Harvesting Image Databases from The Web
The research work presented here includes data mining needs and study of their algorithm for various extraction purpose. It also includes work that has been done in the field of harvesting images from web. Here the proposed method is to harvest image databases from web. We can automatically generate a large number of images for a specified object. By applying concept of data mining and the algorithm from data mining which is used for extraction of data or harvesting images. A multimodal approach employing text ,metadata and visual features is used to gather many high-quality images from the web. The modules can be made to find query images by selecting images where nearby text is top ranked by the topic i.e., formation of image clusters then download associate images by using approaches like web search, image search and Google images. Apply re-ranking algorithm and then filtering process to harvest the images.Currently, image search gives a very low precision (only about 4%) and is not used for the harvesting experiments. Since the movements of the technologies are growing rapidly the kinds of work also need to be grown up. This work shows an approach to harvest a large number of images of a particular class automatically and to achieve this with high precision by providing training databases so that a new object model can be learned effortlessly. Many other tools also are available for harvesting images from web .An approach in this paper is original and up to the mark. EVALUATION OF IN VITRO ANTI-UROLITHIATIC ACTIVITY OF SOLANUM TUBEROSUM ON CALCIUM OXALATE CRYSTALS
Objective: Present study was designed to evaluate the anti-urolithiatic activity of Indian species of Solanun tuberosum on calcium oxalate crystal In vitro model.
Methods: Potato powder was prepared by cutting, grinding and dried in hot air oven at 60-65 ℃. In vitro model of Calcium oxalate crystal was used.
Results: It was observed that the sample prepared from Solanumtuberosum, both potato ash as well as potato powder, had ability to act as anti-urolithical drug due to the presence of saponin and potassium.
Conclusion: From the present study, we can conclude that the sample prepared from Solanumtuberosum, had the ability to act as anti-urolithical drug and in vivo study can be carried out for further investigation
THE USE OF HPLC AS A TOOL FOR NEONATAL CORD BLOOD SCREENING OF HAEMOGLOBINOPATHY - A VALIDATION STUDY
Background: Newborn cord blood screening identifies infants with underlying haemoglobinopathies before they develop the characteristic symptoms or sequelae.
Aims: This study was performed to validate the interpretation high-performance chromatography (HPLC) along with complete blood count (CBC) results as a tool for universal neonatal screening of hemoglobin disorders in Oman.
Methods: HPLC and CBC data on subjects who participated in the National Neonatal screening program at birth were obtained from archival records. The results recorded at birth were compared with a second study performed on the same subjects, after approval from the local medical research and ethics committee.
Results: Only 290 subjects from amongst the original cohort of 3740 newborns could be recalled between April 2010 to March 2011, to repeat HPLC and CBC, as well as perform confirmatory DNA studies, wherever necessary. All these subjects had been documented to show an initial abnormal result. 31 cases who had no HbA at birth on HPLC were confirmed as either homozygous β-thalassaemia major (n=5 subjects) or homozygous sickle cell anemia (n=26 subjects) by appropriate DNA analysis. Additionally, amongst 151 subjects, 72 subjects were studied in the initial study by Hb Bart’s quantitation using aalpha thalassaemia short program at birth. In this cohort, 42 subjects with Hb Bart’s >1% at birth could be confirmed as having either deletional or non-deletional thalassaemia by GAP PCR studies. No case of HbH was detected in this cohort. Further, carrier status for structural hemoglobin variants (HbS, HbC, HbD, HbE) (n=67) and beta thalassaemia allele with low HbA at birth (n=29 out of 41) were confirmed by relevant molecular studies.
Conclusions: The study validated the earlier observation by 100% concordance with results of CBC and HPLC. Presence of Hb Bart’s at birth does not always mean the presence of alpha thalassemia, as subjects with Hb Bart’s was below 1% by quantitation, were shown to be normal by molecular studies.

Non-cell autonomous OTX2 transcription factor regulates anxiety-related behaviors in the mouse
The Otx2 homeoprotein transcription factor is expressed in the dopaminergic neurons of the ventral tegmental area, a mesencephalic nucleus involved in the control of complex behaviors through its projections to limbic structures, including the ventral hippocampus, amygdala, nucleus accumbens and prefrontal cortex. We find adult mice heterozygous for Otx2 show a hypoanxious phenotype in light-dark box and elevated plus maze paradigms. However, the number of dopaminergic neurons, the integrity of their axons, their projection patterns in target structures, and the amounts of dopamine and dopamine metabolites in targets structures were not modified in the Otx2 mutant. Because OTX2 is expressed by the choroid plexus, secreted into cerebrospinal fluid and transferred to parvalbumin interneurons of the cortex, hippocampus, and amygdala, we investigated if the hypoanxiety of Otx2 heterozygous mice could result from the decreased synthesis of Otx2 in the choroid plexus. Indeed, hypoanxious phenotype was reversed by the overexpression of Otx2 specifically in choroid plexus of adult Otx2 heterozygous mice, while hypoanxious phenotype could be induced in adult wild type mice by lowering OTX2 levels in the cerebrospinal fluid. Taken together, OTX2 synthesis by the choroid plexus followed by its secretion into the cerebrospinal fluid is an important regulator of the anxiety phenotype in the mouse. All rights reserved. Evaluation of bruising susceptibility and response of pears under impact loading through finite element analysis
Mechanical damage and bruising of fruit is a critical problem in the food industry. Minimizing brusing and damage can be achieved by designing energy-absorbing structures and packaging systems in order to ensure the long-term quality of fresh produce. The aim of this study is to investigate the response and bruise susceptibility of pears under impact loading conditions through finite element analysis (FEA) methods. In this paper, three impact heights (0.25 m, 0.5 m, and 1.0 m), four impact material surfaces (poplar wood, rubber, cardboard, and acrylonitrile butadiene styrene (ABS) plastic), two packaging sizes (standard 0.22″ and sandwich lattice 2.1″), and three impact design structures (rigid, corrugated, and honeycomb) are considered. Based on mesh sensitivity analysis, a mesh element of 1.5 mm was adopted for all simulations, assuring the accuracy of results and considering the trade-off between mesh size and computational time. The response surface analysis approach was utilized in order to develop predictive empirical models related to pear bruising. Results revealed that the rubber-based impact platform yielded minimal bruise susceptibility at all heights, while standard-sized corrugated cardboard performed best at a height of 0.25 m. Furthermore, single, double, and triple layers of packaging cardboard were tested. We observed that adding a second soft layer of corrugated cardboard reduced the stress on the pear by around 33%. However, adding a third layer only reduced stress by 5%. The 3D-printed honeycomb ABS has potential as protective packaging but would require further investigations and parameter optimization. Stacking multiple layers of cardboard on top of each other is a cost-effective solution that could improve damping and, therefore, ensure good quality and increase the shelf life of the fresh produce. This study will help decision-makers select the optimal energy-absorbing material for cushioning and packaging designs in order to improve the handling and post-harvesting logistics of fresh produce

Spray washes of organic acids to decontaminate pork carcass tissues in India
Medical and commercial requirements to extend safe, high quality shelf life of food, have focussed attention on decontamination systems. Organic acid sprays of acetic, propionic and lactic at 3Kg/cm\u27 pressure and 1%, 2% and 3% concentrations, individually and in combination, were used to decontaminate pork samples. It was observed that out of 3 acids lactic acid at 2% was found to reduce the total viable count (TVC) by 0.07 log units whereas the combination treatment of 3% acetic and propionic acids resulted in 0.09 log unit reduction of TVC without affecting the color and odour score. The shelf life, on analysis, was sound to almost double at refrigeration storage as compared to control. Thus, 2 % lactic acid and 3% acetic + propionic acids offer scope for reduction in microflora and suppression of pathogen proliferation on pork samples during refrigeration storage and transport, thereby improving meat quality and shelf life
Crystal structure of the proteasomal deubiquitylation module Rpn8-Rpn11
The ATP-dependent degradation of polyubiquitylated proteins by the 26S proteasome is essential for the maintenance of proteome stability and the regulation of a plethora of cellular processes. Degradation of substrates is preceded by the removal of polyubiquitin moieties through the isopeptidase activity of the subunit Rpn11. Here we describe three crystal structures of the heterodimer of the Mpr1-Pad1-N-terminal domains of Rpn8 and Rpn11, crystallized as a fusion protein in complex with a nanobody. This fusion protein exhibits modest deubiquitylation activity toward a model substrate. Full activation requires incorporation of Rpn11 into the 26S proteasome and is dependent on ATP hydrolysis, suggesting that substrate processing and polyubiquitin removal are coupled. Based on our structures, we propose that premature activation is prevented by the combined effects of low intrinsic ubiquitin affinity, an insertion segment acting as a physical barrier across the substrate access channel, and a conformationally unstable catalytic loop in Rpn11. The docking of the structure into the proteasome EM density revealed contacts of Rpn11 with ATPase subunits, which likely stabilize the active conformation and boost the affinity for the proximal ubiquitin moiety. The narrow space around the Rpn11 active site at the entrance to the ATPase ring pore is likely to prevent erroneous deubiquitylation of folded proteins
