    Longitudinal study of the effects of teat condition on the risk of new intramammary infections in dairy cows

    Machine milking–induced alterations of teat tissue may impair local defense mechanisms and increase the risk of new intramammary infections. The objective of the current study was to assess the influence of short-term and long-term alterations of teat tissue and infectious status of the udder quarter on the risk of naturally occurring new intramammary infections, inflammatory responses, and mastitis. Short-term and long-term changes in teat condition of right udder quarters of 135 cows of a commercial dairy farm in Saxony-Anhalt, Germany, were recorded monthly for 10 mo using simple classification schemes. Quarter milk samples were collected from all examined quarters at each farm visit. Bacteriological culture results and somatic cell counts of quarter milk samples were used to determine new inflammatory responses (increase from ≤100,000 cells/mL to >100,000 cells/mL between 2 samples), new infections (detection of a pathogen from a quarter that was free of the same pathogen at the preceding sampling), and new mastitis (combination of new inflammatory response and new infection). Separate Poisson mixed models for new inflammatory responses, new infections, and new mastitis caused by specific pathogens or groups of pathogens (contagious, environmental, major, minor, or any) were used to estimate risk ratios and 95% confidence intervals. Data preparation and parameter estimation were performed using the open source statistical analysis software R. We observed no effect of any variable describing teat condition on the risk of new intramammary infections, inflammatory responses, or mastitis. Intramammary infections of the same udder quarter in the preceding month did not affect risk either

    Architecture of the fungal nuclear pore inner ring complex

    The nuclear pore complex (NPC) constitutes the sole gateway for bidirectional nucleocytoplasmic transport. We present the reconstitution and interdisciplinary analyses of the ~425-kDa inner ring complex (IRC), which forms the central transport channel and diffusion barrier of the NPC, revealing its interaction network and equimolar stoichiometry. The Nsp1•Nup49•Nup57 channel nucleoporin hetero-trimer (CNT) attaches to the IRC solely through the adaptor nucleoporin Nic96. The CNT•Nic96 structure reveals that Nic96 functions as an assembly sensor that recognizes the three dimensional architecture of the CNT, thereby mediating the incorporation of a defined CNT state into the NPC. We propose that the IRC adopts a relatively rigid scaffold that recruits the CNT to primarily form the diffusion barrier of the NPC, rather than enabling channel dilation

    Gesundheit und Leistungsfähigkeit von Milchkühen im ökologischen Landbau interdisziplinär betrachtet – eine (Interventions-) Studie zu Stoffwechselstörungen und Eutererkrankungen unter Berücksichtigung von Grundfuttererzeugung, Fütterungsmanagement und Tierhaltung

    Im Mittelpunkt des Projektes stand die Stoffwechsel- und Eutergesundheit von ökologisch gehaltenen Milchkühen im prä- und peripartalen Zeitraum sowie in den ersten 100 Laktationstagen und deren Beeinflussung durch die Futter- und Nährstoffversorgung und die Haltungsumwelt im umfassenden Sinn. In einer bundesweiten Feldstudie auf 106 ökologisch wirtschaftenden Milchviehbetrieben erfolgten Erhebungen mit dem Ziel einer Risikomodellierung zu Stoffwechsel- und Eutererkrankungen. Vor diesem Hintergrund wurden die Produktionssysteme von der Pflanzenzusammensetzung im Grünland und im Ackerfutter über die Grobfutterproduktion, Futterqualität und Rationsgestaltung, Haltungsumwelt bis hin zur Tiergesundheit und Milchqualität analysiert, um hier einzelbetriebliche Risikoeinschätzungen vorzunehmen, Optimierungspotenziale aufzuzeigen und Handlungsempfehlungen abzuleiten, die anschließend betriebsindividuell implementiert wurden. Die Effektivität des so geschaffenen präventiv orientierten Tiergesundheitsmanagements wurde anhand der Entwicklung ausgewählter Kennzahlen der Euter- und Stoffwechselgesundheit geprüft. Es konnte gezeigt werden, dass sich mit dieser Vorgehensweise auch unter Praxisbedingungen die Tiergesundheitssituation signifikant verbes-sern lässt. Die Feldstudie wurde mit experimentellen Untersuchungen ergänzt, die sich speziellen Fragen der Analyse von nXP in Grasprodukten, des Kraftfuttereinsatzes, der Wahl der geeigneten Rasse, dem Infektionsgeschehen, der Nutzung von Haltungstechniken im Fütterungsmanagement und der Verbesserung der Grasnarbe widmeten. Die im Projekt generierten, aufgrund ihrer Ableitung aus der Praxis widerspruchsarmen Erkenntnisse wurden über vielfältige Formen des Wissenstransfers an die Akteure in der Ökologischen Milchviehhaltung vermittelt. Ein Merkblatt zur Euter- und Stoffwechselgesundheit bei Biomilchkühen und ein modular aufgebautes Wissenstransferkonzept wurden erarbeitet, um die Projektergebnisse nachhaltig nutzen zu können

    Nitric Oxide (NO) and Cyclooxygenase-2 (COX-2) Cross-Talk in Co-Cultures of Tumor Spheroids with Normal Cells

    Cyclooxygenases (COX), prostaglandin E2 (PGE2) and nitric oxide (NO) are believed to be some of the most important factors related to colon cancer growth and metastasis. In this study, we aimed to investigate the associations between COX-2, PGE2 and NO in co-cultures of human colon cancer spheroids obtained from different tumor grades with normal human colonic epithelium and myofibroblast monolayers. L-arginine (2 mM), a substrate for nitric oxide synthases (NOS), decreased COX-2 and PGE2 levels, while NG-nitro-L-arginine methyl ester (L-NAME) (2 mM), a NOS inhibitor, had no influence on COX-2 and PGE2 levels but limited tumor cell motility. NS398 (75 μM), a selective COX-2 inhibitor, had no significant influence on NO level but decreased motility of tumor cells. COX-2, PGE2 and NO levels depended on the tumor grade of the cells, being the highest in Duke’s stage III colon carcinoma. Summing up, we showed that addition of L-arginine at doses which did not stimulate NO level caused a significant decrease in COX-2 and PGE2 amounts in co-cultures of colon tumor spheroids with normal epithelial cells and myofibroblasts. Any imbalances in NO level caused by exogenous factors influence COX-2 and PGE2 amounts depending on the kind of cells, their reciprocal interactions and the local microenvironmental conditions. The knowledge of these effects may be useful in limiting colon carcinoma progression and invasion

    Extensive recombination events and horizontal gene transfer shaped the Legionella pneumophila genomes

    <p>Abstract</p> <p>Background</p> <p><it>Legionella pneumophila </it>is an intracellular pathogen of environmental protozoa. When humans inhale contaminated aerosols this bacterium may cause a severe pneumonia called Legionnaires' disease. Despite the abundance of dozens of <it>Legionella </it>species in aquatic reservoirs, the vast majority of human disease is caused by a single serogroup (Sg) of a single species, namely <it>L. pneumophila </it>Sg1. To get further insights into genome dynamics and evolution of Sg1 strains, we sequenced strains Lorraine and HL 0604 1035 (Sg1) and compared them to the available sequences of Sg1 strains Paris, Lens, Corby and Philadelphia, resulting in a comprehensive multigenome analysis.</p> <p>Results</p> <p>We show that <it>L. pneumophila </it>Sg1 has a highly conserved and syntenic core genome that comprises the many eukaryotic like proteins and a conserved repertoire of over 200 Dot/Icm type IV secreted substrates. However, recombination events and horizontal gene transfer are frequent. In particular the analyses of the distribution of nucleotide polymorphisms suggests that large chromosomal fragments of over 200 kbs are exchanged between <it>L. pneumophila </it>strains and contribute to the genome dynamics in the natural population. The many secretion systems present might be implicated in exchange of these fragments by conjugal transfer. Plasmids also play a role in genome diversification and are exchanged among strains and circulate between different <it>Legionella </it>species.</p> <p>Conclusion</p> <p>Horizontal gene transfer among bacteria and from eukaryotes to <it>L. pneumophila </it>as well as recombination between strains allows different clones to evolve into predominant disease clones and others to replace them subsequently within relatively short periods of time.</p
