    The Association Between AURKA Gene rs2273535 Polymorphism and Gastric Cancer Risk in a Chinese Population

    The Aurora kinase A (AURKA) gene is frequently amplified and overexpressed in gastric cancer (GC). The overexpression of AURKA promotes inflammation and tumorigenesis in GC. We performed co-expression analysis to identify genes associated with AURKA and speculated its function through the COXPRESdb and STRING databases. We also conducted a hospital-based case-control study involving 385 GC cases and 470 controls in a Chinese population to evaluate the role of AURKA gene rs2273535 polymorphism in the risk of GC. Genotyping was performed using a custom-by-design 48-Plex single nucleotide polymorphism (SNP) Scanâ„¢ Kit. Co-expression analysis indicated that the overexpression of AURKA may be associated with poor prognosis of GC. In addition, TT genotypes of rs2273535 polymorphism increased the risk of GC by 72% compared to the AA genotypes. This significant correlation was also observed in the allelic and dominant models. The stratified analysis suggested that TT+AT genotypes showed positive correlation with the risk of GC among female, age <55 years group and non-smokers compared to AA genotypes. In conclusion, AURKA plays an important role in the development of GC. Larger studies with more diverse ethnic populations are needed to confirm these results

    3D printing of hybrid MoS2-graphene aerogels as highly porous electrode materials for sodium ion battery anodes

    This study reports a 3D freeze-printing method that integrates inkjet printing and freeze casting to control both the microstructure and macroporosity via formation of ice microcrystals during printing. A viscous aqueous ink consisting of a molecular MoS2 precursor (ammonium thiomolybdate) mixed with graphene oxide (GO) nanosheets is used in the printing process. Post-treatments by freeze-drying and reductive thermal annealing convert the printed intermediate mixture into a hybrid structure consisting of MoS2 nanoparticles anchored on the surface of 2D rGO nanosheets in a macroporous framework, which is fully characterized with FESEM, TEM, XRD, Raman spectroscopy and TGA. The resulting hybrid MoS2-rGO aerogels are studied as anodes for sodiumion batteries. They present a high initial specific capacity over 429 mAh/g at C/3.3 rate in the potential range of 2.5–0.10 V (vs Na+/Na). The process involves both reversible 2 Na+ insertion and slow irreversible conversion of MoS2 to metallic Mo. At higher rates, the conversion reaction is suppressed and the electrode is dominated by fast Na+ intercalation with good stability. This demonstrates that the 3D printing technology can be used as a processing technique to control the materials properties for energy storage

    Drug-herb interactions between Scutellaria baicalensis and pharmaceutical drugs : insights from experimental studies, mechanistic actions to clinical applications

    Whilst the popular use of herbal medicine globally, it poses challenges in managing potential drug-herb interaction. There are two folds of the drug-herb interaction, a beneficial interaction that may improve therapeutic outcome and minimise the toxicity of drug desirably; by contrast, negative interaction may evoke unwanted clinical consequences, especially with drugs of narrow therapeutic index. Scutellaria baicalensis Georgi is one of the most popular medicinal plants used in Asian countries. It has been widely used for treating various diseases and conditions such as cancer, diabetes, inflammation, and oxidative stress. Studies on its extract and bioactive compounds have shown pharmacodynamic and pharmacokinetic interactions with a wide range of pharmaceutical drugs as evidenced by plenty of in vitro, in vivo and clinical studies. Notably, S. baicalensis and its bioactives including baicalein, baicalin and wogonin exhibited synergistic interactions with many pharmaceutical drugs to enhance their efficacy, reduce toxicity or overcome drug resistance to combat complex diseases such as cancer, diabetes and infectious diseases. On the other hand, S. baicalensis and its bioactives also affected the pharmacokinetic profile of many drugs in absorption, distribution, metabolism and elimination via the regulatory actions of the efflux pumps and cytochrome P450 enzymes. This review provides comprehensive references of the observed pharmacodynamic and pharmacokinetic drug interactions of Scutellaria baicalensis and its bioactives. We have elucidated the interaction with detailed mechanistic actions, identified the knowledge gaps for future research and potential clinical implications. Such knowledge is important for the practice of both conventional and complementary medicines, and it is essential to ensure the safe use of related herbal medicines.     Ask One More Time: Self-Agreement Improves Reasoning of Language Models in (Almost) All Scenarios

    Although chain-of-thought (CoT) prompting combined with language models has achieved encouraging results on complex reasoning tasks, the naive greedy decoding used in CoT prompting usually causes the repetitiveness and local optimality. To address this shortcoming, ensemble-optimization tries to obtain multiple reasoning paths to get the final answer assembly. However, current ensemble-optimization methods either simply employ rule-based post-processing such as \textit{self-consistency}, or train an additional model based on several task-related human annotations to select the best one among multiple reasoning paths, yet fail to generalize to realistic settings where the type of input questions is unknown or the answer format of reasoning paths is unknown. To avoid their limitations, we propose \textbf{self-agreement}, a generalizable ensemble-optimization method applying in almost all scenarios where the type of input questions and the answer format of reasoning paths may be known or unknown. Self-agreement firstly samples from language model's decoder to generate a \textit{diverse} set of reasoning paths, and subsequently prompts the language model \textit{one more time} to determine the optimal answer by selecting the most \textit{agreed} answer among the sampled reasoning paths. Self-agreement simultaneously achieves remarkable performance on six public reasoning benchmarks and superior generalization capabilities.Comment: Work in progres

    Two kinds of β-SiAlON nanostructure whiskers, rod-like and wool-like whiskers, were synthesized by pressure-less sintering method at 1773 K for 5 hours. The whiskers synthesized were characterized by powder X-ray diffraction (XRD), scanning electron microscope (SEM), transmission electron microscope (TEM), and high-resolution electron microscope (HREM) techniques. It was found that diameter distribution of rod-like whiskers was about 80–250 nm, while it was about 45–55 nm in diameter for the wool-like whiskers. The growth mechanisms of β-SiAlON nanostructure whiskers are discussed by the vapor-solid (VS) and vapor-liquid-solid (VLS) mechanisms, respectively

    Interfacial Interactions and Nanostructure Changes in DPPG/HD Monolayer at the Air/Water Interface

    Lung surfactant (LS) plays a crucial role in regulating surface tension during normal respiration cycles by decreasing the work associated with lung expansion and therefore decreases the metabolic energy consumed. Monolayer surfactant films composed of a mixture of phospholipids and spreading additives are of optional utility for applications in lung surfactant-based therapies. A simple, minimal model of such a lung surfactant system, composed of 1,2-dipalmitoyl-sn-glycero-3-[phosphor-rac-(1-gylcerol)] (DPPG) and hexadecanol (HD), was prepared, and the surface pressure-area (π-A) isotherms and nanostructure characteristics of the binary mixture were investigated at the air/water interface using a combination of Langmuir-Blodgett (LB) and atomic force microscopy (AFM) techniques. Based on the regular solution theory, the miscibility and stability of the two components in the monolayer were analyzed in terms of compression modulus (Cs-1) , excess Gibbs free energy (ΔGexcπ) , activity coefficients (γ), and interaction parameter (ξ). The results of this paper provide valuable insight into basic thermodynamics and nanostructure of mixed DPPG/HD monolayers; it is helpful to understand the thermodynamic behavior of HD as spreading additive in LS monolayer with a view toward characterizing potential improvements to LS performance brought about by addition of HD to lung phospholipids

    KwaiYiiMath: Technical Report

    Recent advancements in large language models (LLMs) have demonstrated remarkable abilities in handling a variety of natural language processing (NLP) downstream tasks, even on mathematical tasks requiring multi-step reasoning. In this report, we introduce the KwaiYiiMath which enhances the mathematical reasoning abilities of KwaiYiiBase1, by applying Supervised Fine-Tuning (SFT) and Reinforced Learning from Human Feedback (RLHF), including on both English and Chinese mathematical tasks. Meanwhile, we also constructed a small-scale Chinese primary school mathematics test set (named KMath), consisting of 188 examples to evaluate the correctness of the problem-solving process generated by the models. Empirical studies demonstrate that KwaiYiiMath can achieve state-of-the-art (SOTA) performance on GSM8k, CMath, and KMath compared with the similar size models, respectively.Comment: technical report. arXiv admin note: text overlap with arXiv:2306.16636 by other author

    The Intracellular Transport and Secretion of Calumenin-1/2 in Living Cells

    Calumenin isoforms 1 and 2 (calu-1/2), encoded by the CALU gene, belong to the CREC protein family. Calu-1/2 proteins are secreted into the extracellular space, but the secretory process and regulatory mechanism are largely unknown. Here, using a time-lapse imaging system, we visualized the intracellular transport and secretory process of calu-1/2-EGFP after their translocation into the ER lumen. Interestingly, we observed that an abundance of calu-1/2-EGFP accumulated in cellular processes before being released into the extracellular space, while only part of calu-1/2-EGFP proteins were secreted directly after attaching to the cell periphery. Moreover, we found the secretion of calu-1/2-EGFP required microtubule integrity, and that calu-1/2-EGFP-containing vesicles were transported by the motor proteins Kif5b and cytoplasmic dynein. Finally, we determined the export signal of calu-1/2-EGFP (amino acid positions 20–46) and provided evidence that the asparagine at site 131 was indispensable for calu-1/2-EGFP stabilization. Taken together, we provide a detailed picture of the intracellular transport of calu-1/2-EGFP, which facilitates our understanding of the secretory mechanism of calu-1/2

    Silkworm Coatomers and Their Role in Tube Expansion of Posterior Silkgland

    Background: Coat protein complex I (COPI) vesicles, coated by seven coatomer subunits, are mainly responsible for Golgito-ER transport. Silkworm posterior silkgland (PSG), a highly differentiated secretory tissue, secretes fibroin for silk production, but many physiological processes in the PSG cells await further investigation. Methodology/Principal Findings: Here, to investigate the role of silkworm COPI, we cloned six silkworm COPI subunits (a,b,b9, d, e, and f-COP), determined their peak expression in day 2 in fifth-instar PSG, and visualized the localization of COPI, as a coat complex, with cis-Golgi. By dsRNA injection into silkworm larvae, we suppressed the expression of a-, b9- and c-COP, and demonstrated that COPI subunits were required for PSG tube expansion. Knockdown of a-COP disrupted the integrity of Golgi apparatus and led to a narrower glandular lumen of the PSG, suggesting that silkworm COPI is essential for PSG tube expansion. Conclusions/Significance: The initial characterization reveals the essential roles of silkworm COPI in PSG. Although silkworm COPI resembles the previously characterized coatomers in other organisms, some surprising findings require further investigation.     International audienceChina currently has 42 million elderly dependents whose long-term care needs can no longer be met by the family system that has prevailed until now. The government at all levels is encouraging the production of nursing homes (yanglaoyuan), but the shortage of caregivers is acute. This article analyzes the characteristics of the supply of nursing aides in the yanglaoyuan of two major provincial capitals with a population of 10 million (Hangzhou, a prosperous coastal city, Zhengzhou, a developing inland city), providing account on public and private initiatives to develop professional care facilities. The survey covered 31 yanglaoyuan in Hangzhou and 27 in Zhengzhou. It revealed a low level of qualification of the nursing staff due to the predominance of rural migrant women between 45 and 55 years of age, a lack of nursing skills and a low general level of remuneration. However, there has been a rise in nursing skills in Hangzhou, owing to two main drivers: financial support from the municipal government for the training and upgrading of nursing assistants' salaries on the one hand, and the contribution of foreign operators to the dissemination of nursing knowledge and techniques on the other. These characteristics are not matched by Zhengzhou, where yanglaoyuan production is still low, mainly due to the less advanced stage of economic development of this inland metropolis.La Chine compte aujourd’hui 42 millions de personnes âgées dépendantes dont les besoins en soins de longue durée ne peuvent plus être pris en charge par le système familial qui prévalait jusque-là. Cet article analyse les caractéristiques de l’offre d’aides-soignantes dans les yanglaoyuan de deux grandes capitales provinciales d’une population de 10 millions d’habitants (Hangzhou, une ville côtière prospère, et Zhengzhou, une ville intérieure en développement), et rend compte des initiatives publiques et privées pour y développer des filières professionnelles de soins. L’enquête a porté sur 31 yanglaoyuan à Hangzhou et 27 à Zhengzhou. Elle révèle un faible niveau de qualification des personnels d’aides-soignantes du fait de la prédominance de femmes migrantes rurales âgées de 45 à 55 ans, d’un manque de compétences en soins infirmiers et d’un faible niveau général des rémunérations. Toutefois, on observe une montée en compétence des soins à Hangzhou, grâce à deux principaux facteurs : un soutien financier du gouvernement municipal à la formation et à la revalorisation des salaires d’aides-soignantes d’une part, et la contribution d’opérateurs étrangers à la diffusion des savoirs et techniques de soins d’autre part. Ces caractéristiques n’ont pas d’équivalent à Zhengzhou où la production de yanglaoyuan est encore faible, principalement en raison du stade de développement économique moins avancé de cette métropole de l’intérieur
