    Data collected from an ecological study of the mayfly Potamanthus rnyops (Walsh) in Michigan showed intraspecific variability in taxonomic characteristics that have been employed by previous investigators for species separation. Nymphal dorsal maculation patterns varied considerably within a single population. Also, the ratio of mandibular tusk length to head length increased with successive nymphal instars. Certain adult taxonomic characteristics, particularly relative male imago eye size and distance of separation, were either too poorly defined or too variable to be conclusive in species identification

    Simulating evolutionary responses of an introgressed insect resistance trait for ecological effect assessment of transgene flow: a model for supporting informed decisionmaking in environmental risk assessment

    Predicting outcomes of transgene flow from arable crops requires a system perspective that considers ecological and evolutionary processes within a landscape context. In Europe, the arable weed Raphanus raphanistrum is a potential hybridization partner of oilseed rape, and the two species are ecologically linked through the common herbivores Meligethes spp. Observations in Switzerland show that high densities of Meligethes beetles maintained by oilseed rape crops can lead to considerable damage on R. raphanistrum. We asked how increased insect resistance in R. raphanistrum – as might be acquired through introgression from transgenic oilseed rape – would affect seed production under natural herbivore pressure. In simulation experiments, plants protected against Meligethes beetles produced about twice as many seeds as unprotected plants. All stages in the development of reproductive structures from buds to pods were negatively affected by the herbivore, with the transition from buds to flowers being the most vulnerable. We conclude that resistance to Meligethes beetles could confer a considerable selective advantage upon R. raphanistrum in regions where oilseed rape is widely grown

    P-values for high-dimensional regression

    Assigning significance in high-dimensional regression is challenging. Most computationally efficient selection algorithms cannot guard against inclusion of noise variables. Asymptotically valid p-values are not available. An exception is a recent proposal by Wasserman and Roeder (2008) which splits the data into two parts. The number of variables is then reduced to a manageable size using the first split, while classical variable selection techniques can be applied to the remaining variables, using the data from the second split. This yields asymptotic error control under minimal conditions. It involves, however, a one-time random split of the data. Results are sensitive to this arbitrary choice: it amounts to a `p-value lottery' and makes it difficult to reproduce results. Here, we show that inference across multiple random splits can be aggregated, while keeping asymptotic control over the inclusion of noise variables. We show that the resulting p-values can be used for control of both family-wise error (FWER) and false discovery rate (FDR). In addition, the proposed aggregation is shown to improve power while reducing the number of falsely selected variables substantially.Comment: 25 pages, 4 figure

    Detection of Formaldehyde Towards the Extreme Carbon Star IRC+10216

    We report the detection of H2CO (formaldehyde) around the carbon-rich AGB star, IRC+10216. We find a fractional abundance with respect to molecular hydrogen of x(H2CO)= (1.3 {+1.5}{-0.8}) x 10^{-8}. This corresponds to a formaldehyde abundance with respect to water vapor of x(H2CO)/x(H2O)=(1.1 +/- 0.2) x 10^{-2}, in line with the formaldehyde abundances found in Solar System comets, and indicates that the putative extrasolar cometary system around IRC+10216 may have a similar chemical composition to Solar System comets. However, we also failed to detect CH3OH (methanol) around IRC+10216 and our upper limit of x(CH3OH)/x(H2O) < 7.7 x 10^{-4}, (3 sigma), indicates that methanol is substantially underabundant in IRC+10216, compared to Solar System comets. We also conclude, based on offset observations, that formaldehyde has an extended source in the envelope of IRC+10216 and may be produced by the photodissociation of a parent molecule, similar to the production mechanism for formaldehyde in Solar System comet comae. Preliminary mapping observations also indicate a possible asymmetry in the spatial distribution of formaldehyde around IRC+10216, but higher signal-to-noise observations are required to confirm this finding.     Mutational signatures of DNA mismatch repair deficiency in C. elegans and human cancers

    Throughout their lifetime, cells are subject to extrinsic and intrinsic mutational processes leaving behind characteristic signatures in the genome. DNA mismatch repair (MMR) deficiency leads to hypermutation and is found in different cancer types. Although it is possible to associate mutational signatures extracted from human cancers with possible mutational processes, the exact causation is often unknown. Here, we use C. elegans genome sequencing of pms-2 and mlh-1 knockouts to reveal the mutational patterns linked to C. elegans MMR deficiency and their dependency on endogenous replication errors and errors caused by deletion of the polymerase epsilon subunit pole-4. Signature extraction from 215 human colorectal and 289 gastric adenocarcinomas revealed three MMR-associated signatures, one of which closely resembles the C. elegans MMR spectrum and strongly discriminates microsatellite stable and unstable tumors (AUC = 98%). A characteristic difference between human and C. elegans MMR deficiency is the lack of elevated levels of N (C) under barG &gt; NTG mutations in C. elegans, likely caused by the absence of cytosine (CpG) methylation in worms. The other two human MMR signatures may reflect the interaction between MMR deficiency and other mutagenic processes, but their exact cause remains unknown. In summary, combining information from genetically defined models and cancer samples allows for better aligning mutational signatures to causal mutagenic processes

    Plant-Mediated Rhizosphere Oxygenation in the Native Invasive Salt Marsh Grass Elymus athericus

    In the last decades, the spread of Elymus athericus has caused significant changes to the plant community composition and ecosystem services of European marshes. The distribution of E. athericus was typically limited by soil conditions characteristic for high marshes, such as low flooding frequency and high soil aeration. However, recently the spread of E. athericus has begun to also include low-marsh environments. A high-marsh ecotype and a low-marsh ecotype of E. athericus have been described, where the latter possess habitat-specific phenotypic traits facilitating a better adaption for inhabiting low-marsh areas. In this study, planar optodes were applied to investigate plant-mediated sediment oxygenation in E. athericus, which is a characteristic trait for marsh plants inhabiting frequently flooded environments. Under waterlogged conditions, oxygen (O2) was translocated from aboveground sources to the roots, where it leaked out into the surrounding sediment generating oxic root zones below the sediment surface. Oxic root zones were clearly visible in the optode images, and no differences were found in the O2-leaking capacity between ecotypes. Concentration profiles measured perpendicular to the roots revealed that the radius of the oxic root zones ranged from 0.5 to 2.6 mm measured from the root surface to the bulk anoxic sediment. The variation of oxic root zones was monitored over three consecutive light–dark cycles (12 h/12 h). The O2 concentration of the oxic root zones was markedly reduced in darkness, yet the sediment still remained oxic in the immediate vicinity of the roots. Increased stomatal conductance improving the access to atmospheric O2 as well as photosynthetic O2 production are likely factors facilitating the improved rhizosphere oxygenation during light exposure of the aboveground biomass. E. athericus’ capacity to oxygenate its rhizosphere is an inheritable trait that may facilitate its spread into low-marsh areas. Furthermore, this trait makes E. athericus a highly competitive species in marshes facing the effects of accelerated sea-level rise, where waterlogged sediment conditions could become increasingly pronounced

    Forest elephant movement and habitat use in a tropical forest-grassland mosaic in Gabon

    Poaching of forest elephants (Loxodonta cyclotis) for ivory has decimated their populations in Central Africa. Studying elephant movement can provide insight into habitat and resource use to reveal where, when, and why they move and guide conservation efforts. We fitted 17 forest elephants with global positioning system (GPS) collars in 2015 and 2016 in the tropical forest-grassland mosaic of the Wonga Wongué Presidential Reserve (WW), Gabon. Using the location data, we quantified movement distances, home ranges, and habitat use to examine the environmental drivers of elephant movements and predict where elephants occur spatially and temporally. Forest elephants, on average, traveled 2,840 km annually and had home ranges of 713 km2, with males covering significantly larger home ranges than females. Forest elephants demonstrated both daily and seasonal movement patterns. Daily, they moved between forest and grassland at dawn and dusk. Seasonally, they spent proportionally more time in grassland than forest during the short-wet season when grasses recruit. Forest elephants also traveled faster during the short-wet season when fruit availability was greatest, likely reflecting long, direct movements to preferred fruiting tree species. Forest elephants tended to select areas with high tree and shrub density that afford cover and browse. When villages occurred in their home ranges elephants spent a disproportionate amount of time near them, particularly in the dry season, probably for access to agricultural crops and preferred habitat. Given the importance of the grassland habitat for elephants, maintenance of the forest-grassland matrix is a conservation priority in WW. Law enforcement, outreach, and education should focus on areas of potential human-elephant conflict near villages along the borders of the reserve. GPS-tracking should be extended into multi-use areas in the peripheries of protected areas to evaluate the effects of human disturbance on elephant movements and to maintain connectivity among elephant populations in Gabon

    Clinical outcomes of patients with estimated low or intermediate surgical risk undergoing transcatheter aortic valve implantation

    Aims Transcatheter aortic valve implantation (TAVI) is an established treatment alternative to surgical aortic valve replacement in high-risk and inoperable patients and outcomes among patients with estimated low or intermediate risk remain to be determined. The aim of this study was to assess clinical outcomes among patients with estimated low or intermediate surgical risk undergoing TAVI. Methods and results Between August 2007 and October 2011, 389 consecutive patients underwent TAVI and were categorized according to the Society of Thoracic Surgeons (STS) score into low (STS 8%; n = 94, 24.2%) groups for the purpose of this study. Significant differences were found between the groups (low risk vs. intermediate risk vs. high risk) for age (78.2 ± 6.7 vs. 82.7 ± 5.7 vs. 83.7 ± 4.9, P < 0.001), body mass index (28.1 ± 6.1 vs. 26.5 ± 4.9 vs. 24.4 ± 4.6, P < 0.001), chronic renal failure (34 vs. 67 vs. 90%, P < 0.001), all-cause mortality at 30 days (2.4 vs. 3.9 vs. 14.9%, P = 0.001), and all-cause mortality at 1 year (10.1 vs. 16.1 vs. 34.5%, P = 0.0003). No differences were observed with regards to cerebrovascular accidents and myocardial infarction during 1-year follow-up. Conclusion In contemporary practice, TAVI is not limited to inoperable or STS-defined high-risk patients and should be guided by the decision of an interdisciplinary Heart Team. Compared with patients at calculated high risk, well-selected patients with STS-defined intermediate or low risk appear to have favourable clinical outcome

    The role of co-morbidity in the selection of antidiabetic pharmacotherapy in type-2 diabetes

    Metformin is, if not contraindicated and if tolerated, usually preferred over other antidiabetic drugs for the first line treatment of type-2 diabetes. The particular decision on which antidiabetic agent to use is based on variables such as efficacy, cost, potential side effects, effects on weight, comorbidities, hypoglycemia, risk, and patient preferences. However, there is no guidance how to consider these in the selection of antidiabetic drug treatment. In this work, we aimed to summarize available evidence and tried to give pragmatic treatment recommendations from a clinical practice perspective. There are clear contraindications for some drugs in those with impaired renal and liver function and precautions in those with heart failure for the use of metformin (NYHA III-IV) and glitazones. On the other hand, GLP-1 analogs, DPP-4 inhibitors and acarbose are generally less critical and can be used in the majority of patients. We identified the following gaps with respect to the selection of antidiabetic drug treatment in patients with co-morbid disease conditions: 1) Guidelines fail to give advice on the use of specific antidiabetic drugs in patients with co-morbidity. 2) The literature is deficient in studies documenting antidiabetic drug use in patients with severely impaired renal function, diabetic retinopathy, cerebrovascular disease and systolic heart failure. 3) Further there are no specific data on patients with multiple of these co-morbid disease conditions. We postulate that differential use of antidiabetic drugs in patients with co-morbid disease constellations will help to reduce treatment related complications and might improve prognosis
