    Assessing The Health Of Fringing Salt Marshes Along The Fore River And Its Tributaries

    Casco Bay has experienced two significant oil spills in recent history - the Tamano Tanker spill in 1972 and the Julie N spill in 1996. In addition, the Fore River and its tributaries have experienced numerous small spills over the years, including the August 2002 fuel oil spill (2,900 gallons) and the April 2003 jet fuel spill (6,000 gallons). The impacts of these spills on the fringing salt marshes that line the edges of the Fore River and its tributaries are not well understood. Are these salt marshes resilient enough to withstand these impacts and still act as healthy marshes should? Or are they functioning at lower levels, compared to other fringing salt marshes in Casco Bay? The salt marshes that line the Fore River provide an important suite of functions and values to the citizens of southern Maine. The results of this study will help resource managers in their efforts to protect these important and unique natural resources

    The Functions And Values Of Fringing Salt Marshes In Northern New England, USA

    Although large salt marshes of the northeastern United States have been studied extensively, very little is known about the smaller, fringing marshes in this area, despite the fact that they are a common habitat type. We compared the functions and values of five fringing salt marshes (FM) to those of five meadow marshes (MM) along the southern Maine/New Hampshire coast. Specifically we compared their primary production, soil organic matter content, plant diversity, sediment trapping ability and wave dampening properties. Also explored were the relationships between these functions and several physical characteristics at each site, including soil salinity, percent surface slope, elevation and size. The results of this study indicate that despite their small size, fringing salt marshes are valuable components of estuaries, performing many ecological functions to the same degree as nearby meadow marshes. More effort should be made to include them in regional efforts to conserve and restore coastal habitats

    Casco Bay is the largest oil port in Maine and northern New England, handling over 20 million tons of crude oil and oil products annually. The susceptibility of the Bay’s estuarine habitats, especially its fringing salt marshes, to potential spill events was the impetus for this study. Although much has been learned to date about the effects of oil spills on estuarine habitats around the world, there is a real need for site-specific knowledge of the structures and functions of local habitats so that resource managers can be prepared in the event of a spill. Our study focused specifically on the value of Casco Bay’s fringing salt marshes to shellfish and finfish production, to vegetation production and diversity, and as buffers against sea level rise and coastal erosion. The work we have accomplished has been the first study of the fringing salt marshes of Casco Bay that has explored the biotic communities (fish, invertebrates and plants) of these marshes in conjunction with the physical properties of these sites. Knowledge of these local fringing salt marsh habitats will be invaluable in improving the effectiveness of oil spill cleanup operations, accurate assessment of natural resource damages caused by spills, and the restoration of impacted sites. In addition, the data acquired in this study provide an initial set of benchmarks upon which to build a program to assess long-term change in Casco Bay tidal marsh habitats

In particular, the project focused on potential impacts to the marshes due to shoreline development pressures.     Enterohemorrhagic Escherichia coli (EHEC) is dependent on acid resistance for gastric passage and low oral infectious dose, and the locus of enterocyte effacement (LEE) for intestinal colonization. Mutation of rpoN, encoding sigma factor N (σ(N)), dramatically alters the growth-phase dependent regulation of both acid resistance and the LEE. This study reports on the determinants of σ(N)-directed acid resistance and LEE expression, and the underlying mechanism attributable to this phenotype. Glutamate-dependent acid resistance (GDAR) in TW14359ΔrpoN correlated with increased expression of the gadX-gadW regulatory circuit during exponential growth, whereas upregulation of arginine-dependent acid resistance (ADAR) genes adiA and adiC in TW14359ΔrpoN did not confer acid resistance by the ADAR mechanism. LEE regulatory (ler), structural (espA and cesT) and effector (tir) genes were downregulated in TW14359ΔrpoN, and mutation of rpoS encoding sigma factor 38 (σ(S)) in TW14359ΔrpoN restored acid resistance and LEE genes to WT levels. Stability, but not the absolute level, of σ(S) was increased in TW14359ΔrpoN; however, increased stability was not solely attributable to the GDAR and LEE expression phenotype. Complementation of TW14359ΔrpoN with a σ(N) allele that binds RNA polymerase (RNAP) but not DNA, did not restore WT levels of σ(S) stability, gadE, ler or GDAR, indicating a dependence on transcription from a σ(N) promoter(s) and not RNAP competition for the phenotype. Among a library of σ(N) enhancer binding protein mutants, only TW14359ΔntrC, inactivated for nitrogen regulatory protein NtrC, phenocopied TW14359ΔrpoN for σ(S) stability, GDAR and ler expression. The results of this study suggest that during exponential growth, NtrC-σ(N) regulate GDAR and LEE expression through downregulation of σ(S) at the post-translational level; likely by altering σ(S) stability or activity. The regulatory interplay between NtrC, other EBPs, and σ(N)-σ(S), represents a mechanism by which EHEC can coordinate GDAR, LEE expression and other cellular functions, with nitrogen availability and physiologic stimuli.Microbiology and Molecular Genetic

    A mobile phone text messaging intervention to manage fatigue for people with multiple sclerosis, spinal cord injury, and stroke: Development and usability testing

    BACKGROUND: Fatigue significantly affects daily functioning in persons with disabilities. Fatigue management can be challenging, and the information provided during routine physician visits to manage fatigue can be overwhelming. One way to address fatigue is to increase knowledge, skills, and confidence for self-management (ie, patient activation). Self-management programs have shown promising effects in targeting fatigue in persons with disabilities. However, satisfaction with self-management programs is low for persons with disabilities, and tailoring interventions to personalized needs has been recommended. SMS text messaging is increasingly being used to implement health behavior change interventions in a person\u27s natural environment. Little has been done to link mobile health approaches with patient activation and self-management to address fatigue in persons with disabilities. OBJECTIVE: This study aimed to develop and test a mobile phone-based fatigue self-management SMS text messaging intervention targeting patient activation in 3 groups of persons with disabilities: persons with multiple sclerosis, persons who had a stroke, and persons with a spinal cord injury. METHODS: We used evidence-based resources and input from a consumer advisory board (CAB; composed of 2 participants from each of the 3 disability groups) and a neurologist to develop the intervention. The study was conducted using a 4-step process: development of the initial SMS text messaging library and categorization of the content into 9 content areas, review and modification of the SMS text messages by the neurologist and CAB, integration of the content library into a digital platform, and utility testing by CAB members. RESULTS: A total of 6 CAB participants rated SMS text messages covering 9 domain areas of fatigue self-management with good clarity (mean ratings=3.5-5.0 out of 5) and relevance (mean ratings=3.2-5.0 out of 5). Overall, SMS text messaging content was reported by CAB participants as helpful, clear, and well suited for a mobile health intervention. The CAB reached consensus on the time of day that SMS text messages should be sent (morning) and their frequency (once per day). This feedback led the research team to narrow down the program to deliver 48 SMS text messages, 1 per day, Monday through Thursday only, a total of 4 SMS text messages per week, over a 12-week period. The final set of SMS text messages was programmed into a digital platform with a predefined delivery schedule. The usability of the intervention was high, with 55 (83%) out of 66 responses endorsing the highest rating. CONCLUSIONS: This study demonstrates a step-by-step process for developing a fatigue self-management SMS text messaging intervention for persons with disabilities. For this population, whose access to health services is often limited, this intervention provides an alternative delivery model to increase access to fatigue information and deliver content that aligns with the person\u27s needs

    A Fire Severity Mapping System for Real-Time Fire Management Applications and Long-Term Planning: The FIRESEV project

    Accurate, consistent, and timely fire severity maps are needed in all phases of fire management including planning, managing, and rehabilitating wildfires. The problem is that fire severity maps are commonly developed from satellite imagery that is difficult to use for planning wildfire responses before a fire has actually happened and can’t be used for real-time wildfire management because of the timing of the imagery delivery. Moreover, imagery is difficult to use for controlled fires such as prescribed burning. This study, called FIRESEV (FIRE SEVerity Mapping Tools) created a comprehensive set of tools and protocols to deliver, create, and evaluate fire severity maps for all phases of fire management. The first tool is a Severe Fire Potential Map (SFPM) that quantifies the potential for fires to burn with high severity, should they occur, for any 30m x 30m piece of ground across the western United States. This map was developed using empirical models that related topographic, vegetation, and fire weather variables to burn severity as mapped using the Monitoring Trends in Burn Severity (MTBS) digital products. This SFPM map is currently available on the Fire Research and Management Exchange System (FRAMES, http://www.frames.gov/firesev) web site and can be used to plan for future wildfires or for managing wildfires in real time, e.g. by including it as a layer in Wildland Fire Decision Support System or other GIS analysis. The next tool was the inclusion of a fire severity mapping algorithm in the Wildland Fire Assessment Tool (WFAT) developed by the National Interagency Fuels Technology Transfer (NIFTT) team. WFAT is used for fuel treatment planning to predict potential fire effects under prescribed fire weather conditions (http://www.frames.gov/partner-sites/niftt/tools/niftt-current-resources/). Now, fire severity can be mapped explicitly from fire effects simulation models (FOFEM, Consume) for real-time and planning wildfire applications. Next, the FIRESEV project showed how results from the WFAT simulated fire severity can be integrated with satellite imagery to improve fire severity mapping. And last, the FIRESEV project produced a suite of research studies, synthesis papers, and popular articles designed to improve the description, interpretation, and mapping of fire severity for wildland fire management: (1) a research study created a completely objective method of quantifying fire severity from fire effects to obtain nine unique classes of fire severity, (2) a research study comprehensively contrasted all current classifications of fire severity using Composite Burn Index (CBI) as measured on over 300 plots across the western United States to determine commonalities and differences, and (3) a synthesis paper was written discussing the problems involved in measuring, describing, and quantifying fire severity. This FIRESEV project yielded over 15 deliverables that we feel provides a comprehensive suite of products to create useful fire severity maps, along with current satellite imagery products, and also FIRESEV provides a thorough background on how to measure, interpret, and apply fire severity in fire management

    AbstractMany herpesviruses, including Marek's disease viruses (MDV1 and MDV2), encode microRNAs. In this study, we report microRNAs of two related herpesviruses, infectious laryngotracheitis virus (ILTV) and herpesvirus of turkeys (HVT), as well as additional MDV2 microRNAs. The genome locations, but not microRNA sequences, are conserved among all four of these avian herpesviruses. Most are clustered in the repeats flanking the unique long region (I/TRL), except in ILTV which lacks these repeats. Two abundant ILTV microRNAs are antisense to the immediate early gene ICP4. A homologue of host microRNA, gga-miR-221, was found among the HVT microRNAs. Additionally, a cluster of HVT microRNAs was found in a region containing two locally duplicated segments, resulting in paralogous HVT microRNAs with 96–100% identity. The prevalence of microRNAs in the genomic repeat regions as well as in local repeats suggests the importance of genetic plasticity in herpesviruses for microRNA evolution and preservation of function
