    Hydrophobic DES Based on Menthol and Natural Organic Acids for Use in Antifouling Marine Coatings

Published by American Chemical Society.Marine biofouling negatively impacts industries with off-shore infrastructures, such as naval, oil, and aquaculture. To date, there are no ideal sustainable, economic, and environmentally benign solutions to deal with this phenomenon. The advances achieved in green solvents, as well as its application in different industries, such as pharmaceutical and biotechnology, have promoted the emergence of deep eutectic systems (DES). These eutectic systems have applications in various fields and can be revolutionary in the marine-based industrial sector. In this study, the main objective was to investigate the potential use of hydrophobic DES (HDES) based on menthol and natural organic acids for their use as marine antifouling coatings. Our strategy encompassed the physicochemical characterization of different formulations, which allowed us to identify the most appropriate molar ratio and intermolecular interactions for HDES formations. The miscibility of the resulting HDES with the marine coating has been evaluated and proven to be successful. The Men/OL (1:1) system proved to be the most promising in terms of cost-production and thus was the one used in subsequent antifouling tests. The cytotoxicity of this HDES was evaluated using an in vitro cell model (HaCat cells) showing no significant toxicity. Furthermore, the application of this system incorporated into coatings that are used in marine structures was also studied using marine species (Mytilus edulis mussels and Patella vulgata limpets) to evaluate both their antifouling and ecotoxicity effects. HDES Men/OL (1:1) incorporated in marine coatings was promising in reducing marine macrofouling and also proved to be effective at the level of microfouling without viability impairment of the tested marine species. It was revealed to be more efficient than using copper oxide, metallic copper, or ivermectin as antifouling agents. Biochemical assays performed on marine species showed that this HDES does not induce oxidative stress in the tested species. These results are a strong indication of the potential of this HDES to be sustainable and efficiently used in marine fouling control technologies.publishersversionpublishe

    Highly regional population structure of Spondyliosoma cantharus depicted by nuclear and mitochondrial DNA data

    Resolution of population structure represents an effective way to define biological stocks and inform efficient fisheries management. In the present study, the phylogeography of the protogynous sparid Spondyliosoma cantharus, in the East Atlantic and Mediterranean Sea, was investigated with nuclear (S7) and mitochondrial (cytochrome b) DNA markers. Significant divergence of four regional genetic groups was observed: North Eastern Atlantic, Mediterranean Sea, Western African Transition (Cape Verde) and Gulf of Guinea (Angola). The two southern populations (Cape Verde and Angola) each comprised reciprocally monophyletic mtDNA lineages, revealed low levels of diversity in Cape Verde and high diversity for Angola despite being represented by only 14 individuals. A complete divergence between North Atlantic and Mediterranean populations was depicted by the mitochondrial marker, but a highly shared nuclear haplotype revealed an incomplete lineage sorting between these regions. Bayesian skyline plots and associated statistics revealed different dynamics among the four regions. Cape Verde showed no expansion and the expansion time estimated for Angola was much older than for the other regions. Mediterranean region seems to have experienced an early population growth but has remained with a stable population size for the last 30000 years while the North Atlantic population has been steadily growing. The lack of genetic structuring within these regions should not be taken as evidence of demographic panmixia in light of potential resolution thresholds and previous evidence of intra-regional phenotypic heterogeneity.info:eu-repo/semantics/publishedVersio

    Antioxidant activity of macroalgae from the Azores

    Free radical scavenging activity (FRSA) was studied to determine the antioxidant activity of 8 common macroalgae species found in the Azores. Species under study were Porphyra sp., Osmundea pinnatifida, Pterocladiella capillacea, Sphaerococcus coronopifolius and Gelidium microdon, from Rhodophyta; Ulva compressa and Chaetomorpha pachynema, from Chlorophyta and Fucus spiralis from Phaeophyta.The antioxidant activity was evaluated in methanolic extract by a decolourisation solution test of 2,2-diphenyl-1-picrylhydrazyl (DPPH), used as a stable radical. The methanolic extracts were obtained from dried biomass by sequential extractions, attaining a final concentration of 2 mg.mL-1. The FRSA values ranged from 19.54% for S. coronopifolius to 60.05% for F. spiralis with standard deviation (SD) varying between 1.41% and 6.80%. Results indicated that the studied seaweeds are a very promising source of biological active compounds with antioxidante properties. The seaweeds were collected in the Azorean islands where seawater pollution levels are low. Consequently, these seaweeds represent a valuable and good source of antioxidante material with superior beneficial effects on human health

    A comparison between pure active pharmaceutical ingredients and therapeutic deep eutectic solvents: solubility and permeability studies

    THEDES, so called therapeutic deep eutectic solvents are here defined as a mixture of two components, which at a particular molar composition become liquid at room temperature and in which one of them is an active pharmaceutical ingredient (API). In this work, THEDES based on menthol complexed with three different APIs, ibuprofen (ibu), BA (BA) and phenylacetic acid (PA), were prepared. The interactions between the components that constitute the THEDES were studied by NMR, confirming that the eutectic system is formed by H-bonds between menthol and the API. The mobility of the THEDES components was studied by PFGSE NMR spectroscopy. It was determined that the self-diffusion of the species followed the same behavior as observed previously for ionic liquids, in which the components migrate via jumping between voids in the suprastructure created by punctual thermal fluctuations. The solubility and permeability of the systems in an isotonic solution was evaluated and a comparison with the pure APIs was established through diffusion and permeability studies carried out in a Franz cell. The solubility of the APIs when in the THEDES system can be improved up to 12 fold, namely for the system containing ibu. Furthermore, for this system the permeability was calculated to be 14 105 cm/s representing a 3 fold increase in comparison with the pure API. With the exception of the systems containing PA an increase in the solubility, coupled with an increase in permeability was observed.     Nutritional value of selected macroalgae

Their protein and fiber content have been widely studied and differ according to the species, their geographic origin and their seasonal conditions. In addition to their value for human nutrition, seaweeds have multiple therapeutically applications (e.g., weight control, hypocholesterolemic, antioxidant and antitumor activities, others) and, in general, contribute and promote human health. In the archipelago of the Azores, the consumption of seaweeds is widespread and accepted as a common practice in some islands. This work is aimed at providing information on the protein and fiber content of the locally consumed species, to promote this regional food product that can be potentially profitable from the biotechnology and commercial perspective, and also benefit public health, particularly, taking into account the low level of marine pollution in the Azores archipelago. Protein and fiber content of eight seaweeds (Porphyra sp., Osmundea pinnatifida, Pterocladiella capillacea, Sphaerococcus coronopifolius, and Gelidium microdon, Rhodophyta; Cystoseira abies-marina and Fucus spiralis, Phaeophyta; Ulva compressa, Chlorophyta) were determined using the Kjeldahl method and the Weende method, respectively. The protein content ranged from 6.81 to 26.62 of dry weight for C. abies-marina and U. compressa, respectively. Fiber content was generally higher as compared with that in seaweeds from other origins and ranged from 33.82 to 63.88 for O. pinnatifida and F. spiralis, respectively

As novas moléculas têm grande interesse científico, económico, social e farmacológico podendo contribuir, em muito, para a saúde das pessoas e para a sua qualidade de vida. O rastreio de novos compostos com atividade biológica tem sido, recentemente, uma das prioridades da comunidade científica e os Açores, pelas suas características particulares e endemismos assumem particular importância nesta área de investigação. O estudo das potencialidades de plantas e organismos marinhos dos Açores tem sido desenvolvido na Universidade dos Açores/CIRN desde a década de noventa, usando a estratégia de investigação seguinte: (i) seleção dos organismos mais promissores para fins industriais e comerciais pela avaliação da sua atividade em ensaios biológicos diversos, (ii) extração, separação e identificação dos compostos bioativos e (iii) estudos da relação estrutura-atividade dos compostos bioativos isolados.ABSTRACT: The chemistry of Natural Products has contributed greatly to the discovery of pharmaceuticals and agrochemicals agents, among others, by the isolation of active substances produced by various organisms. The new molecules are of great scientific, economic, social and pharmacological importance and may contribute greatly to the people's health and their quality of life. The screening for new compounds with biological activity has been a recent priority for the scientific community and the Azores, by its particular characteristics and endemism is of crucial importance in this area of research. The study of the Azorean plants and marine organisms potential has been investigated in Azores University/CIRN since the '90s decade, using the following research strategy: (i) screening of the most promising organisms for industrial and commercial purposes by evaluating their activity in several biological assays, (ii) extraction, purification and molecular structure elucidation of the bioactive compounds, and (iii) studies of structure-activity relationships of the isolated bioactive compounds
