    Dendritic Epidermal T Cells: Lessons from Mice for Humans

    Dendritic epidermal T cells (DETC) in mice form part of a primitive system of epithelial-resident T cells characterized by the expression of gd T-cell receptors (TCR). Critical attributes that characterize DETC include their highly restricted T-cell receptor gene utilization, proliferation and maturation within epidermis, a capacity to kill relevant skin-derived tumor targets, and the ability to modulate immune responses that are initiated and expressed in skin. Contemporary knowledge suggests that DETC and the related skin-directed gd T cells found in humans play important roles in maintaining the immunologic integrity of skin

    Expression of the Tumor Necrosis Factor Gene by Dermal Fibroblasts in Response to Ultraviolet Irradiation or Lipopolysaccharide

    To examine the effects of different wavelengths of ultraviolet (UV) radiation on tumor necrosis factor (TNF) production, we took advantage of mice carrying a chloramphenicol acetyl transferase (CAT) reporter transgene bearing the entire TNF promoter and 3'-untranslated region. Aside from constitutive expression in the thymus, CAT activity was detected only in locally UVB- or UVC-irradiated skin. After UVB irradiation, markedly greater amounts of CAT activity were traced to the dermis rather than the epidermis; by contrast, almost all CAT activity was localized to the epidermis after UVC irradiation, Fibroblasts have not been shown previously to express the TNF gene, i.e., the TNF gene is highly methylated and inaccessible to exogenous modulation in 3T3 fibroblasts, However, the present report reveals that cultured dermal fibroblasts are capable of producing both CAT and TNF in response to treatment in vitro with either UVB irradiation, UVC irradiation, or lipopolysaccharide. These findings indicate that dermal fibroblasts may serve not only as a target for but also as a source of TNF

Inicialmente, demonstraremos os resultados do exame das teses e das dissertações. Em seguida, refletiremos acerca dos artigos. Para a leitura e análise dos artigos e da produção discente, utilizamos como subsídio os referenciais da análise de conteúdo, por meio da análise categorial (BARDIN, 2008). Ao final, apresentaremos os caminhos abertos para pesquisas futuras e as recomendações da produção investigada para a implementação da Lei nº 10.639/2003

    Excess Insulin Binding to Insulin-Like Growth Factor Receptors: Proposed Mechanism for Acanthosis Nigricans

    Clinical and epidemiologic evidence has shown acanthosis nigricans to be closely related to defective tissue utilization of insulin in a number of previously recognized (e.g., obesity, lipodystrophy, and leprechaunism) as well as recently characterized (e.g., type A and type B syndromes) disorders. This article reviews the relationship of acanthosis nigricans to these insulin-resistant states. It also focuses attention on the possibility that interaction between excessive amounts of circulating insulin with insulin-like growth factor receptors on keratinocytes and dermal fibroblasts leads to the development of acanthosis nigricans
