    Among the native tropical underutilized fruits of promising potential, durian assumes great importance particularly in Malaysia, Thailand and Indonesia. Global interest in this fruit is catching up fast and its commercial prospects increasing. With promising clones now available and considering the export potential of durian, more area under its cultivation has led to loss of primitive diversity and wild/semi-wild/domesticated species. Further, research and development efforts are still required to understand durian flora1 biology and physiology of fruit set, and assessing wild and domesticated primitive genepools for their usefulness in diseases/pest resistance, physiological stress and other attributes imparting better characteristics. Specific treatises dealing with studies on taxonomy, genetic diversity, flora1 biology, breeding, improvement, conservation and utilization aspects are wanting. In the above context, the detailed synthesis provided on durian by Dr. Michael J. Brown is welcomed. It is perhaps for the first time that such a comprehensive account has been attempted on an underutilized fruit crop genepool. This publication on Durian which is a review of bibliography of about 1000 references, provides a well-synthesized information on different aspects of Durio. It deals with its origin, history, taxonomy of wild and cultivated species, identification, morphology of flower and fruit and other organs, its edibility, composition and uses, nutritional aspects, medicinal and toxicological prospects, seed physiology, pollination biology, ecology, forest resources, crop improvement efforts, agronomic requirements, propagation, cultivation and maintenance, post harvest technology, genetic resource and conservation

    Ex vivo adenoviral vector-mediated neurotrophin gene transfer to olfactory ensheathing glia: Effects on rubrospinal tract regeneration, lesion size, and functional recovery after implantation in the injured rat spinal cord

    The present study uniquely combines olfactory ensheathing glia (OEG) implantation with ex vivo adenoviral (AdV) vector-based neurotrophin gene therapy in an attempt to enhance regeneration after cervical spinal cord injury. Primary OEG were transduced with AdV vectors encoding rat brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3), or bacterial marker protein -galactosidase (LacZ) and subsequently implanted into adult Fischer rats directly after unilateral transection of the dorsolateral funiculus. Implanted animals received a total of 2 x 105 OEG that were subjected to transduction with neurotrophin-encoding AdV vector, AdV-LacZ, or no vector, respectively. At 4 months after injury, lesion volumes were smaller in all OEG implanted rats and significantly reduced in size after implantation of neurotrophin-encoding AdV vector-transduced OEG. All OEG grafts were filled with neurofilament-positive axons, and AdV vector-mediated expression of BDNF by implanted cells significantly enhanced regenerative sprouting of the rubrospinal tract. Behavioral analysis revealed that OEG-implanted rats displayed better locomotion during horizontal rope walking than unimplanted lesioned controls. Recovery of hind limb function was also improved after implantation of OEG that were transduced with a BDNF- or NT-3-encoding AdV vector. Hind limb performance during horizontal rope locomotion did directly correlate with lesion size, suggesting that neuroprotective effects of OEG implants contributed to the level of functional recovery. Thus, our results demonstrate that genetic engineering of OEG not only resulted in a cell that was more effective in promoting axonal outgrowth but could also lead to enhanced recovery after injury, possibly by sparing of spinal tissue

    Nuclear Cogeneration of Methanol and Acetaldehyde from Ethylene Glycol Using Ionizing Radiation

    Despite offering low-carbon and reliable energy, the utilization of nuclear energy is declining globally due to high upfront capital costs and longer returns on investments. Nuclear cogeneration of valuable chemicals from waste biomass-derived feedstocks could have beneficial impacts while harnessing the underutilized resource of ionizing energy. Here, we demonstrate selective methanol or acetaldehyde production from ethylene glycol, a feedstock derived from glycerol, a byproduct of biodiesel, using irradiations from a nuclear fission reactor. The influence of radiation quality, dose rate, and the absorbed dose of irradiations on radiochemical yields (G-value) has been studied. Under low-dose-rate, γ-only radiolysis during reactor shutdown rate (<0.018 kGy min–1), acetaldehyde is produced at a maximum G-value of 8.28 ± 1.05 μmol J–1 and a mass productivity of 0.73 ± 0.06% from the 20 kGy irradiation of neat ethylene glycol. When exposed to a high-dose-rate (6.5 kGy min–1), 100 kGy mixed-field of neutron + γ-ray radiations, the radiolytic selectivity is adjusted from acetaldehyde to generate methanol at a G-value of 2.91 ± 0.78 μmol J–1 and a mass productivity of 0.93 ± 0.23%. Notably, utilizing 422 theoretical systems could contribute to 4.96% of worldwide acetaldehyde production using a spent fuel pool γ-ray scheme. This research reports G-values and production capacities for acetaldehyde for high-dose scenarios and shows the potential selectivity of a nuclear cogeneration process to synthesize chemicals based on their irradiation conditions from the same reagent

    Carbohydrate esterase family 16 contains fungal hemicellulose acetyl esterases (HAEs) with varying specificity

    Acetyl esterases are an important component of the enzymatic machinery fungi use to degrade plant biomass and are classified in several Carbohydrate Esterase families of the CAZy classification system. Carbohydrate Esterase family 16 (CE16) is one of the more recently discovered CAZy families, but only a small number of its enzyme members have been characterized so far, revealing activity on xylan-derived oligosaccharides, as well as activity related to galactoglucomannan. The number of CE16 genes differs significantly in the genomes of filamentous fungi. In this study, four CE16 members were identified in the genome of Aspergillus niger NRRL3 and it was shown that they belong to three of the four phylogenetic Clades of CE16. Significant differences in expression profiles of the genes and substrate specificity of the enzymes were revealed, demonstrating the diversity within this family of enzymes. Detailed characterization of one of these four A. niger enzymes (HaeA) demonstrated activity on oligosaccharides obtained from acetylated glucuronoxylan, galactoglucomannan and xyloglucan, thus establishing this enzyme as a general hemicellulose acetyl esterase. Their broad substrate specificity makes these enzymes highly interesting for biotechnological applications in which deacetylation of polysaccharides is required.Peer reviewe

    Immunohistochemical, ultrastructural and functional analysis of axonal regeneration through peripheral nerve grafts containing Schwann cells expressing BDNF, CNTF or NT3

    Objective  To establish reference values for activated coagulation time (ACT) in normal cats and dogs, by visual assessment of clot formation using the MAX-ACTTM tube. Subjects  We recruited 43 cats and 50 dogs for the study; 11 cats and 4 dogs were excluded from the statistical analysis because of abnormalities on clinical examination or laboratory testing including anaemia, prolonged prothrombin time (PT) or activated partial thromboplastin time (APTT), or insufficient plasma volume for comprehensive laboratory coagulation testing. Procedure  Blood samples were collected via direct venipuncture for MAX-ACT, packed cell volume/total solids, manual platelet estimation and PT/APTT measurement. Blood (0.5 mL) was mixed gently in the MAX-ACT tube at 37°C for 30 s, then assessed for clot formation every 5 to 10 s by tipping the tube gently on its side and monitoring for magnet movement. The endpoint was defined as the magnet lodging in the clot. The technique was tested with 10 dogs by collecting two blood samples from the same needle insertion and running a MAX-ACT on each simultaneously. Results  In normal cats the mean MAX-ACT was 66 s (range 55–85 s). In normal dogs the mean was 71 s (range 55–80 s). There was no statistical difference between the first and second samples collected from the same needle insertion. Conclusions and Clinical Relevance  In both cats and dogs, a MAX-ACT result >85 s should be considered abnormal and further coagulation testing should be performed. Additionally, failure to discard the first few drops of the sample does not appear to significantly affect results

    The balance of power: accretion and feedback in stellar mass black holes

    In this review we discuss the population of stellar-mass black holes in our galaxy and beyond, which are the extreme endpoints of massive star evolution. In particular we focus on how we can attempt to balance the available accretion energy with feedback to the environment via radiation, jets and winds, considering also possible contributions to the energy balance from black hole spin and advection. We review quantitatively the methods which are used to estimate these quantities, regardless of the details of the astrophysics close to the black hole. Once these methods have been outlined, we work through an outburst of a black hole X-ray binary system, estimating the flow of mass and energy through the different accretion rates and states. While we focus on feedback from stellar mass black holes in X-ray binary systems, we also consider the applicability of what we have learned to supermassive black holes in active galactic nuclei. As an important control sample we also review the coupling between accretion and feedback in neutron stars, and show that it is very similar to that observed in black holes, which strongly constrains how much of the astrophysics of feedback can be unique to black holes.Comment: To be published in Haardt et al.
