    De Novo Transcriptome Assembly of Cucurbita Pepo L. Leaf Tissue Infested by Aphis Gossypii

    Zucchini (Cucurbita pepo L.), extensively cultivated in temperate areas, belongs to the Cucurbitaceae family and it is a species with great economic value. One major threat related to zucchini cultivation is the damage imposed by the cotton/melon aphid Aphis gossypii Glover (Homoptera: Aphididae). We performed RNA-sequencing on cultivar "San Pasquale" leaves, uninfested and infested by A. gossypii, that were collected at three time points (24, 48, and 96 h post infestation). Then, we combined all high-quality reads for de novo assembly of the transcriptome. This resource was primarily established to be used as a reference for gene expression studies in order to investigate the transcriptome reprogramming of zucchini plants following aphid infestation. In addition, raw reads will be valuable for new experiments based on the latest bioinformatic tools and analytical approaches. The assembled transcripts will serve as an important reference for sequence-based studies and for primer design. Both datasets can be used to support/improve the prediction of protein-coding genes in the zucchini genome, which has been recently released into the public domain

    An Environmentally Friendly Nb–P–Si Solid Catalyst for Acid-Demanding Reactions

    Here, we report the structural characteristics, the surface properties, and the catalytic performances of a Nb–P–Si ternary oxide material (2.5Nb2O5·2.5P2O5·95SiO2, 2.5NbP) in two reactions of importance for biomass valorisation and green industrial production: hydrolysis of inulin and esterification of oleic acid with polyalcohol for biolubricant production. High dispersion of the Nb centers, ascertained by UV–vis–DRS, 29Si, 31P, and 1H solid-state NMR spectroscopy, is the key point for the successful activity of 2.5NbP. Intrinsic and effective acidities of the sample were studied by FT-IR of adsorbed pyridine in the absence and presence of water and by volumetric titrations of the acid sites in cyclohexane and in water, to enlighten the nature and amount of acid sites in different environments. For both studied reactions, 2.5NbP catalyst exhibits water-tolerant acidic sites, mainly BrĂžnsted ones, giving higher activity and better stability in the reaction medium than well-known niobium oxophosphate catalyst, which is considered one of the best water-tolerant acid catalysts

    Towards an agriculture knowledge ecosystem :A social life network for farmers in Sri Lanka

    We have developed and successfully trialled a Social Life Network (SLN); a Mobile Based Information System to support farming activities in Sri Lanka. It provides information required to support activities such as crop selection and cultivation planning in the context of farmer, farm location, season and task being performed. The system also provides a facility for farmers to sell farming related products and services to other farmers. The final system architecture evolved through a series of iterative relevance and design cycles based on Design Science Research methodology. In the first relevance cycle we identified farmer information needs, their current decision making patterns, and some possible ways to enhance their decision making process. In the first design cycles we developed the initial prototype to visualise a possible solution and in subsequent cycles a crop ontology to reorganise published crop information that would be queried in context and processes to empower farmers. Next we went through 2 cycles of creating functional prototypes, field testing with farmers and improving these to arrive at the final system. We noted that this system can enhance the flow of information in the agriculture domain by aggregating or disaggregating information produced by some stakeholders to be consumed by others. Based on this observation the overall architecture was reconceptualised as a Digital Knowledge Ecosystem

    BNT162b2 mRNA COVID-19 vaccine does not impact the honeymoon phase in type 1 diabetes: a case report

    Type 1 diabetes (T1D), which is caused by the autoimmune destruction of insulin-secreting pancreatic beta cells, represents a high-risk category requiring COVID-19 vaccine prioritization. Although COVID-19 vaccination can lead to transient hyperglycemia (vaccination-induced hyperglycemia; ViHG), its influence on the course of the clinical remission phase of T1D (a.k.a. "honeymoon phase") is currently unknown. Recently, there has been an increasing concern that COVID-19 vaccination may trigger autoimmune phenomena. We describe the case of a 24-year-old young Italian man with T1D who received two doses of the BNT162b2 mRNA (Pfizer-BioNTech) COVID-19 vaccine during a prolonged honeymoon phase. He experienced a transient impairment in glucose control (as evidenced by continuous glucose monitoring) that was not associated with substantial changes in stimulated C-peptide levels and islet autoantibody titers. Nonetheless, large prospective studies are needed to confirm the safety and the immunometabolic impact of the BNT162b2 vaccine in T1D patients during the honeymoon phase. Thus far, T1D patients who are going to receive COVID-19 vaccination should be warned about the possible occurrence of transient ViHG and should undergo strict postvaccination surveillance

    Mitigating Errors on Superconducting Quantum Processors through Fuzzy Clustering

    Quantum utility has been severely limited in superconducting quantum hardware until now by the modest number of qubits and the relatively high level of control and readout errors, due to the intentional coupling with the external environment required for manipulation and readout of the qubit states. Practical applications in the Noisy Intermediate Scale Quantum (NISQ) era rely on Quantum Error Mitigation (QEM) techniques, which are able to improve the accuracy of the expectation values of quantum observables by implementing classical post-processing analysis from an ensemble of repeated noisy quantum circuit runs. In this work, we focus on a recent QEM technique that uses Fuzzy C-Means (FCM) clustering to specifically identify measurement error patterns. For the first time, we report a proof-of-principle validation of the technique on a 2-qubit register, obtained as a subset of a real NISQ 5-qubit superconducting quantum processor based on transmon qubits. We demonstrate that the FCM-based QEM technique allows for reasonable improvement of the expectation values of single- and two-qubit gates based quantum circuits, without necessarily invoking state-of-the-art coherence, gate, and readout fidelities

    Evolution of the HIV-1 protease region in heavily pretreated HIV-1 infected patients receiving Atazanavir

    Background: Previous in vitro studies indicated that Atazanavir (ATV) has a distinct resistance profile than other protease inhibitors (PIs). In treatment-experienced patients ATV resistance is characterised by the accumulation of at least four mutations among those that confer cross-resistance to the PIs. Objective: We studied the evolution of PIs resistance mutations in 10 HAART-failed patients undergoing ATV enrolled in an early access program. Study design: Virus genotypic resistance was determined from plasma collected at baseline and during treatment. HIV-RNA was extracted and the pol region amplified and sequenced. Genotypic data were used to determine drug susceptibility. Phylogenetic analysis was performed. Results: At baseline, genotypic data showed cross-resistance patterns to approved PIs in 6 patients. In two of these subjects new mutations (I54V and A71V) conferring cross-resistance emerged after 3 months of therapy. The I50L mutation was evidenced in one subject after 12 months of treatment. The "virtual" phenotype analysis mirrored the resistance profiles to ATV and other PIs and evidenced differences with tipranavir and darunavir. Conclusion: Genotype evolution within the protease region did not emerge at significant levels during salvage therapy of multidrug-experienced patients. ATV exhibited certain/same virologic effect on the majority of our patients

    Multi-omics data integration provides insights into the post-harvest biology of a long shelf-life tomato landrace

    In this study we investigated the transcriptome and epigenome dynamics of the tomato fruit during post-harvest in a landrace belonging to a group of tomatoes (Solanum lycopersicum L.) collectively known as "Piennolo del Vesuvio", all characterized by a long shelf-life. Expression of protein-coding genes and microRNAs as well as DNA methylation patterns and histone modifications were analysed in distinct post-harvest phases. Multi-omics data integration contributed to the elucidation of the molecular mechanisms underlying processes leading to long shelf-life. We unveiled global changes in transcriptome and epigenome. DNA methylation increased and the repressive histone mark H3K27me3 was lost as the fruit progressed from red ripe to 150 days post-harvest. Thousands of genes were differentially expressed, about half of which were potentially epi-regulated as they were engaged in at least one epi-mark change in addition to being microRNA targets in ~5% of cases. Down-regulation of the ripening regulator MADS-RIN and of genes involved in ethylene response and cell wall degradation was consistent with the delayed fruit softening. Large-scale epigenome reprogramming that occurred in the fruit during post-harvest likely contributed to delayed fruit senescence

    Continuous vs Intermittent Meropenem Administration in Critically Ill Patients With Sepsis

    Importance: Meropenem is a widely prescribed ÎČ-lactam antibiotic. Meropenem exhibits maximum pharmacodynamic efficacy when given by continuous infusion to deliver constant drug levels above the minimal inhibitory concentration. Compared with intermittent administration, continuous administration of meropenem may improve clinical outcomes. Objective: To determine whether continuous administration of meropenem reduces a composite of mortality and emergence of pandrug-resistant or extensively drug-resistant bacteria compared with intermittent administration in critically ill patients with sepsis. Design, setting, and participants: A double-blind, randomized clinical trial enrolling critically ill patients with sepsis or septic shock who had been prescribed meropenem by their treating clinicians at 31 intensive care units of 26 hospitals in 4 countries (Croatia, Italy, Kazakhstan, and Russia). Patients were enrolled between June 5, 2018, and August 9, 2022, and the final 90-day follow-up was completed in November 2022. Interventions: Patients were randomized to receive an equal dose of the antibiotic meropenem by either continuous administration (n = 303) or intermittent administration (n = 304). Main outcomes and measures: The primary outcome was a composite of all-cause mortality and emergence of pandrug-resistant or extensively drug-resistant bacteria at day 28. There were 4 secondary outcomes, including days alive and free from antibiotics at day 28, days alive and free from the intensive care unit at day 28, and all-cause mortality at day 90. Seizures, allergic reactions, and mortality were recorded as adverse events. Results: All 607 patients (mean age, 64 [SD, 15] years; 203 were women [33%]) were included in the measurement of the 28-day primary outcome and completed the 90-day mortality follow-up. The majority (369 patients, 61%) had septic shock. The median time from hospital admission to randomization was 9 days (IQR, 3-17 days) and the median duration of meropenem therapy was 11 days (IQR, 6-17 days). Only 1 crossover event was recorded. The primary outcome occurred in 142 patients (47%) in the continuous administration group and in 149 patients (49%) in the intermittent administration group (relative risk, 0.96 [95% CI, 0.81-1.13], P = .60). Of the 4 secondary outcomes, none was statistically significant. No adverse events of seizures or allergic reactions related to the study drug were reported. At 90 days, mortality was 42% both in the continuous administration group (127 of 303 patients) and in the intermittent administration group (127 of 304 patients). Conclusions and relevance: In critically ill patients with sepsis, compared with intermittent administration, the continuous administration of meropenem did not improve the composite outcome of mortality and emergence of pandrug-resistant or extensively drug-resistant bacteria at day 28.
