and general business education literature. This paper describes the use of an instrument which can be used to measure four identified levels of a reflection hierarchy: habitual action, understanding, reflection and intensive reflection and two conditions for reflection: instructor to student interaction and student to student interaction. Further we demonstrate the importance of reflective learning in predicting graduates’ perception of program quality. Although the focus was on assessment of MBA level curricula, the findings have great importance to marketing education and educators
