Extension of the GATE Monte-Carlo simulation package to model bioluminescence and fluorescence imaging
International audienceThe Geant4 Application for Emission Tomography (GATE) is an advanced open-source software dedicated to Monte-Carlo (MC) simulations in medical imaging involving photon transportation (Positron emission tomography, single photon emission computed tomography, computed tomography) and in particle therapy. In this work, we extend the GATE to support simulations of optical imaging, such as bioluminescence or fluo-rescence imaging, and validate it against the MC for multilayered media standard simulation tool for biomedical optics in simple geometries. A full simulation setup for molecular optical imaging (bioluminescence and fluo-rescence) is implemented in GATE, and images of the light distribution emitted from a phantom demonstrate the relevance of using GATE for optical imaging simulations
Multispectral reflectance imaging of brain activation in rodents: methodological study of the differential path length estimations and first in vivo recordings in the rat olfactory bulb
International audienceDynamic maps of relative changes in blood volume and oxygenation following brain activation are obtained using multispectral reflectance imaging. The technique relies on optical absorption modifications linked to hemodynamic changes. The relative variation of hemodynamic parameters can be quantified using the modified Beer-Lambert Law if changes in reflected light intensities are recorded at two wavelengths or more and the differential path length (DP) is known. The DP is the mean path length in tissues of backscattered photons and varies with wavelength. It is usually estimated using Monte Carlo simulations in simplified semi-infinite homogeneous geometries. Here we consider the use of multilayered models of the somatosensory cortex (SsC) and olfactory bulb (OB), which are common physiological models of brain activation. Simulations demonstrate that specific DP estimation is required for SsC and OB, specifically for wavelengths above 600 nm. They validate the hypothesis of a constant path length during activation and show the need for specific DP if imaging is performed in a thinned-skull preparation. The first multispectral reflectance imaging data recorded in vivo during OB activation are presented, and the influence of DP on the hemodynamic parameters and the pattern of oxymetric changes in the activated OB are discusse
Experimental and analytical comparative study of optical coefficient of fresh and frozen rat tissues
International audienceOptical properties of fresh and frozen tissues of rat heart, kidney, brain, liver, and muscle were measured in the 450-to 700-nm range. The total reflectance and transmittance were measured using a well-calibrated integral sphere setup. Absorption coefficient μ a and reduced scattering coefficient μ 0 s were derived from the experimental measurements using the inverse adding doubling technique. The influence of cryogenic processing on optical properties was studied. Interindividual and intraindividual variations were assessed. These new data aim at filling the lack of validated optical properties in the visible range especially in the blue-green region of particular interest for fluorescence and optogenetics preclinical studies. Furthermore, we provide a unique comparison of the optical properties of different organs obtained using the same measurement setup for fresh and frozen tissues as well as an estimate of the intraindividual and interindividual variability
Experimental and analytical comparative study of optical coefficient of fresh and frozen rat tissues
Search for composite and exotic fermions at LEP 2
A search for unstable heavy fermions with the DELPHI detector at LEP is
reported. Sequential and non-canonical leptons, as well as excited leptons and
quarks, are considered. The data analysed correspond to an integrated
luminosity of about 48 pb^{-1} at an e^+e^- centre-of-mass energy of 183 GeV
and about 20 pb^{-1} equally shared between the centre-of-mass energies of 172
GeV and 161 GeV. The search for pair-produced new leptons establishes 95%
confidence level mass limits in the region between 70 GeV/c^2 and 90 GeV/c^2,
depending on the channel. The search for singly produced excited leptons and
quarks establishes upper limits on the ratio of the coupling of the excited
fermio
Search for lightest neutralino and stau pair production in light gravitino scenarios with stau NLSP
Promptly decaying lightest neutralinos and long-lived staus are searched for
in the context of light gravitino scenarios. It is assumed that the stau is the
next to lightest supersymmetric particle (NLSP) and that the lightest
neutralino is the next to NLSP (NNLSP). Data collected with the Delphi detector
at centre-of-mass energies from 161 to 183 \GeV are analysed. No evidence of
the production of these particles is found. Hence, lower mass limits for both
kinds of particles are set at 95% C.L.. The mass of gaugino-like neutralinos is
found to be greater than 71.5 GeV/c^2. In the search for long-lived stau,
masses less than 70.0 to 77.5 \GeVcc are excluded for gravitino masses from 10
to 150 \eVcc . Combining this search with the searches for stable heavy leptons
and Minimal Supersymmetric Standard Model staus a lower limit of 68.5 \GeVcc
may be set for the stau mas
Instrumentation et méthodologies pour les études cérébrales in vivo sur modèles animaux : mesure cinétiques de radiotraceurs et imageries optiques en champ large.
Résumé : Les recherches que j’ai menées se sont successivement organisées selon deux thématiques avec comme fil directeur l’interface entre l’instrumentation et les neurosciences. Dans une première phase j’ai participé au développement et à la mise en œuvre d’une sonde radiosensible béta pour les études pharmacocinétiques in vivo à l’aide de traceurs radio marqués. La sonde intracérébrale (SIC) est née de la collaboration de radio-pharmaciens et neurobiologistes des centres de Tomographie par Emission de Positrons (TEP) de Lyon et d’Orsay et d’instrumentalistes du groupe Interface Physique Biologie de l’institut de Physique Nucléaire d’Orsay. Les atouts majeurs de SIC dans son positionnement comme alternative aux micro-tomographes, sont : (i) la résolution temporelle, (ii) la facilité de couplage avec d’autres modalités d’imagerie comme l’IRM et (iii) la souplesse d’utilisation et d’intégration de l’instrument pour les laboratoires de Neurosciences. Après avoir établit la faisabilité et les caractéristiques physiques du dispositif par simulations Monte Carlo, j’ai développé ce détecteur durant ma thèse et jusqu’en 2005. Pour son application en Neurobiologie, deux thématiques de recherche ont été poursuivies dans le cadre de collaborations: (i) les études pharmacocinétique des traceurs du système dopaminergiques et sérotoninergiques et plus particulièrement les études de marquage compétitif ; (ii) l’étude du métabolisme du glucose lors d’une activation cérébrale ou sur un modèle de pathologie. Parallèlement à ces travaux, j’ai participé à plusieurs avancées méthodologiques autour de la sonde : son association avec l’imagerie IRM, son utilisation pour la mesure cinétique de la radioactivité dans la circulation sanguine. Avec la création en 2006 du laboratoire interdisciplinaire « Imagerie et Modélisation en Neurobiologie et Cancérologie » avec en son sein un groupe de neurobiologistes, j’ai réorienté mes recherches vers des travaux d’interface plus intégrés avec pour finalité l’étude du métabolisme énergétique cérébral. Du point de vue instrumental, le constat de leur excellente résolution spatio-temporelle pour les études in vivo de l’activation cérébrale à l’échelle des structures fonctionnelles m’a conduit à privilégier les approches optiques. Nous avons développé successivement l’imagerie de réflectance, l’imagerie d’autofluorescence et l’imagerie du contraste laser speckle qui permettent la cartographie en deux dimensions de paramètres métaboliques complémentaires. Instrumentalement, ces techniques reposent sur la synchronisation d’un microscope en champ large, d’une ou plusieurs caméras CCD, d’une source d’illumination à balayage spectral rapide, d’une diode laser modulée, des filtres interférentiels associés et du dispositif de stimulation sensorielle. Le travail instrumental et méthodologique a consisté à construire les dispositifs d’imagerie mais également à comprendre et quantifier par simulation Monte Carlo les effets de la diffusion et de l’absorption de la lumière dans les tissus sur la formation des cartes d’activation. Les trois modalités développées successivement au cours de deux thèses menées entre 2006 et 2012 ont permis d’obtenir chez le rongeur anesthésié des données originales lors d’une stimulation sensorielle du bulbe olfactif chez le rongeur. Je conclurai en présentant les perspectives de ces travaux en particulier, la combinaison des techniques de cartographie en champ large et des techniques d’imagerie optique à la résolution cellulaire
