    Gemini Planet Imager Observational Calibrations III: Empirical Measurement Methods and Applications of High-Resolution Microlens PSFs

    The newly commissioned Gemini Planet Imager (GPI) combines extreme adaptive optics, an advanced coronagraph, precision wavefront control and a lenslet-based integral field spectrograph (IFS) to measure the spectra of young extrasolar giant planets between 0.9-2.5 um. Each GPI detector image, when in spectral model, consists of ~37,000 microspectra which are under or critically sampled in the spatial direction. This paper demonstrates how to obtain high-resolution microlens PSFs and discusses their use in enhancing the wavelength calibration, flexure compensation and spectral extraction. This method is generally applicable to any lenslet-based integral field spectrograph including proposed future instrument concepts for space missions.Comment: 10 pages, 6 figures.     Early and reversible neuropathology induced by tetracycline-regulated lentiviral overexpression of mutant huntingtin in rat striatum

    The ability to overexpress full-length huntingtin or large fragments represents an important challenge to mimic Huntington's pathology and reproduce all stages of the disease in a time frame compatible with rodent life span. In the present study, tetracycline-regulated lentiviral vectors leading to high expression levels were used to accelerate the pathological process. Rats were simultaneously injected with vectors coding for the transactivator and wild type (WT) or mutated huntingtin (TRE-853-19Q/82Q) in the left and right striatum, respectively, and analyzed in the ‘on' and ‘off' conditions. Overexpression of TRE-853-19Q protein or residual expression of TRE-853-82Q in ‘off' condition did not cause any significant neuronal pathology. Overexpressed TRE-853-82Q protein led to proteolytic release of N-terminal htt fragments, nuclear aggregation, and a striatal dysfunction as revealed by decrease of DARPP-32 staining but absence of NeuN down-regulation. The differential effect on the DARPP-32/NeuN neuronal staining was observed as early as 1 month after injection and maintained at 3 months. In contrast, expression of a shorter htt form (htt171-82Q) did not require processing prior formation of nuclear aggregates and caused decrease of both DARPP-32 and NeuN neuronal markers at one month post-injection suggesting that polyQ pathology may be dependent on protein context. Finally, the reversibility of the pathology was assessed. Huntingtin expression was turn ‘on' for 1 month and then shut ‘off' for 2 months. Recovery of DARPP-32 immunoreactivity and clearance of huntingtin aggregates were observed in animals treated with doxycycline.     Le fût d'un arbre sur pied est soumis à l'action de la pesanteur, qui induit un effet de flexion sur un arbre déséquilibré. La qualification de cette action réclame donc d'évaluer non seulement la masse de l'arbre, mais aussi son déséquilibre, c'est-à-dire la position de son centre de gravité dans un plan horizontal. Une méthode d'estimation de cette position à partir de relevés dendrométriques simples (mesure d'une inclinaison du fût, de huit rayons de la projection au sol du houppier) est proposée. Appliquée à 9 peupliers, elle permet de classer les individus en trois groupes : droits, inclinés, flexueux. Confrontée aux résultats d'un essai mécanique de suppression de la masse supportée, elle apparaît suffisante pour estimer la direction de l'effort de flexion sur les individus de conformation simple, droits ou inclinés, mais doit être affinée dans les cas plus complexes. L'essai comme la modélisation confirment en outre la prépondérance des effets de flexion sur ceux de compression, même sur des individus apparemment équilibrés. (Résumé d'auteur

    Meet-in-the-Middle Attacks and Structural Analysis of Round-Reduced PRINCE

    NXP Semiconductors and its academic partners challenged the cryptographic community with finding practical attacks on the block cipher they designed, PRINCE. Instead of trying to attack as many rounds as possible using attacks which are usually impractical despite being faster than brute-force, the challenge invites cryptographers to find practical attacks and encourages them to actually implement them. In this paper, we present new attacks on round-reduced PRINCE including the ones which won the challenge in the 4, 6 and 8-round categories --- the highest for which winners were identified. Our first attacks rely on a meet-in-the-middle approach and break up to 10 rounds of the cipher. We also describe heuristic methods we used to find practical SAT-based and differential attacks. Finally, we also present an analysis of the cycle structure of the internal rounds of PRINCE leading both to a low complexity distinguisher for 4-round PRINCE-core and an alternative representation of the cipher valid in particular contexts and which highlights, in this cases, a poor diffusion

    Non-equilibrium sedimentation of colloids on the particle scale

    We investigate sedimentation of model hard sphere-like colloidal dispersions confined in horizontal capillaries using laser scanning confocal microscopy, dynamical density functional theory, and Brownian dynamics computer simulations. For homogenized initial states we obtain quantitative agreement of the results from the respective approaches for the time evolution of the one-body density distribution and the osmotic pressure on the walls. We demonstrate that single particle information can be obtained experimentally in systems that were initialized further out-of-equilibrium such that complex lateral patterns form.Comment: to be published in Phys.     No widespread induction of cell death genes occurs in pure motoneurons in an amyotrophic lateral sclerosis mouse model

    To identify candidate genes that may be involved in motoneuron degeneration, we combined laser capture microdissection with microarray technology. Gene expression in motoneurons was analyzed during the progression of the disease in transgenic SOD1G93A mice that develop motoneuron loss. Three major observations were made: first, there was only a small number of genes that were differentially expressed in motoneurons at a pre-symptomatic age (27 out of 34 000 transcripts). Secondly, there is an early specific up-regulation of the gene coding for the intermediate filament vimentin that is increased even further during disease progression. Using in situ hybridization and immunohistochemical analysis, we show that vimentin expression was not only elevated in motoneurons but that the protein formed inclusions in the motoneuron cytoplasm. Thirdly, a time-course analysis of the motoneurons at a symptomatic age (90 and 120 days) showed a modest de-regulation of only a few genes associated with cell death pathways; however, a massive up-regulation of genes involved in cell growth and/or maintenance was observed.
