katedra: KPV; přílohy: CD ROM; rozsah: 115 s.This diploma thesis is focused on the possibilities of using television and video in the primary schools. The theoretical part defines basic terms a gives a theoretical background which is used in the practical part. This part also deals with television broadcasting and its new trends. The theoretical part describes how television broadcasting influences infant TV viewers and it deals with the possibilities of using television and video as an instrument of education. The practical part is focused on the analysis of the program offer for children and the young. This part also shows questionnaire investigation which was done among teachers in primary schools. The aim of this questionnaire was to find what teachers think about a possibility of using television and video as an instrument of education and whether they take benefits from this possibility or not. In the end of the practical part we create the concept for education in primary schools, which is based on the questionnaire and information from studied sources. This concept contains activities dealing with television and video as an instrument of education and uses TV programs as audio-visual material.

Art in public space in Prague with case study of Krištof Kintera‘s memorials

    No full text
The theoretical part of the work focuses on public space, art in public space its function and the evolution of public art in the Czech Republic. The next part discusses the process of placing art in public spaces in Prague. The practical part consists of a case study of art in public space in Prague from Czech artist Krištof Kintera, two memorials Bike to heaven and Of One's Own Volition – Memento mori

Fluorescent magnetic nanoparticles for cell labeling: Flux synthesis of manganite particles and novel functionalization of silica shell

International audienceNovel synthetic approaches for the development of multimodal imaging agents with high chemical stability are demonstrated. The magnetic cores are based on La0.63Sr0.37MnO3 manganite prepared as individual grains using a flux method followed by additional thermal treatment in a protective silica shell allowing to enhance their magnetic properties. The cores are then isolated and covered de novo with a hybrid silica layer formed through the hydrolysis and polycondensation of tetraethoxysilane and a fluorescent silane synthesized from rhodamine, piperazine spacer, and 3-iodopropyltrimethoxysilane. The aminoalkyltrialkoxysilanes are strictly avoided and the resulting particles are hydrolytically stable and do not release dye. The high colloidal stability of the material and the long durability of the fluorescence are reinforced by an additional silica layer on the surface of the particles. Structural and magnetic studies of the products using XRD, TEM, and SQUID magnetometry confirm the importance of the thermal treatment and demonstrate that no mechanical treatment is required for the flux-synthesized manganite. Detailed cell viability tests show negligible or very low toxicity at concentrations at which excellent labeling is achieved. Predominant localization of nanoparticles in lysosomes is confirmed by immunofluorescence staining. Relaxometric and biological studies suggest that the functionalized nanoparticles are suitable for imaging applications

Four strains of the fungus Quambalaria cyanescens (Basidiomycota: Microstromatales), were used for the determination of secondary metabolites production and their antimicrobial and biological activities. A new naphthoquinone named quambalarine A, (S)-(+)-3-(5-ethyl-tetrahydrofuran-2-yliden)-5,7,8-trihydroxy-2-oxo-1,4-naphthoquinone (1), together with two known naphthoquinones, 3-hexanoyl-2,5,7,8-tetrahydroxy-1,4-naphthoquinone (named here as quambalarine B, 2) and mompain, 2,5,7,8-tetrahydroxy-1,4-naphthoquinone (3) were isolated. Their structures were determined by single-crystal X-ray diffraction crystallography, NMR and MS spectrometry. Quambalarine A (1) had a broad antifungal and antibacterial activity and is able inhibit growth of human pathogenic fungus Aspergillus fumigatus and fungi co-occurring with Q. cyanescens in bark beetle galleries including insect pathogenic species Beauveria bassiana. Quambalarine B (2) was active against several fungi and mompain mainly against bacteria. The biological activity against human-derived cell lines was selective towards mitochondria (2 and 3); after long-term incubation with 2, mitochondria were undetectable using a mitochondrial probe. A similar effect on mitochondria was observed also for environmental competitors of Q. cyanescens from the genus Geosmithia

Effect of quambalarine B (2) and mompain (3) on the adenocarcinoma cell line HeLa.

<p>0.25% DMSO was used as a solvent and as a control. Upper panels: visualization of mitochondria using MitoTracker Red CMXRos (red), actin cytoskeleton using Phalloidin (green), lysosomes using monoclonal antibody MEM 259 recognizing the lysosomal protein LAMP2 (white) and nuclei with DAPI (blue). Lower panels: simultaneous visualization of mitochondria, lysosomes and nuclei only.</p
