    Office-Based Educational Handout for Influenza Vaccination: A Randomized Controlled Trial

METHODS: A convenience sample of parents of children ≥6 months old with a visit at 2 New York City pediatric clinics between August 2016 and March 2017 were randomly assigned (1:1:1) to receive either usual care, an educational handout about influenza disease that was based on local data, or an educational handout about influenza disease that was based on national data. Parents received the handout in the waiting room before their visit. Primary outcomes were child influenza vaccine receipt on the day of the clinic visit and by the end of the season. A multivariable logistic regression was used to assess associations between intervention and vaccination, with adjustment for variables that were significantly different between arms. RESULTS: Parents who received an intervention (versus usual care) had greater odds of child influenza vaccine receipt by the end of the season (74.9% vs 65.4%; adjusted odds ratio 1.68; 95% confidence interval: 1.06-2.67) but not on the day of the clinic visit. Parents who received the national data handout (versus usual care) had greater odds of child influenza vaccine receipt on the day of the clinic visit (59.0% vs 52.6%; adjusted odds ratio 1.79; 95% confidence interval: 1.04-3.08) but not by the end of the season. CONCLUSIONS: Providing an educational intervention in the waiting room before a pediatric provider visit may help increase child influenza vaccine receipt.European CommissionNIH - Ruth L.     The report makes an in depth analysis of what to expect of future urban population growth in cities across India. Cities play a critical role in India\u27s development. While its one billion-plus population is predominantly rural, over 300 million people live in urban areas. One-third of this population lives in 35 urban agglomerations or cities exceeding one million. Cities\u27 governments are responsible for delivering various public services, yet severe infrastructures shortages in water supply and sanitation, roads, transportation, housing and waste management, and inefficient management have resulted in poor quality services. These inadequate services and worsening environmental conditions affect the poor. Between 1950 and 2000 India\u27s urban population increased from 62 to 288 million. Already strained to provide services and quality of life to existing urban residents, cities will face tremendous challenges in expanding existing infrastructure and avoiding deterioration of living standards due to congestion, pollution, and lack of basic services. A doubling of the population over 30 years means that by 2030 there will be a second Mumbai, a second Calcutta, and a second Bangalore that must be fed, supplied with water, sanitation, electricity, give public and private transportation options; and where garbage must be disposed of. The report concludes by laying out a series of state and local actions over the short-medium and long-term to enhance fiscal sustainability and strengthening institutional capacity building of state and local governments

    Edaxadiene: A New Bioactive Diterpene from Mycobacterium tuberculosis

Presented here is the characterization of an atypical class I diterpene cyclase from M. tuberculosis that catalyzes an unusual cyclization reaction in converting the known M. tuberculosis metabolite halimadienyl diphosphate to a further cyclized novel diterpene, which we have termed edaxadiene, as it directly inhibits maturation of the phagosomal compartment in which the bacterium is taken up during infection

    Mycobacterium tuberculosis is a major human pathogen and the causative agent for the pulmonary disease, tuberculosis (TB). Current treatment programs to combat TB are under threat due to the emergence of multi-drug and extensively-drug resistant TB. As part of our efforts towards the discovery of new anti-tubercular leads, a number of potent tetrahydropyrazolo[1,5-a]pyrimidine-3-ca​rboxamide(THPP) and N-benzyl-6′,7′-dihydrospiro[piperidine-4,​4′-thieno[3,2-c]pyran](Spiro) analogues were recently identified against Mycobacterium tuberculosis and Mycobacterium bovis BCG through a high-throughput whole-cell screening campaign. Herein, we describe the attractive in vitro and in vivo anti-tubercular profiles of both lead series. The generation of M. tuberculosis spontaneous mutants and subsequent whole genome sequencing of several resistant mutants identified single mutations in the essential mmpL3 gene. This ‘genetic phenotype’ was further confirmed by a ‘chemical phenotype’, whereby M. bovis BCG treated with both the THPP and Spiro series resulted in the accumulation of trehalose monomycolate. In vivo efficacy evaluation of two optimized THPP and Spiro leads showed how the compounds were able to reduce >2 logs bacterial cfu counts in the lungs of infected mice

    Genetic diversity studies in kodo millet (Paspalum scrobiculatum L.)

    The present investigation was undertaken to assess genetic diversity in seventy genotypes of kodo millet. The analysis of variance revealed the presence of significant variation among the genotypes for all 13 characters. Higher genotypic coefficient of variation (GCV), phenotypic coefficient of variation (PCV), heritability and genetic advance as per cent mean were recorded for thumb raceme length, number of productive tillers per plant, length of panicle, raceme length and grain yield per plant, indicating that simple selection could be practised for improving these traits. Seventy genotypes were grouped into seven different clusters on the basis of magnitude of D2 values by Mahalanobis D2 analysis. Cluster I had 51 genotypes followed by cluster II with 14 genotypes, while clusters III, IV, V, VI and VII were mono genotypic. The inter–cluster distance was high between clusters II and III and therefore it is suggested to use these genotypes as parents for hybridization to evolve potential segregants

    Detection of Pathogenic Mycobacteria Based on Functionalized Quantum Dots Coupled with Immunomagnetic Separation

    Mycobacteria have always proven difficult to identify due to their low growth rate and fastidious nature. Therefore molecular biology and more recently nanotechnology, have been exploited from early on for the detection of these pathogens. Here we present the first stage of development of an assay incorporating cadmium selenide quantum dots (QDs) for the detection of mycobacterial surface antigens. The principle of the assay is the separation of bacterial cells using magnetic beads coupled with genus-specific polyclonal antibodies and monoclonal antibodies for heparin-binding hemagglutinin. These complexes are then tagged with anti-mouse biotinylated antibody and finally streptavidin-conjugated QDs which leads to the detection of a fluorescent signal. For the evaluation of performance, the method under study was applied on Mycobacterium bovis BCG and Mycobacterium tuberculosis (positive controls), as well as E. coli and Salmonella spp. that constituted the negative controls. The direct observation of the latter category of samples did not reveal fluorescence as opposed to the mycobacteria mentioned above. The minimum detection limit of the assay was defined to 104 bacteria/ml, which could be further decreased by a 1 log when fluorescence was measured with a spectrofluorometer. The method described here can be easily adjusted for any other protein target of either the pathogen or the host, and once fully developed it will be directly applicable on clinical samples
