    Measurement of Epstein-Barr virus DNA load using a novel quantification standard containing two EBV DNA targets and SYBR Green I dye

    BACKGROUND Reactivation of Epstein-Barr virus (EBV) infection may cause serious, life-threatening complications in immunocompromised individuals. EBV DNA is often detected in EBV-associated disease states, with viral load believed to be a reflection of virus activity. Two separate real-time quantitative polymerase chain reaction (QPCR) assays using SYBR Green I dye and a single quantification standard containing two EBV genes, Epstein-Barr nuclear antigen-1 (EBNA-1) and BamHI fragment H rightward open reading frame-1 (BHRF-1), were developed to detect and measure absolute EBV DNA load in patients with various EBV-associated diseases. EBV DNA loads and viral capsid antigen (VCA) IgG antibody titres were also quantified on a population sample. RESULTS EBV DNA was measurable in ethylenediaminetetraacetic acid (EDTA) whole blood, peripheral blood mononuclear cells (PBMCs), plasma and cerebrospinal fluid (CSF) samples. EBV DNA loads were detectable from 8.0 × 10² to 1.3 × 10⁸ copies/ml in post-transplant lymphoproliferative disease (n = 5), 1.5 × 10³ to 2.0 × 10⁵ copies/ml in infectious mononucleosis (n = 7), 7.5 × 10⁴ to 1.1 × 10⁵ copies/ml in EBV-associated haemophagocytic syndrome (n = 1), 2.0 × 10² to 5.6 × 10³ copies/ml in HIV-infected patients (n = 12), and 2.0 × 10² to 9.1 × 10⁴ copies/ml in the population sample (n = 218). EBNA-1 and BHRF-1 DNA were detected in 11.0% and 21.6% of the population sample respectively. There was a modest correlation between VCA IgG antibody titre and BHRF-1 DNA load (rho = 0.13, p = 0.05) but not EBNA-1 DNA load (rho = 0.11, p = 0.11). CONCLUSION Two sensitive and specific real-time PCR assays using SYBR Green I dye and a single quantification standard containing two EBV DNA targets, were developed for the detection and measurement of EBV DNA load in a variety of clinical samples.     Maximum Entropy for Gravitational Wave Data Analysis: Inferring the Physical Parameters of Core-Collapse Supernovae

    The gravitational wave signal arising from the collapsing iron core of a Type II supernova progenitor star carries with it the imprint of the progenitor's mass, rotation rate, degree of differential rotation, and the bounce depth. Here, we show how to infer the gravitational radiation waveform of a core collapse event from noisy observations in a network of two or more LIGO-like gravitational wave detectors and, from the recovered signal, constrain these source properties. Using these techniques, predictions from recent core collapse modeling efforts, and the LIGO performance during its S4 science run, we also show that gravitational wave observations by LIGO might have been sufficient to provide reasonable estimates of the progenitor mass, angular momentum and differential angular momentum, and depth of the core at bounce, for a rotating core collapse event at a distance of a few kpc.Comment: 44 pages, 12 figures; accepted version scheduled to appear in Ap J 1 April 200

    Observational analytic studies in multiple sclerosis: controlling bias through study design and conduct. The Australian Multicentre Study of Environment and Immune Function

    Rising multiple sclerosis incidence over the last 50 years and geographic patterns of occurrence suggest an environmental role in the causation of this multifactorial disease. Design options for epidemiological studies of environmental causes of multiple sclerosis are limited by the low incidence of the disease, possible diagnostic delay and budgetary constraints. We describe scientific and methodological issues considered in the development of the Australian Multicentre Study of Environment and Immune Function (the Ausimmune Study), which seeks, in particular, to better understand the causes of the well-known MS positive latitudinal gradient. A multicentre, case-control design down the eastern seaboard of Australia allows the recruitment of sufficient cases for adequate study power and provides data on environmental exposures that vary by latitude. Cases are persons with an incident first demyelinating event (rather than prevalent multiple sclerosis), sourced from a population base using a two tier notification system. Controls, matched on sex, age (within two years) and region of residence, are recruited from the general population. Biases common in case-control studies, eg, prevalence-incidence bias, admission-rate bias, non-respondent bias, observer bias and recall bias, as well as confounding have been carefully considered in the study design and conduct of the Ausimmune Study

    Investigating International Time Trends in the Incidence and Prevalence of Atopic Eczema 1990-2010: A Systematic Review of Epidemiological Studies

    The prevalence of atopic eczema has been found to have increased greatly in some parts of the world. Building on a systematic review of global disease trends in asthma, our objective was to study trends in incidence and prevalence of atopic eczema. Disease trends are important for health service planning and for generating hypotheses regarding the aetiology of chronic disorders. We conducted a systematic search for high quality reports of cohort, repeated cross-sectional and routine healthcare database-based studies in seven electronic databases. Studies were required to report on at least two measures of the incidence and/or prevalence of atopic eczema between 1990 and 2010 and needed to use comparable methods at all assessment points. We retrieved 2,464 citations, from which we included 69 reports. Assessing global trends was complicated by the use of a range of outcome measures across studies and possible changes in diagnostic criteria over time. Notwithstanding these difficulties, there was evidence suggesting that the prevalence of atopic eczema was increasing in Africa, eastern Asia, western Europe and parts of northern Europe (i.e. the UK). No clear trends were identified in other regions. There was inadequate study coverage worldwide, particularly for repeated measures of atopic eczema incidence. Further epidemiological work is needed to investigate trends in what is now one of the most common long-term disorders globally. A range of relevant measures of incidence and prevalence, careful use of definitions and description of diagnostic criteria, improved study design, more comprehensive reporting and appropriate interpretation of these data are all essential to ensure that this important field of epidemiological enquiry progresses in a scientifically robust manner

    Jet aircraft lubrication oil droplets as contrail ice-forming particles

    The radiative characteristics and lifetimes of contrails are dependent on the number concentration of ice-forming particles in the engine exhaust plume. Aircraft gas turbine engines produce a variety of particles, yet it is understood that non-volatile black carbon aggregates are the dominant source of ice-forming particles with typical, fossil-derived jet fuel. However, with cleaner combustion technologies and the adoption of alternative fuels (e.g. hydrogen or synthetic aviation fuel), non-volatile black carbon particle emissions are expected to decrease or even be eliminated. Under these conditions, contrail properties will depend upon the concentration and characteristics of particles other than black carbon. Ultrafine (&lt; 100 nm) jet lubrication oil droplets constitute a significant fraction of the total organic particulate matter released by aircraft; however, their ability to form contrail ice crystals has hitherto been unexplored. In this work, we experimentally investigate the activation and freezing behaviour of lubrication oil droplets using an expansion chamber, assessing their potential as ice-forming particles. We generate lubrication oil droplets with a geometric mean mobility diameter of (100.9 ± 0.6) nm and show that these activate to form water droplets, which subsequently freeze when the temperature is below ∼ 235 K. We find that nucleation on lubrication oil droplets should be considered in future computational studies – particularly under soot-poor conditions – and that these studies would benefit from particle size distribution measurements at cruise altitude. Overall, taking steps to reduce lubrication oil number emissions would help reduce the climate impact of contrail cirrus.</p

    Ground-based contrail observations: comparisons with reanalysis weather data and contrail model simulations

    Observations of contrails are vital for improving our understanding of the contrail formation and life cycle, informing models, and assessing mitigation strategies. Here, we developed a methodology that utilises ground-based cameras for tracking and analysing young contrails (&lt; 35 min) formed under clear-sky conditions, comparing these observations against reanalysis meteorology and simulations from the contrail cirrus prediction model (CoCiP) with actual flight trajectories. Our observations consist of 14 h of video footage recorded over 5 different days in Central London, capturing 1582 flight waypoints from 281 flights. The simulation correctly predicted contrail formation and absence for around 75 % of these waypoints, with incorrect contrail predictions occurring at warmer temperatures than those with true-positive predictions (7.8 K vs. 12.8 K below the Schmidt–Appleman criterion threshold temperature). When evaluating contrails with observed lifetimes of at least 2 min, the simulation's correct prediction rate for contrail formation increases to over 85 %. Among all waypoints with contrail observations, 78 % of short-lived contrails (observed lifetimes &lt; 2 min) formed under ice-subsaturated conditions, whereas 75 % of persistent contrails (observed lifetimes &gt; 10 min) formed under ice-supersaturated conditions. On average, the simulated contrail geometric width was around 100 m smaller than the observed (visible) width over its observed lifetime, with the mean underestimation reaching up to 280 m within the first 5 min. Discrepancies between the observed and simulated contrail formation, lifetime, and width can be associated with uncertainties in reanalysis meteorology due to known model limitations and sub-grid-scale variabilities, contrail model simplifications, uncertainties in aircraft performance estimates, and observational challenges, among other possible factors. Overall, this study demonstrates the potential of ground-based cameras to create essential observational and benchmark datasets for validating and improving existing weather and contrail models.</p

    Objectively Measured Physical Activity and the Subsequent Risk of Incident Dysglycemia: The Australian Diabetes, Obesity and Lifestyle Study (AusDiab)

    OBJECTIVE - To investigate pedometer-measured physical activity (PA) in 2000 and change in PA over 5 years with subsequent risk of dysglycemia by 2005. RESEARCH DESIGN AND METHODS - This prospective cohort study in Tasmania, Australia, analyzed 458 adults with normal glucose tolerance and a mean (SD) age of 49.7 (12.1) years in 2000. Variables assessed in 2000 and 2005 included PA, by pedometer and questionnaire, nutrient intake, and other lifestyle factors. Incident dysglycemia was defined as the development of impaired fasting glucose or impaired glucose tolerance revealed by oral glucose tolerance testing in 2005, without type 2 diabetes. RESULTS - Incident dysglycemia developed in 26 participants during the 5-year period. Higher daily steps in 2000 were independently associated with a lower 5-year risk of incident dysglycemia (adjusted odds ratio [AOR] 0.87 [95% CI 0.77-0.97] per 1,000-step increment). Higher daily steps in 2005, after controlling for baseline steps in 2000 (thus reflecting change in steps over 5 years), were not associated with incident dysglycemia (AOR 1.02 [0.92-1.14]). Higher daily steps in 2000 were also associated with lower fasting blood glucose, but not 2-h plasma glucose by 2005. Further adjustment for BMI or waist circumference did not remove these associations. CONCLUSIONS - Among community-dwelling adults, a higher rate of daily steps is associated with a reduced risk of incident dysglycemia. This effect appears to be not fully mediated through reduced adiposity
