    A deep ocean acoustic noise floor, 1–800 Hz

The definitive version was published in Journal of the Acoustical Society of America 143 (2018): 1223, doi:10.1121/1.5025042.The ocean acoustic noise floor (observed when the overhead wind is low, ships are distant, and marine life silent) has been measured on an array extending up 987 m from 5048 m depth in the eastern North Pacific, in what is one of only a few recent measurements of the vertical noise distribution near the seafloor in the deep ocean. The floor is roughly independent of depth for 1–6 Hz, and the slope (∼ f−7) is consistent with Longuet-Higgins radiation from oppositely-directed surface waves. Above 6 Hz, the acoustic floor increases with frequency due to distant shipping before falling as ∼ f−2 from 40 to 800 Hz. The noise floor just above the seafloor is only about 5 dB greater than during the 1975 CHURCH OPAL experiment (50–200 Hz), even though these measurements are not subject to the same bathymetric blockage. The floor increases up the array by roughly 15 dB for 40–500 Hz. Immediately above the seafloor, the acoustic energy is concentrated in a narrow, horizontal beam that narrows as f−1 and has a beam width at 75 Hz that is less than the array resolution.     Excisional treatment in women with cervical adenocarcinoma in situ (AIS): a prospective randomised controlled noninferiority trial to compare AIS persistence/recurrence after loop electrosurgical excision procedure with cold knife cone biopsy: protocol for a pilot study

    Introduction: Adenocarcinoma in situ (AIS) of the uterine cervix is the precursor to invasive endocervical adenocarcinoma. An excisional biopsy such as a cold knife cone biopsy (CKC) should be performed to exclude invasive adenocarcinoma. Loop electrosurgical excision procedure (LEEP) is an alternative modality to CKC but is controversial in AIS. There is a perception that there is a greater likelihood of incomplete excision of AIS with LEEP because the depth of excised tissue tends to be smaller and the tissue margins may show thermal artefact which can interfere with pathology assessment. In the USA, guidelines recommend that any treatment modality can be used to excise AIS, provided that the specimen remains intact with interpretable margins. However, there are no high-quality studies comparing LEEP with CKC and well-designed prospective studies are needed. If such a study were to show that LEEP was non-inferior to CKC for the outcomes of post-treatment persistence, recurrence and adenocarcinoma, LEEP could be recommended as an appropriate treatment option for AIS in selected patients. This would benefit women because, unlike CKC, LEEP does not require general anaesthesia and may be associated with reduced morbidity. Methods and analysis: The proposed exploratory study is a parallel group trial with an allocation ratio of 2:1 in favour of the intervention (LEEP: CKC). Participants are women aged ≥18 to ≤45 years diagnosed with AIS on cervical screening and/or colposcopically directed biopsy in Australia and New Zealand, who are to receive excisional treatment in a tertiary level centre.     Estimating the contribution of stimulant injection to HIV and HCV epidemics among people who inject drugs and implications for harm reduction:A modeling analysis

    BACKGROUND: Stimulants, such as amphetamines and cocaine, are widely injected among people who inject drugs (PWID). Systematic reviews indicate stimulant injection is associated with HIV and HCV among PWID. Using these associations, we estimated the contribution of stimulant injection to HIV and HCV transmission among PWID. METHODS: We modeled HIV and HCV transmission among PWID, incorporating excess injecting and sexual risk among PWID who inject stimulants. We simulated three illustrative settings with different stimulants injected, prevalence of stimulant injecting, and HIV/HCV epidemiology. We estimated one-year population attributable fractions of stimulant injection on new HIV and HCV infections, and impact of scaling up needle-syringe programs (NSP). RESULTS: In low prevalence settings of stimulant injection (St. Petersburg-like, where 13% inject amphetamine), 9% (2.5-97.5% interval [95%I]: 6-15%) and 7% (95%I 4-11%) of incident HIV and HCV cases, respectively, could be associated with stimulant injection in the next year. With moderate stimulant injection (Montreal-like, where 34% inject cocaine), 29% (95%I: 19–37%) and 19% (95%I: 16-21%) of incident HIV and HCV cases, respectively, could be associated with stimulant injection. In high-burden settings like Bangkok where 65% inject methamphetamine, 23% (95%I:10–34%) and 20% (95%I: 9-27%) of incident HIV and HCV cases could be due to stimulant injection. High-coverage NSP (60%) among PWID who inject stimulants could reduce HIV (by 22-65%) and HCV incidence (by 7-11%) in a decade. DISCUSSION: Stimulant injection contributes substantially to HIV and HCV among PWID. NSP scale-up and development of novel interventions among PWID who inject stimulants are warranted

    Wind sea behind a cold front and deep ocean acoustics

The definitive version was published in Journal of Physical Oceanography 46 (2016): 1705-1716, doi:10.1175/JPO-D-15-0221.1.A rapid and broadband (1 h, 1 < f < 400 Hz) increase in pressure and vertical velocity on the deep ocean floor was observed on seven instruments comprising a 20-km array in the northeastern subtropical Pacific. The authors associate the jump with the passage of a cold front and focus on the 4- and 400-Hz spectra. At every station, the time of the jump is consistent with the front coming from the northwest. The apparent rate of progress, 10–20 km h−1 (2.8–5.6 m s−1), agrees with meteorological observations. The acoustic radiation below the front is modeled as arising from a moving half-plane of uncorrelated acoustic dipoles. The half-plane is preceded by a 10-km transition zone, over which the radiator strength increases linearly from zero. With this model, the time derivative of the jump at a station yields a second and independent estimate of the front’s speed, 8.5 km h−1 (2.4 m s−1). For the 4-Hz spectra, the source physics is taken to be Longuet-Higgins radiation. Its strength depends on the quantity , where Fζ is the wave amplitude power spectrum and I the overlap integral. Thus, the 1-h time constant observed in the bottom data implies a similar time constant for the growth of the wave field quantity behind the front. The spectra at 400 Hz have a similar time constant, but the jump occurs 25 min later.     Computer simulations of domain growth and phase separation in two-dimensional binary immiscible fluids using dissipative particle dynamics

    We investigate the dynamical behavior of binary fluid systems in two dimensions using dissipative particle dynamics. We find that following a symmetric quench the domain size R(t) grows with time t according to two distinct algebraic laws R(t) = t^n: at early times n = 1/2, while for later times n = 2/3. Following an asymmetric quench we observe only n = 1/2, and if momentum conservation is violated we see n = 1/3 at early times. Bubble simulations confirm the existence of a finite surface tension and the validity of Laplace's law. Our results are compared with similar simulations which have been performed previously using molecular dynamics, lattice-gas and lattice-Boltzmann automata, and Langevin dynamics. We conclude that dissipative particle dynamics is a promising method for simulating fluid properties in such systems.Comment: RevTeX; 22 pages, 5 low-resolution figures.     Epigenome-wide profiling identifies significant differences in DNA methylation between matched-pairs of T- and B-lymphocytes from healthy individuals

    Multiple reports now describe changes to the DNA methylome in rheumatoid arthritis and in many cases have analyzed methylation in mixed cell populations from whole blood. However, these approaches may preclude the identification of cell type-specific methylation, which may subsequently bias identification of disease-specific changes. To address this possibility, we conducted genome-wide DNA methylation profiling using HumanMethylation450 BeadChips to identify differences within matched pairs of T-lymphocytes and B-lymphocytes isolated from the peripheral blood of 10 healthy females. Array data were processed and differential methylation identified using NIMBL software. Validation of array data was performed by bisulfite Pyrosequencing. Genome-wide DNA methylation was initially determined by analysis of LINE-1 sequences and was higher in B-lymphocytes than matched T-lymphocytes (69.8 vs. 65.2%, p ≤ 0.01). Pairwise analysis identified 679 CpGs, representing 250 genes, which were differentially methylated between T-lymphocytes and B-lymphocytes. The majority of sites (76.6%) were hypermethylated in B-lymphocytes. Pyrosequencing of selected candidates confirmed the array data in all cases. Hierarchical clustering revealed perfect segregation of samples into two distinct clusters based on cell type. Differentially methylated genes showed enrichment for biological functions/pathways associated with leukocytes and T-lymphocytes. Our work for the first time shows that T-lymphocytes and B-lymphocytes possess intrinsic differences in DNA methylation within a restricted set of functionally-related genes. These data provide a foundation for investigating DNA methylation in diseases in which these cell types play important and distinct roles

    Lattice-gas simulations of Domain Growth, Saturation and Self-Assembly in Immiscible Fluids and Microemulsions

    We investigate the dynamical behavior of both binary fluid and ternary microemulsion systems in two dimensions using a recently introduced hydrodynamic lattice-gas model of microemulsions. We find that the presence of amphiphile in our simulations reduces the usual oil-water interfacial tension in accord with experiment and consequently affects the non-equilibrium growth of oil and water domains. As the density of surfactant is increased we observe a crossover from the usual two-dimensional binary fluid scaling laws to a growth that is {\it slow}, and we find that this slow growth can be characterized by a logarithmic time scale. With sufficient surfactant in the system we observe that the domains cease to grow beyond a certain point and we find that this final characteristic domain size is inversely proportional to the interfacial surfactant concentration in the system.Comment: 28 pages, latex, embedded .eps figures, one figure is in colour, all in one uuencoded gzip compressed tar file, submitted to Physical Review
