    Rapid and selective surveillance of Arabidopsis thaliana genome annotations with Centrifuge

    Summary: Centrifuge is a user-friendly system to simultaneously access Arabidopsis gene annotations and intra- and inter-organism sequence comparison data. The tool allows rapid retrieval of user-selected data for each annotated Arabidopsis gene providing, in any combination, data on the following features: predicted protein properties such as mass, pI, cellular location and transmembrane domains; SWISS-PROT annotations; Interpro domains; Gene Ontology records; verified transcription; BLAST matches to the proteomes of A.thaliana, Oryza sativa (rice), Caenorhabditis elegans, Drosophila melanogaster and Homo sapiens. The tool lends itself particularly well to the rapid analysis of contigs or of tens or hundreds of genes identified by high-throughput gene expression experiments. In these cases, a summary table of principal predicted protein features for all genes is given followed by more detailed reports for each individual gene.     Cet article présente des attributs caractérisant les contours extraits par les détecteurs haute-échelle, dans des situations de natures différentes : texture, bruit ou contours divers. Les attributs sont géométriques, topologiques ou relatifs à la texture, et sont calculés sur les contours ou leurs zones d'influence. Ils permettent la création de cartes des différentes zones de texture/bruit sur l'image. Les contours haute-échelle sont conservés à l'intérieur des régions non bruitées et non texturées ; ce qui permet d'obtenir une très bonne localisation des contours dans ces zones. Dans les régions bruitées ou texturées, les contours haute-échelle sont supprimés ; ce qui permet de s'affranchir de la sur-segmentation et offre la possibilité d'appliquer un détecteur différent pour une coopération de plusieurs méthodes de segmentation, adaptées à la nature des zones

    The role of stress coping strategies for life impairments in ADHD

    Attention-deficit/hyperactivity disorder (ADHD) is commonly accompanied by functional impairments within personal, professional, and social context as well as further psychological distress. The role of perceived stress has been examined in this regard, although little is known about the influence of stress coping strategies. We examined the dynamics between ADHD symptomatology, stress coping strategies, and life impairments in a sample of 230 clinical and non-clinical adult individuals. ADHD was associated with low probability of choosing adaptive but high probability of implementing maladaptive stress coping strategies as well as with increased life impairments. Adaptive stress coping showed little effects beyond the associations between ADHD and life impairments, whereas some maladaptive coping strategies increased current life impairments over and above the effects of ADHD and further psychological distress. The present findings emphasize the need of professional support in adults with ADHD and the necessity to include stress coping strategies in respective treatment approaches aimed at reducing life impairments

    Manual versus microfluidic-assisted nanoparticle manufacture : impact of silk fibroin stock on nanoparticle characteristics

    Silk has a long track record of clinical use in the human body, and new formulations, including silk nanoparticles, continue to reveal the promise of this natural biopolymer for healthcare applications. Native silk fibroin can be isolated directly from the silk gland, but generating sufficient material for routine studies is difficult. Consequently, silk fibroin, typically extracted from cocoons, serves as the source for nanoparticle formation. This silk requires extensive processing (e.g., degumming, dissolution, etc.) to yield a hypoallergenic aqueous silk stock, but the impact of processing on nanoparticle production and characteristics is largely unknown. Here, manual and microfluidic-assisted silk nanoparticle manufacturing from 60- and 90-min degummed silk yielded consistent particle sizes (100.9-114.1 nm) with low polydispersity. However, the zeta potential was significantly lower (P 30 min) had no significant effect on particle attributes. Overall, the results showed that silk fibroin processing directly impacts nanoparticle characteristics

    Affective speech modulates a cortico-limbic network in real time

    Affect signaling in human communication involves cortico-limbic brain systems for affect information decoding, such as expressed in vocal intonations during affective speech. Both, the affecto-acoustic speech profile of speakers and the cortico-limbic affect recognition network of listeners were previously identified using non-social and non-adaptive research protocols. However, these protocols neglected the inherent socio-dyadic nature of affective communication, thus underestimating the real-time adaptive dynamics of affective speech that maximize listeners' neural effects and affect recognition. To approximate this socio-adaptive and neural context of affective communication, we used an innovative real-time neuroimaging setup that linked speakers' live affective speech production with listeners' limbic brain signals that served as a proxy for affect recognition. We show that affective speech communication is acoustically more distinctive, adaptive, and individualized in a live adaptive setting and more efficiently capitalizes on neural affect decoding mechanisms in limbic and associated networks than non-adaptive affective speech communication. Only live affective speech produced in adaption to listeners' limbic signals was closely linked to their emotion recognition as quantified by speakers' acoustics and listeners' emotional rating correlations. Furthermore, while live and adaptive aggressive speaking directly modulated limbic activity in listeners, joyful speaking modulated limbic activity in connection with the ventral striatum that is, amongst others, involved in the processing of pleasure. Thus, evolved neural mechanisms for affect decoding seem largely optimized for interactive and individually adaptive communicative contexts

    ESCAlate – Adaptive treatment approach for adolescents and adults with ADHD: study protocol for a randomized controlled trial

    Background: Over the last decade, a wide range of attention-deficit/hyperactivity disorder (ADHD) treatment approaches for adults, including both pharmacological interventions and psychosocial treatments, have been proposed and observed to be efficient. In practice, individual treatment concepts are based on results of clinical studies as well as international guidelines (NICE Guidelines) that recommend a step-by-step treatment approach. Since the evidence supporting this approach is limited, the aim of the present study is to determine an optimal intervention regarding severity levels of ADHD symptomatology conducting a randomized controlled trial. Method: We aim to include 279 ADHD subjects aged between 16 and 45 years. First, participants are randomized to either a face-to-face psychoeducation, telephone assisted self-help (TASH), or a waiting control group (Step 1). All participants assigned to the control group are treated using TASH after a 3-month waiting period. Participants are then allocated to one of three groups, based on their remaining severity level of ADHD symptoms, as (1) full responder, (2) partial responder, or (3) non-responder (Step 2). Full responders receive counseling, partial responders receive either counseling only or counseling and neurofeedback (NF), and non-responders receive either pharmacological treatment only or pharmacological treatment and NF, followed by a 3 month period without intervention. Discussion: The naturalistic sample is one of the study’s advantages, avoiding highly selective inclusion or exclusion criteria. The efficacy of an evidence-based stepped care intervention is explored by primary (reduction of severity of ADHD symptoms) and secondary outcomes (functional outcomes, e.g., quality of life, anger management, enhancement of psychosocial well-being). Predictors of therapeutic response and non-response are being investigated at each step of intervention. Further, sex differences are also being explored.     Sports-related sudden cardiac arrest in young adults

    AIMS Data on sports-related sudden cardiac arrest (SrSCA) among young adults in the general population are scarce. We aimed to determine the overall SrSCA incidence, characteristics, and outcomes in young adults. METHODS AND RESULTS Prospective cohort study of all cases of SrSCA between 2012 and 2019 in Germany and Paris area, France, involving subjects aged 18-35 years. Detection of SrSCA was achieved via multiple sources, including emergency medical services (EMS) reporting and web-based screening of media releases. Cases and aetiologies were centrally adjudicated. Overall, a total of 147 SrSCA (mean age 28.1 ± 4.8 years, 95.2% males) occurred, with an overall burden of 4.77 [95% confidence interval (CI) 2.85-6.68] cases per million-year, including 12 (8.2%) cases in young competitive athletes. While bystander cardiopulmonary resuscitation (CPR) was initiated in 114 (82.6%), automated external defibrillator (AED) use by bystanders occurred only in a minority (7.5%). Public AED use prior to EMS arrival (odds ratio 6.25, 95% CI 1.48-43.20, P = 0.02) was the strongest independent predictor of survival at hospital discharge (38.1%). Among cases that benefited from both immediate bystander CPR and AED use, survival rate was 90.9%. Coronary artery disease was the most frequent aetiology (25.8%), mainly through acute coronary syndrome (86.9%). CONCLUSION Sports-related sudden cardiac arrest in the young occurs mainly in recreational male sports participants. Public AED use remains disappointingly low, although survival may reach 90% among those who benefit from both bystander CPR and early defibrillation. Coronary artery disease is the most prevalent cause of SrSCA in young adults
