    Deforming solutions of geometric variational problems with varying symmetry groups

    We prove an equivariant implicit function theorem for variational problems that are invariant under a varying symmetry group (corresponding to a bundle of Lie groups). Motivated by applications to families of geometric variational problems lacking regularity, several non-smooth extensions of the result are discussed. Among such applications is the submanifold problem of deforming the ambient metric preserving a given variational property of a prescribed family of submanifolds, e.g., constant mean curvature, up to the action of the corresponding ambient isometry groups.Comment: LaTeX2e, 26 pages, to appear in Transform.     An Assessment of the Psychometric Properties of Italian Version of CPGI

    The aim of this study was to adapt to the Italian context a very commonly used international instrument to detect problem gambling, the canadian problem gambling index (CPGI), and assess its psychometric properties. Cross-cultural adaptation of CPGI was performed in several steps and the questionnaire was administered as a survey among Italian general population (n = 5,292). Cronbach\u27s alpha reliability coefficient was 0.87 and can be considered to be highly reliable. Construct validity was assessed first by means of a principal component analysis and then by means of confirmatory factor analysis, showing that only one factor, problem gambling, was extracted from the CPGI questionnaire (an eigenvalues of 4,684 with percentage of variance 52 %). As far as convergent validity is concerned, CPGI was compared with Lie/Bet questionnaire, a two-item screening tool for detecting problem gamblers, and with both depression and stress scales. A short form DSM-IV CIDI questionnaire was used for depression and VRS scale, a rating scale, was used for rapid stress evaluation. A strong convergent validity with these instruments was found and these findings are consistent with past research on problem gambling, where another way to confirm the validity is to determine the extent to which it correlates with other qualities or measures known to be directly related to problem gambling. In sum, despite the lack of a direct comparison with a classic gold-standard such as DSM-IV, the Italian version of CPGI exhibits good psychometric properties and can be used among the Italian general population to identify at-risk problem gamblers.

    Sex and the single embryo: early deveopment in the Mediterranean fruit fly, Ceratitis capitata

    <p>Abstract</p> <p>Background</p> <p>In embryos the maternal-to-zygotic transition (MTZ) integrates post-transcriptional regulation of maternal transcripts with transcriptional activation of the zygotic genome. Although the molecular mechanisms underlying this event are being clarified in <it>Drosophila melanogaster</it>, little is know about the embryogenic processes in other insect species. The recent publication of expressed sequence tags (ESTs) from embryos of the global pest species <it>Ceratitis capitata </it>(medfly) has enabled the investigation of embryogenesis in this species and has allowed a comparison of the embryogenic processes in these two related dipteran species, <it>C. capitata </it>and <it>D. melanogaster</it>, that shared a common ancestor 80-100 mya.</p> <p>Results</p> <p>Using a novel PCR-based sexing method, which takes advantage of a putative LTR retrotransposon MITE insertion on the medfly Y chromosome, the transcriptomes of individual early male and female embryos were analysed using RT-PCR. This study is focused on two crucial aspects of the onset of embryonic development: sex determination and cellular blastoderm formation. Together with the three known medfly genes (<it>Cctransformer</it>, <it>Cctransformer2 </it>and <it>Ccdoublesex</it>), the expression patterns of other medfly genes that are similar to the <it>D. melanogaster </it>sex-determination genes (<it>sisterlessA, groucho, deadpan, Sex-lethal, female lethal d, sans fille </it>and <it>intersex</it>) and four cellular blastoderm formation genes (<it>Rho1, spaghetti squash, slow-as-molasses </it>and <it>serendipity-α</it>) were analyzed, allowing us to sketch a preliminary outline of the embryonic process in the medfly. Furthermore, a putative homologue of the <it>Zelda </it>gene has been considered, which in <it>D. melanogaster </it>encodes a DNA-binding factor responsible for the maternal-to-zygotic transition.</p> <p>Conclusions</p> <p>Our novel sexing method facilitates the study of i) when the MTZ transition occurs in males and females of <it>C. capitata</it>, ii) when and how the maternal information of "female-development" is reprogrammed in the embryos and iii) similarities and differences in the regulation of gene expression in <it>C. capitata </it>and <it>D. melanogaster</it>. We suggest a new model for the onset of the sex determination cascade in the medfly: the maternally inherited <it>Cctra </it>transcripts in the female embryos are insufficient to produce enough active protein to inhibit the male mode of <it>Cctra </it>splicing. The slow rate of development and the inefficiency of the splicing mechanism in the pre-cellular blastoderm facilitates the male-determining factor (M) activity, which probably acts by inhibiting CcTRA protein activity.</p

    European large-scale farmland investments and the land-water-energy-food nexus

    The escalating human demand for food, water, energy, fibres and minerals have resulted in increasing commercial pressures on land and water resources, which are partly reflected by the recent increase in transnational land investments. Studies have shown that many of the land-water issues associated with land acquisitions are directly related to the areas of energy and food production. This paper explores the land-water-energy-food nexus in relation to large-scale farmland investments pursued by investors from European countries. The analysis is based on a “resource assessment approach” which evaluates the linkages between land acquisitions for agricultural (including both energy and food production) and forestry purposes, and the availability of land and water in the target countries. To that end, the water appropriated by agricultural and forestry productions is quantitatively assessed and its impact on water resource availability is analysed. The analysis is meant to provide useful information to investors from EU countries and policy makers on aspects of resource acquisition, scarcity, and access to promote responsible land investments in the target countries

    New endoscopic capsule vs upper gastrointestinal endoscopy in preoperative work-up of obese candidate for bariatric surgery: Relevance of a pilot study in the COVID-19 era

    : Background and study aims  Obesity represents a major health concern; bariatric surgery is the most effective treatment reducing and maintaining weight loss. The role of a routine esophagogastroduodenoscopy (EGD) prior bariatric surgery is still debated. Moreover, in this scenario of COVID-19 pandemic, EGD is even more questionable due to the procedural risk of viral transmission. A new model of video-endoscopic capsule (VEC) recently has been introduced as a good alternative to the EGD. The aim of this study was to determine if this new capsule is an adequate diagnostic alternative to EGD in the work-up of patients selected for bariatric surgery, particularly in the setting of COVID-19. Patients and methods  From January to November 2020, 27 patients selected for bariatric surgery were enrolled in this pilot study to assess for noninferiority of VEC compared to EGD in detection of upper gastrointestinal disease. Results  VEC had sensitivity, specificity, and positive and negative predictive values in identification of significant lesions of 91.3 %, 83.33 %, 98.01 %, and 51.57 %, respectively, compared with EGD as the standard criterion. The accuracy was 90.51 % (95 % CI, 73.75 %-98.18 %) and the chi-square statistic is 0.1153 ( P  = 0.73). Conclusions  Our report confirms the diagnostic noninferiority of VEC in preoperative work-up of patients selected for bariatric surgery, compared to EGD. This is very important, particularly during the COVID-19 pandemic, given the high risk of contamination with EGD. Larger multicenter studies are required to confirm our preliminary results

    Antifungal and Antibiofilm Activity of Cyclic Temporin L Peptide Analogues against Albicans and Non-Albicans Candida Species

    Temporins are one of the largest families of antimicrobial peptides with both anti-inflammatory and antimicrobial activity. Herein, for a panel of cyclic temporin L isoform analogues, the antifungal and antibiofilm activities were determined against representative Candida strains, including C. albicans, C. glabrata, C. auris, C. parapsilosis and C. tropicalis. The outcomes indicated a significant anti-candida activity against planktonic and biofilm growth for four peptides (3, 7, 15 and 16). The absence of toxicity up to high concentrations and survival after infection were assessed in vivo by using Galleria mellonella larvae, and the correlation between conformation and cytotoxicity was investigated by fluorescence assays and circular dichroism (CD). By combining fluorescence spectroscopy, CD, dynamic light scattering, confocal and atomic force microscopy, the mode of action of four analogues was hypothesized. The results pinpointed that peptide 3 emerged as a non-toxic compound showing a potent antibiofilm activity and represents a promising compound for biomedical applications
