    Effectiveness of different cellulose-based filtration materials against inhalation of SARS-CoV-2-like particles

    The respiratory protection equipment (RPE) used by health professionals consists of an essential device to prevent infectious diseases, especially those caused by biological agents such as the coronavirus (SARS-CoV-2). The current epidemiological panorama is worrying, and the context of creation and production of the mask has emerged as an alternative to RPE to face the public health crisis worldwide. The aim of this work is to present a low-cost alternative as an FFP2-like filter for a reusable respirator face mask. This study presents the comparison of different cellulose-based filtering materials performed by retention testing, time saturation testing, aerosol penetration testing, nanoparticle (~140 nm) filtration testing, bacterial filtration efficiency (BFE), analysis of material morphology and usability. The reusable respirator face mask used in this study is an open-source innovation, using 3D printing. Cotton disc proved to be the best filter material for the reusable mask, with satisfactory results and a performance similar to that shown by the N95-type mask. The cotton disc ensured effectiveness over 6 h of use, and after that, the reusable respirator face mask (here, Delfi-TRON®) needed to be sanitized and replenished with a new cotton disc.     Harlequin ichthyosis is a rare autosomal recessive congenital disease in which neonates present generalized hyperkeratotic plaques and deep fissures, ectropion, eclabium, malformation of the auricular pavilion and typical facies. Although several complications related to the skin restriction may occur, support in intensive care and early introduction of systemic retinoids, such as acitretin, have significantly contributed to patients' survival and improved prognosis. The purpose of this report is to present a rare case of harlequin ichthyosis and to discuss strategies for early diagnosis and first supportive care.Ictiose arlequim é uma doença congênita autossómica recessiva rara, na qual os recém-nascidos apresentam placas de hiperqueratose generalizadas e fissuras profundas, ectrópio, eclábio, malformação do pavilhão auricular e fácies típicas. Embora várias complicações relacionadas à restrição cutânea possam ocorrer, o suporte em terapia intensiva e a introdução precoce de retinóides sistémicos, como a acitretina, têm contribuído significativamente para a melhoria da sobrevida e do prognóstico dos doentes.     Hepatitis C Virus Infection as a Traumatic Experience

    Objective The purpose of this study was to evaluate whether individuals consider their HCV infection to be a potentially traumatic experience. Additionally, we investigated its association with Post-Traumatic Stress Disorder (PTSD) and the impact of PTSD diagnosis on health-related quality of life (HRQoL) in HCV infected subjects. Methods We conducted a cross-sectional survey of 127 HCV-infected outpatients recruited at a University Hospital in Salvador, Brazil. All subjects answered an orally-administered questionnaire to gather clinical and socio-demographic data. We investigated traumatic experiences and the subject's perception of the disease using the Trauma History Questionnaire. PTSD and other psychiatric diagnoses were assessed through the Mini International Neuropsychiatric Interview-Brazilian Version 5.0.0 (M.I.N.I. PLUS). HRQoL was assessed using Short-Form 36 (SF-36). Results Approximately 38.6% of the patients considered hepatitis C to be a traumatic experience. Of these, 60.7% had a PTSD diagnosis. PTSD was associated with significant impairment in quality of life for individuals in seven SF-36 domains as shown bymultivariate analysis: Role-Physical (β: −24.85; 95% CI: −42.08; −7.61), Bodily Pain (β: −19.36; 95% CI: −31.28; −7.45), General Health (β: −20.79; 95% CI: −29.65; −11.92), Vitality (β: −11.92; 95% CI: −20.74; −3.1), Social Functioning (β: −34.73; 95% CI: −46.79; −22.68), Role-Emotional (β: −26.07; 95% CI: −44.61; −7.53), Mental Health (β: −17.46; 95% CI: −24.38; −10.54). Conclusion HCV is frequently a traumatic experience and it is strongly associated with PTSD diagnosis. PTSD significantly impaired HRQoL

    Reference genes for quantitative reverse transcription-polymerase chain reaction expression studies in wild and cultivated peanut

    <p>Abstract</p> <p>Background</p> <p>Wild peanut species (<it>Arachis </it>spp.) are a rich source of new alleles for peanut improvement. Plant transcriptome analysis under specific experimental conditions helps the understanding of cellular processes related, for instance, to development, stress response, and crop yield. The validation of these studies has been generally accomplished by quantitative reverse transcription-polymerase chain reaction (qRT-PCR) which requires normalization of mRNA levels among samples. This can be achieved by comparing the expression ratio between a gene of interest and a reference gene which is constitutively expressed. Nowadays there is a lack of appropriate reference genes for both wild and cultivated <it>Arachis</it>. The identification of such genes would allow a consistent analysis of qRT-PCR data and speed up candidate gene validation in peanut.</p> <p>Results</p> <p>A set of ten reference genes were analyzed in four <it>Arachis </it>species (<it>A. magna</it>; <it>A. duranensis</it>; <it>A. stenosperma </it>and <it>A. hypogaea</it>) subjected to biotic (root-knot nematode and leaf spot fungus) and abiotic (drought) stresses, in two distinct plant organs (roots and leaves). By the use of three programs (GeNorm, NormFinder and BestKeeper) and taking into account the entire dataset, five of these ten genes, <it>ACT1 </it>(actin depolymerizing factor-like protein), <it>UBI1 </it>(polyubiquitin), <it>GAPDH </it>(glyceraldehyde-3-phosphate dehydrogenase), <it>60S </it>(60S ribosomal protein L10) and <it>UBI2 </it>(ubiquitin/ribosomal protein S27a) emerged as top reference genes, with their stability varying in eight subsets. The former three genes were the most stable across all species, organs and treatments studied.</p> <p>Conclusions</p> <p>This first in-depth study of reference genes validation in wild <it>Arachis </it>species will allow the use of specific combinations of secure and stable reference genes in qRT-PCR assays. The use of these appropriate references characterized here should improve the accuracy and reliability of gene expression analysis in both wild and cultivated Arachis and contribute for the better understanding of gene expression in, for instance, stress tolerance/resistance mechanisms in plants.</p

    CRESCIMENTO INICIAL DE AROEIRA DO SERTÃO (Myracrodruon urundeuva Allemão) EM DIFERENTES SUBSTRATOS

    RESUMOEste trabalho buscou avaliar o crescimento de Myracrodruon urundeuvaAllemão produzida em substratos preparados com lodo de esgoto, composto orgânico e esterco bovino. O trabalho foi conduzido em casa de vegetação por 120 dias no Viveiro do Centro de Referência em Conservação da Natureza e Recuperação de Áreas Degradadas – CRAD, Brasília-DF. Foram testados quatro tipos de adubos: (1) Osmocote®; (2) esterco bovino; (3) composto orgânico; e (4) lodo de esgoto seco. Cada adubo foi submetido a três composições diferentes: (1) 25%, (2) 50% e (3) 75%, com exceção da testemunha e do Osmocote®, totalizando 11 tratamentos com 10 repetições cada. As variáveis analisadas foram: Diâmetro do Coleto (DC), Altura da Muda (H), Número de Folhas (NF), Matéria Fresca de Parte Aérea (MFPA), Matéria Seca de Parte Aérea (MSPA), Matéria Fresca de Raiz (MFR), Matéria Seca de Raiz (MSR) e Índice de Qualidade de Dickson (IQD). As médias foram comparadas pelo teste de Tukey a 5% de probabilidade. Os resultados indicaram interação significativa entre os tratamentos. As maiores médias foram obtidas nos tratamentos com esterco bovino, seguidas do tratamento com composto. As plantas produzidas apenas com Osmocote® e com lodo de esgoto morreram. O resultado apontou a importância de incorporar matéria orgânica ao substrato para produção de mudas de M. urundeuva e o composto orgânico na proporção de 25% equivale ao esterco bovino na proporção de 25, 50 e 75%

    Inhibitory Receptors Are Expressed by Trypanosoma cruzi-Specific Effector T Cells and in Hearts of Subjects with Chronic Chagas Disease

    We had formerly demonstrated that subjects chronically infected with Trypanosoma cruzi show impaired T cell responses closely linked with a process of T cell exhaustion. Recently, the expression of several inhibitory receptors has been associated with T cell dysfunction and exhaustion. In this study, we have examined the expression of the cytotoxic T lymphocyte antigen 4 (CTLA-4) and the leukocyte immunoglobulin like receptor 1 (LIR-1) by peripheral T. cruzi antigen-responsive IFN-gamma (IFN-γ)-producing and total T cells from chronically T. cruzi-infected subjects with different clinical forms of the disease. CTAL-4 expression was also evaluated in heart tissue sections from subjects with severe myocarditis. The majority of IFN-γ-producing CD4+ T cells responsive to a parasite lysate preparation were found to express CTLA-4 but considerably lower frequencies express LIR-1, irrespective of the clinical status of the donor. Conversely, few IFN-γ-producing T cells responsive to tetanus and diphtheria toxoids expressed CTLA-4 and LIR-1. Polyclonal stimulation with anti-CD3 antibodies induced higher frequencies of CD4+CTAL-4+ T cells in patients with severe heart disease than in asymptomatic subjects. Ligation of CTLA-4 and LIR-1 with their agonistic antibodies, in vitro, reduces IFN-γ production. Conversely, CTLA-4 blockade did not improved IFN-γ production in response to T. cruzi antigens. Subjects with chronic T. cruzi infection had increased numbers of CD4+LIR-1+ among total peripheral blood mononuclear cells, relative to uninfected individuals and these numbers decreased after treatment with benznidazole. CTLA-4 was also expressed by CD3+ T lymphocytes infiltrating heart tissues from chronically infected subjects with severe myocarditis. These findings support the conclusion that persistent infection with T. cruzi leads to the upregulation of inhibitory receptors which could alter parasite specific T cell responses in the chronic phase of Chagas disease
