    Speech as a pilot input medium

    The speech recognition system under development is a trainable pattern classifier based on a maximum-likelihood technique. An adjustable uncertainty threshold allows the rejection of borderline cases for which the probability of misclassification is high. The syntax of the command language spoken may be used as an aid to recognition, and the system adapts to changes in pronunciation if feedback from the user is available. Words must be separated by .25 second gaps. The system runs in real time on a mini-computer (PDP 11/10) and was tested on 120,000 speech samples from 10- and 100-word vocabularies. The results of these tests were 99.9% correct recognition for a vocabulary consisting of the ten digits, and 99.6% recognition for a 100-word vocabulary of flight commands, with a 5% rejection rate in each case. With no rejection, the recognition accuracies for the same vocabularies were 99.5% and 98.6% respectively

    APRIL:TACI axis is dispensable for the immune response to rabies vaccination.

    There is significant need to develop a single-dose rabies vaccine to replace the current multi-dose rabies vaccine regimen and eliminate the requirement for rabies immune globulin in post-exposure settings. To accomplish this goal, rabies virus (RABV)-based vaccines must rapidly activate B cells to secrete antibodies which neutralize pathogenic RABV before it enters the CNS. Increased understanding of how B cells effectively respond to RABV-based vaccines may improve efforts to simplify post-exposure prophylaxis (PEP) regimens. Several studies have successfully employed the TNF family cytokine a proliferation-inducing ligand (APRIL) as a vaccine adjuvant. APRIL binds to the receptors TACI and B cell maturation antigen (BCMA)-expressed by B cells in various stages of maturation-with high affinity. We discovered that RABV-infected primary murine B cells upregulate APRIL ex vivo. Cytokines present at the time of antigen exposure affect the outcome of vaccination by influencing T and B cell activation and GC formation. Therefore, we hypothesized that the presence of APRIL at the time of RABV-based vaccine antigen exposure would support the generation of protective antibodies against RABV glycoprotein (G). In an effort to improve the response to RABV vaccination, we constructed and characterized a live recombinant RABV-based vaccine vector which expresses murine APRIL (rRABV-APRIL). Immunogenicity testing in mice demonstrated that expressing APRIL from the RABV genome does not impact the primary antibody response against RABV G compared to RABV alone. In order to evaluate the necessity of APRIL for the response to rabies vaccination, we compared the responses of APRIL-deficient and wild-type mice to immunization with rRABV. APRIL deficiency does not affect the primary antibody response to vaccination. Furthermore, APRIL expression by the vaccine did not improve the generation of long-lived antibody-secreting plasma cells (PCs) as serum antibody levels were equivalent in response to rRABV-APRIL and the vector eight weeks after immunization. Moreover, APRIL is dispensable for the long-lived antibody-secreting PC response to rRABV vaccination as anti-RABV G IgG levels were similar in APRIL-deficient and wild-type mice six months after vaccination. Mice lacking the APRIL receptor TACI demonstrated primary anti-RABV G antibody responses similar to wild-type mice following immunization with the vaccine vector indicating that this response is independent of TACI-mediated signals. Collectively, our findings demonstrate that APRIL and associated TACI signaling is dispensable for the immune response to RABV-based vaccination

    Incorporating B cell activating factor (BAFF) into the membrane of rabies virus (RABV) particles improves the speed and magnitude of vaccine-induced antibody responses.

    B cell activating factor (BAFF) is a member of the tumor necrosis factor (TNF) superfamily of cytokines that links innate with adaptive immunity. BAFF signals through receptors on B cells, making it an attractive molecule to potentiate vaccine-induced B cell responses. We hypothesized that a rabies virus (RABV)-based vaccine displaying both antigen and BAFF on the surface of the same virus particle would target antigen-specific B cells for activation and improve RABV-specific antibody responses. To test this hypothesis, we constructed a recombinant RABV-based vector expressing virus membrane-anchored murine BAFF (RABV-ED51-mBAFF). BAFF was incorporated into the RABV particle and determined to be biologically functional, as demonstrated by increased B cell survival of primary murine B cells treated ex-vivo with RABV-ED51-mBAFF. B cell survival was inhibited by pre-treating RABV-ED51-mBAFF with an antibody that blocks BAFF functions. RABV-ED51-mBAFF also activated primary murine B cells ex-vivo more effectively than RABV as shown by significant upregulation of CD69, CD40, and MHCII on the surface of infected B cells. In-vivo, RABV-ED51-mBAFF induced significantly faster and higher virus neutralizing antibody (VNA) titers than RABV while not adversely affecting the longevity of the vaccine-induced antibody response. Since BAFF was incorporated into the virus particle and genome replication was not required for BAFF expression in-vivo, we hypothesized that RABV-ED51-mBAFF would be effective as an inactivated vaccine. Mice immunized with 250 ng/mouse of β-propriolactone-inactivated RABV-ED51-mBAFF showed faster and higher anti-RABV VNA titers compared to mice immunized with inactivated RABV. Together, this model stands as a potential foundation for exploring other virus membrane-anchored molecular adjuvants to make safer, more effective inactivated RABV-based vaccines

    Automatic speech recognition research at NASA-Ames Research Center

    A trainable acoustic pattern recognizer manufactured by Scope Electronics is presented. The voice command system VCS encodes speech by sampling 16 bandpass filters with center frequencies in the range from 200 to 5000 Hz. Variations in speaking rate are compensated for by a compression algorithm that subdivides each utterance into eight subintervals in such a way that the amount of spectral change within each subinterval is the same. The recorded filter values within each subinterval are then reduced to a 15-bit representation, giving a 120-bit encoding for each utterance. The VCS incorporates a simple recognition algorithm that utilizes five training samples of each word in a vocabulary of up to 24 words. The recognition rate of approximately 85 percent correct for untrained speakers and 94 percent correct for trained speakers was not considered adequate for flight systems use. Therefore, the built-in recognition algorithm was disabled, and the VCS was modified to transmit 120-bit encodings to an external computer for recognition

    Understanding uncertainty in temperature effects on vector-borne disease: A Bayesian approach

    Extrinsic environmental factors influence the distribution and population dynamics of many organisms, including insects that are of concern for human health and agriculture. This is particularly true for vector-borne infectious diseases, like malaria, which is a major source of morbidity and mortality in humans. Understanding the mechanistic links between environment and population processes for these diseases is key to predicting the consequences of climate change on transmission and for developing effective interventions. An important measure of the intensity of disease transmission is the reproductive number R0R_0. However, understanding the mechanisms linking R0R_0 and temperature, an environmental factor driving disease risk, can be challenging because the data available for parameterization are often poor. To address this we show how a Bayesian approach can help identify critical uncertainties in components of R0R_0 and how this uncertainty is propagated into the estimate of R0R_0. Most notably, we find that different parameters dominate the uncertainty at different temperature regimes: bite rate from 15-25^\circ C; fecundity across all temperatures, but especially \sim25-32^\circ C; mortality from 20-30^\circ C; parasite development rate at \sim15-16^\circC and again at \sim33-35^\circC. Focusing empirical studies on these parameters and corresponding temperature ranges would be the most efficient way to improve estimates of R0R_0. While we focus on malaria, our methods apply to improving process-based models more generally, including epidemiological, physiological niche, and species distribution models.Comment: 27 pages, including 1 table and 3 figure

    I propose an apparatus for handling intrasentential change in context. The standard approach has problems with sentences with multiple occurrences of the same demonstrative or indexical. My proposal involves the idea that contexts can be complex. Complex contexts are built out of (“simple”) Kaplanian contexts by ordered n-tupling. With these we can revise the clauses of Kaplan’s Logic of Demonstratives so that each part of a sentence is taken in a different component of a complex context. I consider other applications of the framework: to agentially distributed utterances (ones made partly by one speaker and partly by another); to an account of scare-quoting; and to an account of a binding-like phenomenon that avoids what Kit Fine calls “the antinomy of the variable.

    Faint Fuzzy Star Clusters in NGC1023 as Remnants of Merged Star Cluster Complexes

    In the lenticular galaxy NGC1023 a third population of globular clusters (GCs), called faint fuzzies (FFs), was discovered next to the blue and red GC populations by Larsen & Brodie. While these FFs have colors comparable to the red population, the new population is fainter, larger (R_eff > 7 pc) and, most importantly, shows clear signs of co-rotation with the galactic disk of NGC1023. We present N-body simulations verifying the hypothesis that these disk-associated FFs are related to the young massive cluster complexes (CCs) observed by Bastian et. al in M51, who discovered a mass-radius relation for these CCs. Our models have an initial configuration based on the observations from M51 and are placed on various orbits in a galactic potential derived for NGC1023. All computations end up with a stable object containing 10 to 60% of the initial CC mass after an integration time of 5 Gyr. A conversion to visual magnitudes demonstrates that the resulting objects cover exactly the observed range for FFs. Moreover, the simulated objects show projected half-mass radii between 3.6 and 13.4 pc, in good agreement with the observed FF sizes. We conclude that objects like the young massive CCs in M51 are likely progenitors of the FFs observed in NGC1023.Comment: Accepted for publication in Ap

    Orientational Defects in Ice Ih: An Interpretation of Electrical Conductivity Measurements

    We present a first-principles study of the structure and energetics of Bjerrum defects in ice Ih and compare the results to experimental electrical conductivity data. While the DFT result for the activation energy is in good agreement with experiment, we find that its two components have quite different values. Aside from providing new insight into the fundamental parameters of the microscopic electrical theory of ice, our results suggest the activity of traps in doped ice in the temperature regime typically assumed to be controlled by the free migration of L defects.Comment: 4 pages, 4 Figures, 1 Tabl

    Envelope Structure of Starless Core L694-2 Derived from a Near-Infrared Extinction Map

    We present a near-infrared extinction study of the dark globule L694-2, a starless core that shows strong evidence for inward motions in molecular line profiles. The J,H, and K band data were taken using the European Southern Observatory New Technology Telescope. The best fit simple spherical power law model has index p=2.6 +/- 0.2, over the 0.036--0.1 pc range in radius sampled in extinction. This power law slope is steeper than the value of p=2 for a singular isothermal sphere, the initial condition of the inside-out model for protostellar collapse. Including an additional extinction component along the line of sight further steepens the inferred profile. Fitting a Bonnor-Ebert sphere results in a super-critical value of the dimensionless radius xi_max=25 +/- 3. The unstable configuration of material may be related to the observed inward motions. The Bonnor-Ebert model matches the shape of the observed profile, but significantly underestimates the amount of extinction (by a factor of ~4). This discrepancy in normalization has also been found for the nearby protostellar core B335 (Harvey et al. 2001). A cylindrical density model with scale height H=0.0164+/- 0.002 pc viewed at a small inclination to the cylinder axis provides an equally good radial profile as a power law model, and reproduces the asymmetry of the core remarkably well. In addition, this model provides a basis for understanding the discrepancy in the normalization of the Bonnor-Ebert model, namely that L694-2 has prolate structure, with the full extent (mass) of the core being missed by assuming symmetry between the profiles in the plane of the sky and along the line-of-sight. The article draws on Goffman's concept of stigma and uses this to explore transitions in a society that retains some traditional characteristics, particularly the code of honour and shame, although mediated by aspects of modernity. Interviews were undertaken with 15 young people with the goal of producing narratives. The article analyses the experience of stigma, its effects and how young people manage its consequences. It concludes by drawing attention to the pervasive nature of stigma and the importance of structure, agency and reflexivity in youth transitions. In particular stigma remains an important feature of societies in which hetero-normative sexuality remains dominant
