    Bleeding Kansas has long been an important topic for political historians exploring how it influenced Congress, presidential campaigns, and the coming of the Civil War. Kristen Tegtmeier Oertel\u27s book uses the insights of recent social historians to add depth to this political narrative, thereby strengthening our understanding of how antebellum Kansas changed America. Bleeding Borders begins with a chapter that positions Kansas as a frontier territory. Oertel\u27s antebellum Kansans are divided by race, but not just between white and black. Starting her history in the 1820s, Oertel finds that white settlers\u27 perceptions of and interactions with Kansas Indians played a crucial role in developing white racial identity. Drawing on studies of whiteness, she finds that almost all white settlers compared themselves favorably to both the area\u27s older and more recent Indian nations, setting the stage for her conclusion that white supremacy would be a common belief among whites on both sides of the slavery question. The only exceptions to that rule, she notes, were a handful of white abolitionists who worked with African Americans to stop attempts to bring slavery (and slaves) into the Territory. Oertel\u27s section on the pervasiveness of slave resistance is particularly strong, and the African American efforts against enslavement that she chronicles help to explain both the desperation of the proslavery settlers and their ultimate failure

    Interactive Effects of Hypoxia and Temperature on Coastal Pelagic Zooplankton and Fish

    Hypoxia, triggered in large part by eutrophication, exerts widespread and expanding stress on coastal ecosystems. Hypoxia is often specifically defined as water having dissolved oxygen (DO) concentrations < 2 mg L−1. However, DO concentration alone is insufficient to categorize hypoxic stress or predict impacts of hypoxia on zooplankton and fish. Hypoxic stress depends on the oxygen supply relative to metabolic demand. Water temperature controls both oxygen solubility and the metabolic demand of aquatic ectotherms. Accordingly, to assess impacts of hypoxia requires consideration of effects of temperature on both oxygen availability and animal metabolism. Temperature differences across ecosystems or across seasons or years within an ecosystem can dramatically impact the severity of hypoxia even at similar DO concentrations. Living under sub-optimum DO can reduce temperature-dependent metabolic efficiencies, prey capture efficiency, growth and reproductive potential, thus impacting production and individual zooplankton and fish fitness. Avoidance of hypoxic bottom water can reduce or eliminate low-temperature thermal refuges for organisms and increase energy demands and respiration rates, and potentially reduce overall fitness if alternative habitats are sub-optimal. Moreover, differential habitat shifts among species can shift predator-prey abundance ratios or interactions and thus modify food webs. For example, more tolerant zooplankton prey may use hypoxic waters as a refuge from fish predation. In contrast, zooplankton avoidance of hypoxic bottom waters can result in prey aggregations at oxyclines sought out by fish predators. Hypoxic conditions that affect spatial ecology can drive taxonomic and size shifts in the zooplankton community, affecting foraging, consumption and growth of fish. Advances in understanding the ecological effects of low DO waters on pelagic zooplankton and fish and comparisons among ecosystems will require development of generic models that estimate the oxygen demand of organisms in relation to oxygen supply which depends on both DO and temperature. We provide preliminary analysis of a metric (Oxygen Stress Level) which integrates oxygen demand in relation to oxygen availability for a coastal copepod and compare the prediction of oxygen stress to actual copepod distributions in areas with hypoxic bottom waters

    Deep generative modeling for single-cell transcriptomics.

    Single-cell transcriptome measurements can reveal unexplored biological diversity, but they suffer from technical noise and bias that must be modeled to account for the resulting uncertainty in downstream analyses. Here we introduce single-cell variational inference (scVI), a ready-to-use scalable framework for the probabilistic representation and analysis of gene expression in single cells ( https://github.com/YosefLab/scVI ). scVI uses stochastic optimization and deep neural networks to aggregate information across similar cells and genes and to approximate the distributions that underlie observed expression values, while accounting for batch effects and limited sensitivity. We used scVI for a range of fundamental analysis tasks including batch correction, visualization, clustering, and differential expression, and achieved high accuracy for each task

    The Fc region of an antibody impacts the neutralization of West Nile viruses in different maturation states

    Flavivirus-infected cells secrete a structurally heterogeneous population of viruses because of an inefficient virion maturation process. Flaviviruses assemble as noninfectious, immature virions composed of trimers of envelope (E) and precursor membrane (prM) protein heterodimers. Cleavage of prM is a required process during virion maturation, although this often remains incomplete for infectious virus particles. Previous work demonstrated that the efficiency of virion maturation could impact antibody neutralization through changes in the accessibility of otherwise cryptic epitopes on the virion. In this study, we show that the neutralization potency of monoclonal antibody (MAb) E33 is sensitive to the maturation state of West Nile virus (WNV), despite its recognition of an accessible epitope, the domain III lateral ridge (DIII-LR). Comprehensive epitope mapping studies with 166 E protein DIII-LR variants revealed that the functional footprint of MAb E33 on the E protein differs subtly from that of the well-characterized DIII-LR MAb E16. Remarkably, aromatic substitutions at E protein residue 306 ablated the maturation state sensitivity of E33 IgG, and the neutralization efficacy of E33 Fab fragments was not affected by changes in the virion maturation state. We propose that E33 IgG binding on mature virions orients the Fc region in a manner that impacts subsequent antibody binding to nearby sites. This Fc-mediated steric constraint is a novel mechanism by which the maturation state of a virion modulates the efficacy of the humoral immune response to flavivirus infection

    Functional analysis of antibodies against dengue virus type 4 reveals strain-dependent epitope exposure that impacts neutralization and protection

    Although prior studies have characterized the neutralizing activities of monoclonal antibodies (MAbs) against dengue virus (DENV) serotypes 1, 2, and 3 (DENV-1, DENV-2, and DENV-3), few reports have assessed the activity of MAbs against DENV-4. Here, we evaluated the inhibitory activity of 81 new mouse anti-DENV-4 MAbs. We observed strain- and genotype-dependent differences in neutralization of DENV-4 by MAbs mapping to epitopes on domain II (DII) and DIII of the envelope (E) protein. Several anti-DENV-4 MAbs inefficiently inhibited at least one strain and/or genotype, suggesting that the exposure or sequence of neutralizing epitopes varies within isolates of this serotype. Remarkably, flavivirus cross-reactive MAbs, which bound to the highly conserved fusion loop in DII and inhibited infection of DENV-1, DENV-2, and DENV-3, more weakly neutralized five different DENV-4 strains encompassing the genetic diversity of the serotype after preincubation at 37°C. However, increasing the time of preincubation at 37°C or raising the temperature to 40°C enhanced the potency of DII fusion loop-specific MAbs and some DIII-specific MAbs against DENV-4 strains. Prophylaxis studies in two new DENV-4 mouse models showed that neutralization titers of MAbs after preincubation at 37°C correlated with activity in vivo. Our studies establish the complexity of MAb recognition against DENV-4 and suggest that differences in epitope exposure relative to other DENV serotypes affect antibody neutralization and protective activity

    This analysis examines supranational constitutionalism in the European Union. In particular, the study focuses on the role of the European Court of Justice in the creation of women’s rights. I examine the interaction between the Court and member state governments in legal integration, and also the integral role that women’s advocates – both individual activists and groups – have played in the development of EU social provisions. The findings suggest that this litigation dynamic can have the effect of fueling the integration process by creating new rights that may empower social actors and EU organizations, with the ultimate effect of diminishing member state government control over the scope and direction of EU law. This study focuses specifically on gender equality law, yet provides a general framework for examining the case law in subsequent legal domains, with the purpose of providing a more nuanced understanding of supranational governance and constitutionalism

    Macondo-1 well oil-derived polycyclic aromatic hydrocarbons

    Mesozooplankton (>200 mm) collected in August and September of 2010 from the northern Gulf of Mexico show evidence of exposure to polycyclic aromatic hydrocarbons (PAHs). Multivariate statistical analysis revealed that distributions of PAHs extracted from mesozooplankton were related to the oil released from the ruptured British Petroleum Macondo-1 (M-1) well associated with the R/V Deepwater Horizon blowout. Mesozooplankton contained 0.03–97.9 ng g 1 of total PAHs and ratios of fluoranthene to fluoranthene + pyrene less than 0.44, indicating a liquid fossil fuel source. The distribution of PAHs isolated from mesozooplankton extracted in this study shows that the 2010 Deepwater Horizon spill may have contributed to contamination in the northern Gulf of Mexico ecosystem
