    MEK1 Inhibits Cardiac PPARα Activity by Direct Interaction and Prevents Its Nuclear Localization

    BACKGROUND: The response of the postnatal heart to growth and stress stimuli includes activation of a network of signal transduction cascades, including the stress activated protein kinases such as p38 mitogen-activated protein kinase (MAPK), c-Jun NH2-terminal kinase (JNK) and the extracellular signal-regulated kinase (ERK1/2) pathways. In response to increased workload, the mitogen-activated protein kinase kinase (MAPKK) MEK1 has been shown to be active. Studies embarking on mitogen-activated protein kinase (MAPK) signaling cascades in the heart have indicated peroxisome-proliferators activated-receptors (PPARs) as downstream effectors that can be regulated by this signaling cascade. Despite the importance of PPARα in controlling cardiac metabolism, little is known about the relationship between MAPK signaling and cardiac PPARα signaling. METHODOLOGY/PRINCIPAL FINDING: Using co-immunoprecipitation and immunofluorescence approaches we show a complex formation of PPARα with MEK1 and not with ERK1/2. Binding of PPARα to MEK1 is mediated via a LXXLL motif and results in translocation from the nucleus towards the cytoplasm, hereby disabling the transcriptional activity of PPARα. Mice subjected to voluntary running-wheel exercise showed increased cardiac MEK1 activation and complex formation with PPARα, subsequently resulting in reduced PPARα activity. Inhibition of MEK1, using U0126, blunted this effect. CONCLUSION: Here we show that activation of the MEK1-ERK1/2 pathway leads to specific inhibition of PPARα transcriptional activity. Furthermore we show that this inhibitory effect is mediated by MEK1, and not by its downstream effector kinase ERK1/2, through a mechanism involving direct binding to PPARα and subsequent stimulation of PPARα export from the nucleus

    O “louva-a-deus-de-cobra”, Phibalosoma sp. (Insecta, Phasmida), segundo a percepção dos moradores de Pedra Branca, Santa Terezinha, Bahia, Brasil

    This paper discusses the way the dwellers from the town of Pedra Branca, located in the state of Bahia, northeastern Brazil, perceive and relate with the stick insects (Insect, Phasmida). Part of the data was obtained in one day in May 2004 by performing open-ended interviews with 12 children and adolescents, whose ages ranged from 5 to 17 years old. Additional data was obtained from February to May 2001 by performing open-ended interviews with 25 men and 26 women, whose ages ranged from 24 to 82 years old. Results show that people usually gather insects from three different taxa (Phasmida, Mantodea, and Tettigoniidae) under the label “snake-mantis”. Interviewees have provided information related to the ethnotaxonomy, behavior, ontogeny, morphology, habitat, and food ecology of the stick insects. The most commented and widespread knowledge regarding these insects refers to their biotransformation or metamorphic process, where it is believed they originate from plants, and then change themselves into snakes locally known as “cobra-de-cipó”. The metamorphosis of the snake-mantis has significance in the way people understand and relate with it.Este artigo discute como os habitantes do povoado de Pedra Branca, localizado no estado da Bahia, nordeste do Brasil, percebem e se relacionam com os bichos-pau (Insecta, Phasmida). Parte dos dados foi obtida em um dia de trabalho de campo realizado em maio de 2004 através de entrevistas semi-estruturadas com 12 crianças e adolescentes, cujas idades variaram dos 5 aos 17 anos. A estes dados foram acrescentados aqueles obtidos de fevereiro a maio de 2001 por meio de entrevistas semi-estruturadas realizadas com 25 homens e 26 mulheres, cuja faixa etária variou de 24 a 82 anos. Os resultados mostram que os indivíduos costumam reunir insetos de três táxons diferentes (Phasmida, Mantodea e Tettigoniidae) sob o rótulo “louva-a-deus-de-cobra”. Os entrevistados forneceram informações sobre etnotaxonomia, comportamento, ontogenia, morfologia, habitat e ecologia trófica do bicho-pau. O conhecimento local mais marcante relacionado com esses insetos se refere a sua biotransformação ou processo metamórfico, onde se acredita que eles se originam de plantas e depois “se transformam” em cobras conhecidas localmente como “cobra-de-cipó”. Conclui-se que a metamorfose do bicho-pau resulta importante no modo como os indivíduos compreendem e se relacionam com esse inseto

    A cultivar BRS 7762 Supremo é resultante do cruzamento biparental entre as linhagens W22-34 e VAN163, realizado na Embrapa Arroz e Feijão, em 1988. Apresenta hábito de crescimento indeterminado, com porte da planta ereto e boa resistência ao acamamento, em qualquer sistema de plantio. A arquitetura da planta possibilita a colheita mecânica; também é interessante os produtores que plantam feijão intercalado nos cafezais, prática muito comum no Estado de Minas Gerais, uma vez que a ausência de "guias" longas, facilita o manejo da cultura do feijoeiro e do café, ao qual está associada.bitstream/CNPAF-2009-09/27960/1/comt_160.pd

    Dietary supplementation with chestnut (Castanea sativa) reduces abdominal adiposity in FVB/n mice: a preliminary study

    The production of chestnut (Castanea sativa Miller) is mostly concentrated in Europe. Chestnut is recognized by its high content of antioxidants and phytosterols. This work aimed to evaluate the e ects of dietary chestnut consumption over physiological variables of FVB/n mice. Eighteen FVB/n male 7-month-old mice were randomly divided into three experimental groups (n = 6): 1 (control group) fed a standard diet; 2 fed a diet supplemented with 0.55% (w/w) chestnut; and 3 supplemented with 1.1% (w/w) chestnut. Body weight, water, and food intake were recorded weekly. Following 35 days of supplementation, the mice were sacrificed for the collection of biological samples. Chestnut supplementation at 1.1% reduced abdominal adipose tissue. Lower serum cholesterol was also observed in animals supplemented with chestnut. There were no significant di erences concerning the incidence of histological lesions nor in biochemical markers of hepatic damage and oxidative stress.     MiR-337-3p Promotes Adipocyte Browning by Inhibiting TWIST1

    The prevalence of metabolic syndrome (MetS) and obesity is an alarming health issue worldwide. Obesity is characterized by an excessive accumulation of white adipose tissue (WAT), and it is associated with diminished brown adipose tissue (BAT) activity. Twist1 acts as a negative feedback regulator of BAT metabolism. Therefore, targeting Twist1 could become a strategy for obesity and metabolic disease. Here, we have identified miR-337-3p as an upstream regulator of Twist1. Increased miR-337-3p expression paralleled decreased expression of TWIST1 in BAT compared to WAT. Overexpression of miR-337-3p in brown pre-adipocytes provoked a reduction in Twist1 expression that was accompanied by increased expression of brown/mitochondrial markers. Luciferase assays confirmed an interaction between the miR-337 seed sequence and Twist1 3 0UTR. The inverse relationship between the expression of TWIST1 and miR-337 was finally validated in adipose tissue samples from non-MetS and MetS subjects that demonstrated a dysregulation of the miR-337-Twist1 molecular axis in MetS. The present study demonstrates that adipocyte miR-337-3p suppresses Twist1 repression and enhances the browning of adipocytes

    É com esse objetivo, que as instituições que trabalham no melhoramento dessa cultura no estado, Universidade Federal de Lavras (UFLA), Universidade Federal de Viçosa (UFV), Embrapa Arroz e Feijão e Empresa de Pesquisa Agropecuária de Minas Gerais (Epamig), têm somado esforços na avaliação de novas linhagens.
