Perspectives On The LEED (Leadership In Energy And Environmental Design) System As A Green Certification Standard
As we begin to reach the limits of Earths biosphere, ensuring that sustainable practices are integrating into new buildings and into older buildings are crucial to remove unsustainable strains on the environment. This paper seeks to examine the merits and disadvantages of LEED as a certification system in the context of environmental sustainability. While there does seem to be legitimate criticism of the LEED system, it does serve to bring and legitimize sustainable development to an area that has a profound and continual effect on the environment. The systems should be continuously examined and revamped in the context of improving the integration of environmental sustainability into commercial and residential development. LEED has the opportunity to be revamped, improved, and to become a powerful force in promoting environmental sustainability in construction
Diversity in case management modalities: the Summit model
Though ubiquitous in community mental health agencies, case management suffers from a lack of consensus regarding its definition, essential components, and appropriate application. Meaningful comparisons of various case management models await such a consensus. Global assessments of case management must be replaced by empirical studies of specific interventions with respect to the needs of specific populations. The authors describe a highly differentiated and prescriptive system of case management involving the application of more than one model of service delivery. Such a diversified and targeted system offers an opportunity to study the technology of case management in a more meaningful manner
Law, Business Strategy, And Social Change In The Global Environment
In this article, we examine the dynamic relationship between law, economics, and sustainability. Increased legal regulation is not a sufficient condition for the mitigation of economic and social crises. Corporations are not legally required to respect global social and human rights under international law. In addition, select legal regulations increase social harm through the preservation of corporate financial interests. Laws that attempt to protect social rights at the expense of corporate interests can provoke hostile reactions from the business community. However, recent trends and examples from within the pharmaceutical and information industries suggest a new era of global corporate citizenship where socially responsible organizations achieve profit through pursuing ethical goals
Identification of a transporter complex responsible for the cytosolic entry of nitrogen-containing bisphosphonates
Nitrogen-containing-bisphosphonates (N-BPs) are widely prescribed to treat osteoporosis and other bone-related diseases. Although previous studies established that N-BPs function by inhibiting the mevalonate pathway in osteoclasts, the mechanism by which N-BPs enter the cytosol from the extracellular space to reach their molecular target is not understood. Here we implemented a CRISPRi-mediated genome-wide screen and identified SLC37A3 (solute carrier family 37 member A3) as a gene required for the action of N-BPs in mammalian cells. We observed that SLC37A3 forms a complex with ATRAID (all-trans retinoic acid-induced differentiation factor), a previously identified genetic target of N-BPs. SLC37A3 and ATRAID localize to lysosomes and are required for releasing N-BP molecules that have trafficked to lysosomes through fluid-phase endocytosis into the cytosol. Our results elucidate the route by which N-BPs are delivered to their molecular target, addressing a key aspect of the mechanism of action of N-BPs that may have significant clinical relevance
Assignment of the four disulfides in the N-terminal somatomedin B domain of native vitronectin isolated from human plasma
The primary sequence of the N-terminal somatomedin B (SMB) domain of native vitronectin contains 44 amino acids, including a framework of four disulfide bonds formed by 8 closely spaced cysteines in sequence patterns similar to those found in the cystine knot family of proteins. The SMB domain of vitronectin was isolated by digesting the protein with endoproteinase Glu-C and purifying the N-terminal 1-55 peptide by reverse-phase high performance liquid chromatography. Through a combination of techniques, including stepwise reduction and alkylation at acidic pH, peptide mapping with matrix-assisted laser desorption ionization mass spectrometry and NMR, the disulfide bonds contained in the SMB domain have been determined to be Cys5:Cys9, Cys 19:Cys31, Cys21:Cys32, and Cys 25:Cys39. This pattern of disulfides differs from two other connectivities that have been reported previously for recombinant forms of the SMB domain expressed in Escherichia coli. This arrangement of disulfide bonds in the SMB domain from native vitronectin forms a rigid core around the Cys19: Cys31 and Cys21:Cys32 disulfides. A small positively charged loop is created at the N terminus by the Cys5: Cys9 cystine. The most prominent feature of this disulfide-bonding pattern is a loop between Cys25 and Cys 39 similar to cystine-stabilized α-helical structures commonly observed in cystine knots. This α-helix has been confirmed in the solution structure determined for this domain using NMR (Mayasundari, A., Whittemore, N. A., Serpersu, E. H., and Peterson, C. B. (2004) J. Biol. Chem. 279, 29359-29366). It confers function on the SMB domain, comprising the site for binding to plasminogen activator inhibitor type-1 and the urokinase receptor
Predicting distributions of known and unknown reptile species in Madagascar
Despite the importance of tropical biodiversity(1), informative species distributional data are seldom available for biogeographical study or setting conservation priorities(2,3). Modelling ecological niche distributions of species offers a potential soluion(4-7); however, the utility of old locality data from museums, and of more recent remotely sensed satellite data, remains poorly explored, especially for rapidly changing tropical landscapes. Using 29 modern data sets of environmental land coverage and 621 chameleon occurrence localities from Madagascar ( historical and recent), here we demonstrate a significant ability of our niche models in predicting species distribution. At 11 recently inventoried sites, highest predictive success (85.1%) was obtained for models based only on modern occurrence data (74.7% and 82.8% predictive success, respectively, for pre-1978 and all data combined). Notably, these models also identified three intersecting areas of over-prediction that recently yielded seven chameleon species new to science. We conclude that ecological niche modelling using recent locality records and readily available environmental coverage data provides informative biogeographical data for poorly known tropical landscapes, and offers innovative potential for the discovery of unknown distributional areas and unknown species.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/62843/1/nature02205.pd
Stability of methylation markers in head and neck squamous cell carcinoma
BackgroundAs cancer progresses, methylation patterns change to promote the tumorigenic phenotype. However, stability of methylation markers over time and the extent that biopsy samples are representative of larger tumor specimens are unknown. This information is critical for clinical use of such biomarkers.MethodsNinety‐eight patients with tumor specimens from 2 timepoints were measured for DNA methylation in the promoter regions across 4 genes.ResultsThere were no significant differences in overall methylation of CCNA1 (cyclin A1), NDN (necdin), deleted in colorectal carcinoma (DCC), and cluster of differentiation 1a (CD1A) within paired specimens (p values = .56, .17, .66, and .58, respectively). All genes showed strong correlations between paired specimens across time. Asymmetric warming over coastal California and its impact on the premium wine industry
Climatic changes over coastal California from 1951 to 1997 may have benefited the premium wine industry, as seen in higher quality wines and larger grape yields. Observed temperature warming trends were asymmetric, with greatest warming at night and during spring. Warming was associated with large increases in eastern Pacific sea surface temperatures (SST) and amounts of atmospheric water vapor. Although the average annual temperature warming trend was modest (1.13°C/47 yr), there was a 20 d reduction in frost occurrence and a 65 d increase in frost-free growing season length. In the Napa and Sonoma valleys, warmer winter and spring temperatures advanced the start of the growing season by 18 to 24 d, and enhanced atmospheric water vapor resulted in a 7 % reduction in evaporative demand. Given the strong coupling between Pacific SSTs and the coastal California climate, and because regional-scale SSTs persist for 6 to 12 mo, additional research may allow the possibility of predicting vintage quantity and quality from previous winter conditions
