    Microbiome Associated with Slovak Traditional Ewe's Milk Lump Cheese

    Worldwide consumers increasingly demand traditional/local products, to which those made from ewe's milk belong. In Slovakia, dairy products made from ewe's milk have a long tradition. A total of seventeen farmhouse fresh ewe's milk lump cheeses from various local farm producers in central Slovakia were sampled at farms and then analyzed. Based on the sequencing data analysis, the phylum Firmicutes dominated (60.92%) in ewe's lump cheeses, followed with the phylum Proteobacteria (38.23%), Actinobacteria (0.38%) and Bacteroidetes (0.35%). The phylum Firmicutes was represented by six genera, among which the highest amount possessed the genus Streptococcus (41.13%) followed with the genus Lactococcus (8.54%), Fructobacillus (3.91%), Enterococcus (3.18%), Staphylococcus (1.80%) and the genus Brochotrix (0.08%). The phylum Proteobacteria in ewe's lump cheeses involved eight Gram-negative genera: Pseudomonas, Acinetobacter, Enterobacter, Ewingella, Escherichia-Shigella, Pantoea and Moraxella. The phylum Bacteroidetes involved three genera: Bacteroides, Sphingobacterium and Chrysobacterium.     MetNH3: Metrology for Ammonia in Ambient Air

    Measuring ammonia in ambient air is a sensitive and priority issue due to its harmful effects on human health and ecosystems. Ammonia is increasingly being globally acknowledged as a key precursor to atmospheric particulate matter. The European Directive 2001/81/EC on “National Emission Ceilings for Certain Atmospheric Pollutants (NEC)” regulates ammonia emissions in the member states. However, due to the chemical characteristics of ambient ammonia traceable on-line measurements still have significant challenges in analytical technology, uncertainty, quality assurance and quality control (QC/QA). Currently the UK National Ammonia Monitoring Network uses an accredited off-line low temporal resolution and on-line denuder–IC methods at the UK Supersites. There is a need for traceable ammonia measurements which will be vitally important for identifying changes in environment policies, climate and agricultural practice. This in turn should lead to improvements emission inventory uncertainties and for providing independent verification of atmospheric model predictions. MetNH3 (EMRP Joint Research Project) has worked with SMEs in testing improved reference gas mixtures by static and dynamic gravimetric generation methods, develop and refine existing laser based optical spectrometric standards and establishing the transfer from high-accuracy standards to field applicable methods. The first results from the metrological characterisation of a commercially available cavity ring-down spectrometer (CRDS) are presented and the results from a new design “Controlled Atmosphere Test Facility (CATFAC)”, which is currently characterising the performance of diffusive samplers. The range and characteristics of instruments are discussed. The plans for a major ammonia field intercomparison in 2016 will be outlined

    Novel ATP-Independent RNA Annealing Activity of the Dengue Virus NS3 Helicase

    The flavivirus nonstructural protein 3 (NS3) bears multiple enzymatic activities and represents an attractive target for antiviral intervention. NS3 contains the viral serine protease at the N-terminus and ATPase, RTPase, and helicase activities at the C-terminus. These activities are essential for viral replication; however, the biological role of RNA remodeling by NS3 helicase during the viral life cycle is still unclear. Secondary and tertiary RNA structures present in the viral genome are crucial for viral replication. Here, we used the NS3 protein from dengue virus to investigate functions of NS3 associated to changes in RNA structures. Using different NS3 variants, we characterized a domain spanning residues 171 to 618 that displays ATPase and RNA unwinding activities similar to those observed for the full-length protein. Interestingly, we found that, besides the RNA unwinding activity, dengue virus NS3 greatly accelerates annealing of complementary RNA strands with viral or non-viral sequences. This new activity was found to be ATP-independent. It was determined that a mutated NS3 lacking ATPase activity retained full-RNA annealing activity. Using an ATP regeneration system and different ATP concentrations, we observed that NS3 establishes an ATP-dependent steady state between RNA unwinding and annealing, allowing modulation of the two opposing activities of this enzyme through ATP concentration. In addition, we observed that NS3 enhanced RNA-RNA interactions between molecules representing the ends of the viral genome that are known to be necessary for viral RNA synthesis. We propose that, according to the ATP availability, NS3 could function regulating the folding or unfolding of viral RNA structures

    Workshop on Regulatory Preparedness for Innovation in Nanotechnology

    This report summarises the presentations and discussions at the first NanoReg2 Workshop on Regulatory Preparedness for Innovation in Nanotechnology held in Ispra, Italy 5 to 6 October 2017 and attended by approximately 60 regulators, industry representatives and other stakeholders. NanoReg2 is a European Union (EU) Horizon 2020 project. At the workshop, Regulatory Preparedness was defined as the regulators' timely awareness of innovations and the regulator's actions to check whether present legislation covers all safety aspects of each innovation, including initiating revision of the legislation as appropriate. Regulatory Preparedness, and Safe-by-Design (SbD) jointly constitute the NanoReg2 Safe Innovation Approach (SIA) for developing innovative products based on nanotechnology. The workshop aimed to gather views and identify current practices in regulatory work on safety of innovative products, tools already in use or needed, and potential difficulties in implementing Regulatory Preparedness in the EU. Presentations addressed the current state of the safety of nanotechnology innovation. The viewpoints included the regulatory framework, the principles behind it and the agencies and authorities enforcing it; nanosafety research projects and their support system (e.g. the current EU Horizon 2020 Framework Programme); national nanosafety initiatives; and the development of tools, such as foresight tools and harmonised test guidelines by the OECD for data generation. The workshop served to generate ideas for achieving Regulatory Preparedness. The participants recognised that while regulators deal with the safety of innovations, only few systematic approaches to this work exist. Some innovative products may reach the market before their safety has been appropriately assessed, as illustrated by RAPEX, the Rapid Exchange of Information System. A continuous and proactive combination of interconnected activities was considered to be required for ensuring Regulatory Preparedness. Thus, anticipation, e.g. horizon scanning, was seen as important, as was communication between regulators, innovators (industry) and other stakeholders. Regulators need to become aware of innovative products under development to ensure that the legislation and methods for safety assessment are available and adequate. Innovators must be aware of regulatory requirements and their likely development. This mutual awareness helps to develop safe products and to avoid delays or other problems in obtaining market approval. Awareness can be achieved through communication, which requires trust, e.g. promoted via "trusted environments" for confidential inquiries and information sharing. Furthermore, regulators need early access to the existing information and data relevant to safety assessment of innovative products to provide timely guidance and advice to Industry as well as to develop strategies for dealing with uncertainty, e.g. by applying the precautionary principle. Regulatory Preparedness was discussed as part of the SIA, and a "road map" of actions was suggested and outlined. The workshop has thus contributed towards acceptance of implementing Regulatory Preparedness for innovation in nanotechnology through the participation of a variety of stakeholders. This paves the way for a better dialogue among stakeholders in a fast economic development cycle, where it is even more important to quickly identify emerging needs for new approaches to regulatory issues for innovationJRC.F.2-Consumer Products Safet

    Detection of structural genes for enterocins production among enterococci isolated from french traditional fermented sausages

    National audienceIn European countries, the demand for traditional food products has increased. Dry fermented sausages, mainly manufactured with pork and/or lean beef and pork fat, account for a significant part of traditional meat products. Many studies have dealt with the microbiology of traditional fermented sausages but there are few detail studies of properties of individual genera. The genus Enterococcus involved about 55 validated species. Its representants are mostly lactic acid producing; they can possess probiotic properties or produce bacteriocins (antimicrobial substances); on the other side, some strains can posess virulence factors genes. Our study has been focused on positive property-on detection of structural genes for enterocins production among enterococci isolated from French traditional fermented sausages. Enterocins are proteinaceous antimicrobial substances produced mostly by enterococci with inhibition spectrum against Gram-positive but also against Gram-negative bacteria. This study was performed in the framework of the EU project TRADISAUSAGE-QLK1-CT2002-02240; that is why also French traditional fermented meat products were analysed. Samples of products from 10 low-capacity processing units (placed in Massif-central-France) were collected. Sampling was performed from the initial meat (Z), during processing (M) and from the final product (F). Sampling and analytical methods for the microbiota analysis were based on the International Organisation for Standardisation (ISO). Enterococci were counted on M-Enterococcus agar (Merck, Germany). Fifty–three pure colonies were identified by PCR (primers) confirmed by Maldi-Tof identification system. Enterocins genes were tested by primers and PCR. Most of strains belonged to the species Enterococcus faecium, followed by E. faecalis and other not specified species. Strains survived in Oxgall bile evironment in high %. The most detected was Ent P gene (34%) followed by Ent A gene (27%) and Ent L50B (20%). The least was detected Ent B gene (19%)

    Contemporary comparison of aortic arch repair by endovascular and open surgical reconstructions

    Objective: This study analyzed total aortic arch reconstruction in a contemporary comparison of current open and endovascular repair.Methods: Endovascular (group 1) and open arch procedures (group 2) performed during 2007 to 2013 were entered in a prospective database and retrospectively analyzed. Endovascular repair (proximal landing zones 0-1), with or without a hybrid adjunct, was selected for patients with a high comorbidity profile and fit anatomy. Operations involving coverage of left subclavian artery only (zone 2 proximal landing: n = 41) and open hemiarch replacement (n = 434) were excluded. Early and midterm mortality and major complications were assessed.Results: Overall, 100 (78 men; mean age, 68 years) consecutive procedures were analyzed: 29 patients in group 2 and 71 in group 1. Seven group 1 patients were treated with branched or chimney stent graft, and 64 with partial or total debranching and straight stent graft. The 29 patients in group 2 were younger (mean age, 61.9 vs 70.3; P = .005), more frequently females (48.2% vs 11.3; P < .001) with less cardiac (6.9% vs 38.2%; P = .001), hypertensive (58.5% vs 88.4%; P =. 002), and peripheral artery (0% vs 16.2%; P = .031) disease. At 30 days, there were six deaths in group 1 and four in group 2 (8.5% vs 13.8%; odds ratio, 1.7; 95% confidence interval, 0.45-6.66; P = .47), and four strokes in group 1 and one in group 2 (odds ratio, 0.59; 95% confidence interval, 0.06-5.59; P = 1). Spinal cord ischemia occurred in two group 1 patients and in no group 2 patients. Three retrograde dissections (1 fatal) were detected in group 1. During a mean follow-up of 26.2 months, two type I endoleaks and three reinterventions were recorded in group 1 (all for persistent endoleak), and one reintervention was performed in group 2. According to Kaplan Meier estimates, survival at 4 years was 79.8% in group 1 and 69.8% in group 2 (P = .62), and freedom from late reintervention was 94.6% and 95.5%, respectively (P = .82).Conclusions: Despite the older age and a higher comorbidity profile in patients with challenging aortic arch disease suitable and selected for endovascular arch repair, no significant differences were detected in perioperative and 4-year outcomes compared with the younger patients undergoing open arch total repair. An endovascular approach might also be a valid alternative to open surgery in average-risk patients with aortic arch diseases requiring 0 to 1 landing zones, when morphologically feasible. However, larger concurrent comparison and longer follow-up are needed to confirm this hypothesis

    Effect of Sustained Administration of Thymol on Its Bioaccessibility and Bioavailability in Rabbits

    The objective of this study was the detection of thymol in rabbit plasma, tissues, large intestinal content, and faeces. Forty-eight rabbits were divided into control and experimental groups (thymol 250 mg/kg feed). Thymol was administered for 21 days and then withdrawn for 7 days. Concentration of thymol in the intestinal wall (IW) was significantly higher than in plasma (p &lt; 0.05) and liver (p &lt; 0.05); in the kidneys it was significantly higher than in plasma (p &lt; 0.05) and liver (p &lt; 0.05) during thymol addition. Thymol in IW was significantly higher than in plasma also after withdrawal (p &lt; 0.01). Significant correlation (rs = −1.000, p &lt; 0.01) between IW and plasma points to the intensive absorption of thymol from the intestine, while the correlation between plasma and liver (rs = 0.786, p &lt; 0.05) indicates intensive biotransformation and excretion processes in liver. Significant correlation between liver and kidney (rs = 0.738, p &lt; 0.05) confirms the intensive metabolism of thymol in the kidney. During the withdrawal period, thymol was detected above trace amounts only in faeces, and was significantly higher than in the colon during both periods (p &lt; 0.01). Results show intensive biotransformation of thymol in the rabbit organism
