    A Silvopastoral System in the North Atlantic Zone of Costa Rica: Combining Indigenous Timber Species with Dairy Pasture Swards

    The objective of this study was to design, establish and monitor a silvo pastoral experiment on a dairy farm in the northern Atlantic region of Costa Rica. Indigenous timber species, Vochysia guatemalensis and Hyeronima alchorneoides were used together with or without the tropical pasture legume, Arachis pintoi in a split plot design with a 4 x 2 x 2 factorial arrangement. First year establishment was good for the tree component (2.3 to 10 % mortality) but poor for A. pintoi (4 to 5 % of sward). Poor legume establishment was attributed to lax grazing management and excess competition from existing Brachiaria sp. A. Pintoi was reestablished in July 1996 after an adjustment of stocking rate. This experimental project is planned for a 10 year period which corresponds to the expected rotation length for harvesting the indigenous timber species

    The re-use of propagation trays in nursery greenhouses is one of the main ways in which fusarium wilt is spread in melon crops (Cucumis melo). The causal agent of the disease is the fungus Fusarium oxysporum f. sp. melonis. This paper reports that exposing these seed trays to the energy produced by a prototype microwave oven during the commercial production of melon plantlets can prevent the spread of this pathogen with only a very small increase in production costs

    Maintaining a Linked Data Cloud and Data Service for Second World War History

    One of the great promises of Linked Data is to provide a shared data infrastructure into which new data can be imported and aligned with, forming a sustainable, ever growing Linked Data Cloud (LDC). This paper studies and evaluates this idea in the context of the WarSampo LDC that provides a data infrastructure for Second World War related ontologies and data in Finland, including several mutually linked graphs, totaling ca 12 million triples. Two data integration case studies are presented, where the original WarSampo LDC and the related semantic portal were first extended by a dataset of hundreds of war cemeteries and thousands of photographs of them, and then by another dataset of over 4450 Finnish prisoners of war. As a conclusion, lessons learned are explicated, based on hands-on experience in maintaining the WarSampo LDC in a production environment.Peer reviewe

    A Recombinant Respiratory Syncytial Virus Vaccine Candidate Attenuated by a Low-Fusion F Protein Is Immunogenic and Protective against Challenge in Cotton Rats

    ABSTRACT Although respiratory syncytial virus (RSV) is the leading cause of lower respiratory tract infections in infants, a safe and effective vaccine is not yet available. Live-attenuated vaccines (LAVs) are the most advanced vaccine candidates in RSV-naive infants. However, designing an LAV with appropriate attenuation yet sufficient immunogenicity has proven challenging. In this study, we implemented reverse genetics to address these obstacles with a multifaceted LAV design that combined the codon deoptimization of genes for nonstructural proteins NS1 and NS2 (dNS), deletion of the small hydrophobic protein (ΔSH) gene, and replacement of the wild-type fusion (F) protein gene with a low-fusion RSV subgroup B F consensus sequence of the Buenos Aires clade (BAF). This vaccine candidate, RSV-A2-dNS-ΔSH-BAF (DB1), was attenuated in two models of primary human airway epithelial cells and in the upper and lower airways of cotton rats. DB1 was also highly immunogenic in cotton rats and elicited broadly neutralizing antibodies against a diverse panel of recombinant RSV strains. When vaccinated cotton rats were challenged with wild-type RSV A, DB1 reduced viral titers in the upper and lower airways by 3.8 log 10 total PFU and 2.7 log 10 PFU/g of tissue, respectively, compared to those in unvaccinated animals ( P < 0.0001). DB1 was thus attenuated, highly immunogenic, and protective against RSV challenge in cotton rats. DB1 is the first RSV LAV to incorporate a low-fusion F protein as a strategy to attenuate viral replication and preserve immunogenicity. IMPORTANCE RSV is a leading cause of infant hospitalizations and deaths. The development of an effective vaccine for this high-risk population is therefore a public health priority. Although live-attenuated vaccines have been safely administered to RSV-naive infants, strategies to balance vaccine attenuation with immunogenicity have been elusive. In this study, we introduced a novel strategy to attenuate a recombinant RSV vaccine by incorporating a low-fusion, subgroup B F protein in the genetic background of codon-deoptimized nonstructural protein genes and a deleted small hydrophobic protein gene. The resultant vaccine candidate, DB1, was attenuated, highly immunogenic, and protective against RSV challenge in cotton rats

    Seed degeneration in potato : the need for an integrated seed health strategy to mitigate the problem in developing countries

    Seed potato degeneration, the reduction in yield or quality caused by an accumulation of pathogens and pests in planting material due to successive cycles of vegetative propagation, has been a long-standing production challenge for potato growers around the world. In developed countries this problem has been overcome by general access to and frequent use of seed, produced by specialized growers, that has been certified to have pathogen and pest incidence below established thresholds, often referred to as certified seed. The success of certified seed in developed countries has concentrated the research and development agenda on the establishment of similar systems in developing countries. Despite these efforts, certified seed has had little penetration into the informal seed systems currently in place in most developing countries. Small-scale farmers in these countries continue to plant seed tubers acquired through the informal seed system, i.e. produced on-farm or acquired from neighbours or local markets. Informal seed tubers frequently have poor health status, leading to significant reductions in yield and/or market value. This review emphasizes the need to refocus management efforts in developing countries on improving the health status of seed tubers in the informal system by integrating disease resistance and on-farm management tools with strategic seed replacement. This 'integrated seed health strategy' can also prolong the good health status of plants derived from certified seed, which would otherwise be diminished due to potential rapid infection from neighbouring fields. Knowledge gaps, development challenges and impacts of this integrated seed health strategy are discussed.PostprintPeer reviewe

    Persistent Leatherback Turtle Migrations Present Opportunities for Conservation

    Effective transboundary conservation of highly migratory marine animals requires international management cooperation as well as clear scientific information about habitat use by these species. Populations of leatherback turtles (Dermochelys coriacea) in the eastern Pacific have declined by >90% during the past two decades, primarily due to unsustainable egg harvest and fisheries bycatch mortality. While research and conservation efforts on nesting beaches are ongoing, relatively little is known about this population of leatherbacks' oceanic habitat use and migration pathways. We present the largest multi-year (2004–2005, 2005–2006, and 2007) satellite tracking dataset (12,095 cumulative satellite tracking days) collected for leatherback turtles. Forty-six females were electronically tagged during three field seasons at Playa Grande, Costa Rica, the largest extant nesting colony in the eastern Pacific. After completing nesting, the turtles headed southward, traversing the dynamic equatorial currents with rapid, directed movements. In contrast to the highly varied dispersal patterns seen in many other sea turtle populations, leatherbacks from Playa Grande traveled within a persistent migration corridor from Costa Rica, past the equator, and into the South Pacific Gyre, a vast, low-energy, low-productivity region. We describe the predictable effects of ocean currents on a leatherback migration corridor and characterize long-distance movements by the turtles in the eastern South Pacific. These data from high seas habitats will also elucidate potential areas for mitigating fisheries bycatch interactions. These findings directly inform existing multinational conservation frameworks and provide immediate regions in the migration corridor where conservation can be implemented.
