Potential Use of MALDI-ToF Mass Spectrometry for Rapid Detection of Antifungal Resistance in the Human Pathogen Candida glabrata.
The echinocandins are relatively new antifungal drugs that represent, together with the older azoles, the recommended and/or preferred agents to treat candidaemia and other forms of invasive candidiasis in human patients. If "time is of the essence" to reduce the mortality for these infections, the administration of appropriate antifungal therapy could be accelerated by the timely reporting of laboratory test results. In this study, we attempted to validate a MALDI-ToF mass spectrometry-based assay for the antifungal susceptibility testing (AFST) of the potentially multidrug-resistant pathogen Candida glabrata against anidulafungin and fluconazole. The practical applicability of the assay, reported here as MS-AFST, was assessed with a panel of clinical isolates that were selected to represent phenotypically and genotypically/molecularly susceptible or resistant strains. The data show the potential of our assay for rapid detection of antifungal resistance, although the MS-AFST assay performed at 3 h of the in vitro antifungal exposure failed to detect C. glabrata isolates with echinocandin resistance-associated FKS2 mutations. However, cell growth kinetics in the presence of anidulafungin revealed important cues about the in vitro fitness of C. glabrata isolates, which may lead to genotypic or phenotypic antifungal testing in clinical practice

Scatterometer and Model Wind and Wind Stress in the Oregon–Northern California Coastal Zone
Measurements of surface wind stress by the SeaWinds scatterometer on NASA's Quick Scatterometer (QuikSCAT) satellite are analyzed and compared with several different atmospheric model products, from an operational model and two high-resolution nested regional models, during two summer periods, June through September 2000 and 2001, in the coastal region west of Oregon and northern California. The mean summer wind stress had a southward component over the entire region in both years. Orographic intensifications of both the mean and fluctuating wind stress occurred near Cape Blanco, Cape Mendocino, and Point Arena. Substantial differences between the model products are found for the mean, variable, and diurnal wind stress fields. Temporal correlations with the QuikSCAT observations are highest for the operational model, and are not improved by either nested model. The highest-resolution nested model most accurately reproduced the mean observed stress fields, but slightly degrades the temporal correlations due to incoherent high-frequency (0.5–2 cpd) fluctuations. The QuikSCAT data reveal surprisingly strong diurnal fluctuations that extend offshore 150 km or more with magnitudes that are a significant fraction of the mean wind stress. Wind stress curl fields from QuikSCAT and the models show local cyclonic and anticyclonic maxima associated with the orographic wind intensification around the capes. The present results are consistent with the hypothesis of a wind-driven mechanism for coastal jet separation and cold water plume and anticyclonic eddy formation in the California Current System south of Cape Blanco
Couples that work full time and are happily married : myth or reality?
Este trabalho apresenta parte de uma pesquisa quantitativa cujo principal objetivo foi avaliar a satisfação no casamento de homens e mulheres que optaram por relacionamentos de duplo trabalho. Participaram do estudo 222 homens e 222 mulheres casados/as, funcionários/as em diversas instituições públicas de Brasília - DF. A maior concentração de respondentes de ambos os sexos está na faixa etária entre 31 a 40 anos. Os participantes responderam a um questionário demográfico e à Escala de Ajustamento Diádico - DAS. Os resultados mostraram que a maioria dos participantes está satisfeita com seus relacionamentos, sendo que as mulheres apresentaram média de satisfação inferior à dos homens. Quanto à percepção do futuro do relacionamento, ficou evidente o comprometimento de homens e mulheres em investirem na manutenção do casamento. Os resultados questionam a idéia vigente de falência do casamento e da família e apontam para uma transformação das relações. _________________________________________________________________________________________________________ ABSTRACTThis paper presents part of a quantitative research regarding marriages in which men and women are engaged in full time work. The main objective was to evaluate marital satisfaction in dual worker couples. Subjects were 222 married men and 222 married women that work in several public institutions in Brasilia - DF. The majority of the participants were between 31 to 40 years old. They answered a demographic questionnaire and the Dyadic Adjustment Scale - DAS. Results showed that the majority of participants were satisfied with their relationships although women presented lower satisfaction scores then men. Regarding the perception of the future of the relationship, the results showed that subjects were committed to preserve their marriage. These data question current ideas that marriages and families are outmoded and point in the direction of a transformation in interpersonal relationships
Real-time Image Generation for Compressive Light Field Displays
With the invention of integral imaging and parallax barriers in the beginning of the 20th century, glasses-free 3D displays have become feasible. Only today—more than a century later—glasses-free 3D displays are finally emerging in the consumer market. The technologies being employed in current-generation devices, however, are fundamentally the same as what was invented 100 years ago. With rapid advances in optical fabrication, digital processing power, and computational perception, a new generation of display technology is emerging: compressive displays exploring the co-design of optical elements and computational processing while taking particular characteristics of the human visual system into account. In this paper, we discuss real-time implementation strategies for emerging compressive light field displays. We consider displays composed of multiple stacked layers of light-attenuating or polarization-rotating layers, such as LCDs. The involved image generation requires iterative tomographic image synthesis. We demonstrate that, for the case of light field display, computed tomographic light field synthesis maps well to operations included in the standard graphics pipeline, facilitating efficient GPU-based implementations with real-time framerates.United States. Computational Opioid Prescribing: A Novel Application of Clinical Pharmacokinetics
We implemented a pharmacokinetics-based mathematical modeling technique using algebra to assist pre-scribers with point-of-care opioid dosing. We call this technique computational opioid prescribing (COP). Because population pharmacokinetic parameter values are needed to estimate drug dosing regimen designs for individual patients using COP, and those values are not readily available to prescribers because they exist scattered in the vast pharmacology literature, we estimated the population pharmacokinetic parameter values for 12 commonly prescribed opioids from various sources using the bootstrap resampling technique. Our results show that opioid dosing regimen design, evaluation, and modification is feasible using COP. We conclude that COP is a new technique for the quantitative assessment of opioid dosing regimen design evaluation and adjustment, which may help prescribers to manage acute and chronic pain at the point-of-care. Potential benefits include opioid dose optimization and minimization of adverse opioid drug events, leading to potential improvement in patient treatment outcomes and safety
Cost Savings of Universal Decolonization to Prevent Intensive Care Unit Infection: Implications of the REDUCE MRSA Trial
ObjectiveTo estimate and compare the impact on healthcare costs of 3 alternative strategies for reducing bloodstream infections in the intensive care unit (ICU): methicillin-resistant Staphylococcus aureus (MRSA) nares screening and isolation, targeted decolonization (ie, screening, isolation, and decolonization of MRSA carriers or infections), and universal decolonization (ie, no screening and decolonization of all ICU patients).DesignCost analysis using decision modeling.MethodsWe developed a decision-analysis model to estimate the health care costs of targeted decolonization and universal decolonization strategies compared with a strategy of MRSA nares screening and isolation. Effectiveness estimates were derived from a recent randomized trial of the 3 strategies, and cost estimates were derived from the literature.ResultsIn the base case, universal decolonization was the dominant strategy and was estimated to have both lower intervention costs and lower total ICU costs than either screening and isolation or targeted decolonization. Compared with screening and isolation, universal decolonization was estimated to save $171,000 and prevent 9 additional bloodstream infections for every 1,000 ICU admissions. The dominance of universal decolonization persisted under a wide range of cost and effectiveness assumptions.ConclusionsA strategy of universal decolonization for patients admitted to the ICU would both reduce bloodstream infections and likely reduce healthcare costs compared with strategies of MRSA nares screening and isolation or screening and isolation coupled with targeted decolonization

Live to cheat another day: bacterial dormancy facilitates the social exploitation of beta-lactamases
The breakdown of antibiotics by β-lactamases may be cooperative, since resistant cells can detoxify their environment and facilitate the growth of susceptible neighbours. However, previous studies of this phenomenon have used artificial bacterial vectors or engineered bacteria to increase the secretion of β-lactamases from cells. Here, we investigated whether a broad-spectrum β-lactamase gene carried by a naturally occurring plasmid (pCT) is cooperative under a range of conditions. In ordinary batch culture on solid media, there was little or no evidence that resistant bacteria could protect susceptible cells from ampicillin, although resistant colonies could locally detoxify this growth medium. However, when susceptible cells were inoculated at high densities, late-appearing phenotypically susceptible bacteria grew in the vicinity of resistant colonies. We infer that persisters, cells that have survived antibiotics by undergoing a period of dormancy, founded these satellite colonies. The number of persister colonies was positively correlated with the density of resistant colonies and increased as antibiotic concentrations decreased. We argue that detoxification can be cooperative under a limited range of conditions: if the toxins are bacteriostatic rather than bacteridical; or if susceptible cells invade communities after resistant bacteria; or if dormancy allows susceptible cells to avoid bactericides. Resistance and tolerance were previously thought to be independent solutions for surviving antibiotics. Here, we show that these are interacting strategies: the presence of bacteria adopting one solution can have substantial effects on the fitness of their neighbours
