    Avaliação do estado do gene Epidermal Growth Factor Receptor (EGFR), por Silver In Situ Hibridization (SISH), tem-se destacado como biomarcador preditivo na resposta à terapêutica. O principal objectivo foi optimizar a etapa de recuperação por calor da metodologia automatizada SISH Dual-Colour, em carcinomas pulmonares fixados em formol durante 24 e 72 horas. A optimização levou a um aumento da preservação do contorno nuclear e da intensidade e contraste dos sinais para os dois tempos de fixação, permitindo avaliar o estado do EGFR em 83,3% dos casos em estudo. A SISH Dual-Colour é uma alternativa para avaliar o estado do EGFR.Assessment of the status of the gene Epidermal Growth Factor Receptor (EGFR), by Silver in Situ Hibridization (SISH) has been highlighted as a predictive biomarker in the response to therapy. The main objective was to optimize the recovery stage of heat-automated methodology SISH Dual-Colour in lung carcinoma with 24 and 72 hours of formaldehyde fixation. The optimization led to an increase in the preservation of the nuclear contour, intensity and contrast of the signals for both fixation times, allowing evaluation of EGFR state in 83.3% of studied cases. This includes individual satisfaction with his/her affective, loving, and sexual relationships.     Selectivity of Rynaxypyr for three species of Phytoseiid mites relevant to coffee in Brazil

    Among the predaceous mites, those of the Phytoseiidae family are the most important and studied. The phytophagous mites Brevipalpus phoenicis (Geijskes, 1939) (Tenuipalpidae) and Oligonychus ilicis (McGregor, 1917) (Tetranychidae), on coffee trees (Coffea spp.), are frequently found in combination with the predaceous mites Iphiseiodes zuluagai Denmark & Muma, 1972, Amblyseius herbicolus (Chant, 1959) and Euseius citrifolius Denmark & Muma, 1970, among others. The purpose of this research was to study the effects of the insecticide rynaxypyr (chlorantraniliprole 200 SC) on these three species of Phytoseiidae, relevant to coffee and citrus, following standard laboratory procedures. Mated female mites were exposed to fresh-dried residues on a glass surface, with 7 treatments, 5 mites per glass plate and 6 replicates, in a completely randomized experimental design. Each test lasted 8 days, with a daily count of the surviving females and of eggs laid. Rynaxypyr, in all tested concentrations (15, 30, 50, 100 and 500 mg a.i./liter of water), was selective for the studied species, A. herbicolus, I. zuluagai, and E. citrifolius. Overall the treatments resulted in low mortality rates and negligible impact on the reproduction. Therefore, based on IOBC standards, rynaxypyr can be classified as not harmful (Class 1), comparable to the agrochemical hexythiazox equivalent to a harmless standard of selectivity in the laboratory. Rynaxypyr is therefore a complement to programs of integrated pest management, to preserve the populations of predatory mites in crops of coffee and citrus, among others, in Brazil.Among the predaceous mites, those of the Phytoseiidae family are the most important and studied.     Ultrasound-assisted biosynthesis of novel methotrexate-conjugates

    New methotrexate-acylglycerols and methotrexate-cyclodextrins (, and -CD) conjugates were obtained via esterification or transesterification reactions. All reactions were catalysed by esterases namely immobilized Lipase from Candida antarctica B and Lipase from Thermomyces lanuginosus. The use of ultrasound to assist the reactions revealed to be a key factor to obtain high conversion yields on both MTX conjugates. Transesterification reactions including long chain triacylglycerols were only successful when ultrasound was applied. In cyclodextrins esterification a higher number of MTX molecules was also linked to cyclodextrins when ultrasound was used.     LC-HRMS profiling of paralytic shellfish toxins in Mytilus galloprovincialis after a Gymnodinium catenatum bloom

    Saxitoxin and its more than 50 analogues are a group of naturally occurring neurotoxins collectively designated as paralytic shellfish toxins (PSTs). PSTs are toxic to humans and maximum legal limits in seafood have been implemented by regulatory authorities worldwide. In the European Union, monitoring of PSTs is performed using the AOAC Official Method 2005.06, based on liquid chromatography coupled with fluorescence detection (LC- FLD). However, this method has been suggested to not effectively detect the emerging C-11 hydroxyl (M-toxins) and benzoate (GC-toxins) analogues, with these analogues currently not being surveyed in monitoring programs. In this study, a liquid chromatography-high resolution mass spectrometry (LC-HRMS) method was used to search for these emerging PSTs in mussels, Mytilus galloprovincialis, contaminated following an intense Gymnodinium catenatum bloom in the Tagus estuary (Lisbon, Portugal). Five M-toxins (M1, M2, M6, dcM6, and dcM10), but no GC-toxins, were detected in the mussels’ whole-soft body tissue. Moreover, the classical PSTs (C1 to C4, GTX 4 to GTX6, dcGTX1 to dcGTX4, dcSTX, dcNEO, and STX) were also found and comprised the largest fraction of the PSTs’ profile. The presence of unregulated PSTs in edible mussel samples suggests potential seafood safety risks and urges further research to determine the frequency of these analogues in seafood and their contribution to toxicityinfo:eu-repo/semantics/publishedVersio
