    O “louva-a-deus-de-cobra”, Phibalosoma sp. (Insecta, Phasmida), segundo a percepção dos moradores de Pedra Branca, Santa Terezinha, Bahia, Brasil

    This paper discusses the way the dwellers from the town of Pedra Branca, located in the state of Bahia, northeastern Brazil, perceive and relate with the stick insects (Insect, Phasmida). Part of the data was obtained in one day in May 2004 by performing open-ended interviews with 12 children and adolescents, whose ages ranged from 5 to 17 years old. Additional data was obtained from February to May 2001 by performing open-ended interviews with 25 men and 26 women, whose ages ranged from 24 to 82 years old. Results show that people usually gather insects from three different taxa (Phasmida, Mantodea, and Tettigoniidae) under the label “snake-mantis”. Interviewees have provided information related to the ethnotaxonomy, behavior, ontogeny, morphology, habitat, and food ecology of the stick insects. The most commented and widespread knowledge regarding these insects refers to their biotransformation or metamorphic process, where it is believed they originate from plants, and then change themselves into snakes locally known as “cobra-de-cipó”. The metamorphosis of the snake-mantis has significance in the way people understand and relate with it.Este artigo discute como os habitantes do povoado de Pedra Branca, localizado no estado da Bahia, nordeste do Brasil, percebem e se relacionam com os bichos-pau (Insecta, Phasmida). Parte dos dados foi obtida em um dia de trabalho de campo realizado em maio de 2004 através de entrevistas semi-estruturadas com 12 crianças e adolescentes, cujas idades variaram dos 5 aos 17 anos. A estes dados foram acrescentados aqueles obtidos de fevereiro a maio de 2001 por meio de entrevistas semi-estruturadas realizadas com 25 homens e 26 mulheres, cuja faixa etária variou de 24 a 82 anos. Os resultados mostram que os indivíduos costumam reunir insetos de três táxons diferentes (Phasmida, Mantodea e Tettigoniidae) sob o rótulo “louva-a-deus-de-cobra”. Os entrevistados forneceram informações sobre etnotaxonomia, comportamento, ontogenia, morfologia, habitat e ecologia trófica do bicho-pau. O conhecimento local mais marcante relacionado com esses insetos se refere a sua biotransformação ou processo metamórfico, onde se acredita que eles se originam de plantas e depois “se transformam” em cobras conhecidas localmente como “cobra-de-cipó”. Conclui-se que a metamorfose do bicho-pau resulta importante no modo como os indivíduos compreendem e se relacionam com esse inseto

    Evidences of herbal medicine-derived natural products effects in inflammatory lung diseases

    Pulmonary inflammation is a hallmark of many respiratory diseases such as asthma, chronic obstructive pulmonary disease (COPD), and acute respiratory syndrome distress (ARDS). Most of these diseases are treated with anti-inflammatory therapy in order to prevent or to reduce the pulmonary inflammation. Herbal medicine-derived natural products have been used in folk medicine and scientific studies to evaluate the value of these compounds have grown in recent years. Many substances derived from plants have the biological effects in vitro and in vivo, such as flavonoids, alkaloids, and terpenoids. Among the biological activities of natural products derived from plants can be pointed out the anti-inflammatory, antiviral, antiplatelet, antitumor anti-allergic activities, and antioxidant. Although many reports have evaluated the effects of these compounds in experimental models, studies evaluating clinical trials are scarce in the literature.     Mitochondrial activity in the frontal cortex area 8 and angular gyrus in Parkinson's disease and Parkinson's disease with dementia

    Altered mitochondrial function is characteristic in the substantia nigra in Parkinson's disease (PD). Information about mitochondria in other brain regions such as the cerebral cortex is conflicting mainly because most studies have not contemplated the possibility of variable involvement depending on the region, stage of disease progression and clinical symptoms such as the presence or absence of dementia. RT-qPCR of 18 nuclear mRNAs encoding subunits of mitochondrial complexes and 12 mRNAs encoding energy metabolism-related enzymes; western blotting of mitochondrial proteins; and analysis of enzymatic activities of complexes I, II, II, IV and V of the respiratory chain were assessed in frontal cortex area 8 and the angular gyrus of middle-aged individuals (MA), and those with incidental PD (iPD), long-lasting PD with parkinsonism without dementia (PD) and long-lasting PD with dementia (PDD). Up-regulation of several genes was found in frontal cortex area 8 in PD when compared with MA and in the angular gyrus in iPD when compared with MA. Marked down-regulation of genes encoding mitochondrial subunits and energy metabolism-related enzymes occurs in frontal cortex but only of genes coding for energy metabolism-related enzymes in the angular gyrus in PDD. Significant decrease in the protein expression levels of several mitochondrial subunits encoded by these genes occurs in frontal cortex area 8 and angular gyrus in PDD. Moreover, expression of MT-ND1 which is encoded by mitochondrial DNA is also reduced in PDD. Reduced enzymatic activity of complex III in frontal cortex area 8 and angular gyrus is observed in PD, but dramatic reduction in the activity of complexes I, II, II and IV in both regions characterizes PDD. Dementia in the context of PD is linked to region-specific deregulation of genomic genes encoding subunits of mitochondrial complexes and to marked reduction in the activity of mitochondrial complexes I, II, III and IV

    Drug discovery for psychiatric disorders using high-content single-cell screening of signaling network responses ex vivo

    There is a paucity of efficacious new compounds to treat neuropsychiatric disorders. We present a novel approach to neuropsychiatric drug discovery based on high-content characterization of druggable signaling network responses at the single-cell level in patient-derived lymphocytes ex vivo. Primary T lymphocytes showed functional responses encompassing neuropsychiatric medications and central nervous system ligands at established (e.g., GSK-3?) and emerging (e.g., CrkL) drug targets. Clinical application of the platform to schizophrenia patients over the course of antipsychotic treatment revealed therapeutic targets within the phospholipase C?1-calcium signaling pathway. Compound library screening against the target phenotype identified subsets of L-type calcium channel blockers and corticosteroids as novel therapeutically relevant drug classes with corresponding activity in neuronal cells. The screening results were validated by predicting in vivo efficacy in an independent schizophrenia cohort. The approach has the potential to discern new drug targets and accelerate drug discovery and personalized medicine for neuropsychiatric conditions

    Exploring cellular markers of metabolic syndrome in peripheral blood mononuclear cells across the neuropsychiatric spectrum

    Recent evidence suggests that comorbidities between neuropsychiatric conditions and metabolic syndrome may precede and even exacerbate long-term side-effects of psychiatric medication, such as a higher risk of type 2 diabetes and cardiovascular disease, which result in increased mortality. In the present study we compare the expression of key metabolic proteins, including the insulin receptor (CD220), glucose transporter 1 (GLUT1) and fatty acid translocase (CD36), on peripheral blood mononuclear cell subtypes from patients across the neuropsychiatric spectrum, including schizophrenia, bipolar disorder, major depression and autism spectrum conditions (n = 25/condition), relative to typical controls (n = 100). This revealed alterations in the expression of these proteins that were specific to schizophrenia. Further characterization of metabolic alterations in an extended cohort of first-onset antipsychotic drug-naïve schizophrenia patients (n = 58) and controls (n = 63) revealed that the relationship between insulin receptor expression in monocytes and physiological insulin sensitivity was disrupted in schizophrenia and that altered expression of the insulin receptor was associated with whole genome polygenic risk scores for schizophrenia. Finally, longitudinal follow-up of the schizophrenia patients over the course of antipsychotic drug treatment revealed that peripheral metabolic markers predicted changes in psychopathology and the principal side effect of weight gain at clinically relevant time points. These findings suggest that peripheral blood cells can provide an accessible surrogate model for metabolic alterations in schizophrenia and have the potential to stratify subgroups of patients with different clinical outcomes or a greater risk of developing metabolic complications following antipsychotic therapy.This work was supported by grants from the Stanley Medical Research Institute (SMRI); the Engineering and Physical Sciences Research Council UK (EPSRC); the Dutch Government-funded Virgo consortium (ref.     Social Security Coverage and the Labor Market in Developing Countries

    This paper analyzes the reasons behind the low rates of contribution to social security programs in developing countries. Using a large set of harmonized household surveys from Latin America we compare contribution patterns among wage employees, for whom participation is compulsory, with contribution patterns among self-employed workers, for whom participation is often voluntary. In all countries, contribution rates among salaried workers are similarly correlated with education, earnings, size of the employer, household characteristics and age. In addition, contribution patterns among salaried workers are highly correlated with contribution patterns among the self-employed. Our results indicate that on average more than 30 percent of the explained within-country variance in contributions patterns may be accounted for by individuals low willingness to participate in old-age pension programs. Nonetheless, we also find evidence suggesting that some workers are rationed out of social security against their will
