A system of three transiting super-Earths in a cool dwarf star
We present the detection of three super-Earths transiting the cool star
LP415-17, monitored by K2 mission in its 13th campaign. High resolution spectra
obtained with HARPS-N/TNG showed that the star is a mid-late K dwarf. Using
spectral synthesis models we infer its effective temperature, surface gravity
and metallicity and subse- quently determined from evolutionary models a
stellar radius of 0.58 R Sun. The planets have radii of 1.8, 2.6 and 1.9 R
Earth and orbital periods of 6.34, 13.85 and 40.72 days. High resolution images
discard any significant contamination by an intervening star in the line of
sight. The orbit of the furthest planet has radius of 0.18 AU, close to the
inner edge of the habitable zone. The system is suitable to improve our
understanding of formation and dynamical evolution of super-Earth systems in
the rocky - gaseous threshold, their atmospheres, internal structure,
composition and interactions with host stars.Comment: Accepted for publication in MNRAS Letter
NPC1 deficiency impairs cerebellar postnatal development of microglia and climbing fiber refinement in a mouse model of Niemann-Pick disease type C.
Little is known about the effects of NPC1 deficiency in brain development and whether these effects contribute to neurodegeneration in Niemann-Pick disease type C (NPC). Degeneration of cerebellar Purkinje cells occurs at an earlier stage and to a greater extent in NPC; therefore, we analyzed the effect of NPC1 deficiency on microglia and on climbing fiber synaptic refinement during cerebellar postnatal development using th
Contribution of microscopy for understanding the mechanism of action against trypanosomatids
Transmission electron microscopy (TEM) has proved to be a useful tool to study the ultrastructural alterations and the target organelles of new antitrypanosomatid drugs. Thus, it has been observed that sesquiterpene lactones induce diverse ultrastructural alterations in both T. cruzi and Leishmania spp., such as cytoplasmic vacuolization, appearance of multilamellar structures, condensation of nuclear DNA, and, in some cases, an important accumulation of lipid vacuoles. This accumulation could be related to apoptotic events. Some of the sesquiterpene lactones (e.g., psilostachyin) have also been demonstrated to cause an intense mitochondrial swelling accompanied by a visible kinetoplast deformation as well as the appearance of multivesicular bodies. This mitochondrial swelling could be related to the generation of oxidative stress and associated to alterations in the ergosterol metabolism. The appearance of multilamellar structures and multiple kinetoplasts and flagella induced by the sesquiterpene lactone psilostachyin C indicates that this compound would act at the parasite cell cycle level, in an intermediate stage between kinetoplast segregation and nuclear division. In turn, the diterpene lactone icetexane has proved to induce the external membrane budding on T. cruzi together with an apparent disorganization of the pericellar cytoskeleton. Thus, ultrastructural TEM studies allow elucidating the possible mechanisms and the subsequent identification of molecular targets for the action of natural compounds on trypanosomatids.Fil: Lozano, Esteban Sebastián. Effect of the break-up on the fusion and elastic scattering of weakly bound projectiles on Zn
We study the behavior of the fusion, break-up, reaction and elastic
scattering of different projectiles on Zn, at near and above barrier
energies. We present fusion and elastic scattering data with the tightly bound
O and the stable weakly bound Li, Li and Be
projectiles. The data were analyzed by coupled channel calculations. The total
fusion cross sections for these systems are not affected by the break-up
process at energies above the barrier. The elastic (non-capture) break-up cross
section is important at energies close and above the Coulomb barrier and
increases the reaction cross sections. In addition we also show that the
break-up process at near and sub-barrier energies is responsible for the
vanishing of the usual threshold anomaly of the optical potential and give rise
to a new type of anomaly.Comment: 24 pages, 10 figure
Sub- and above barrier fusion of loosely bound Li with Si
Fusion excitation functions are measured for the system Li+Si
using the characteristic -ray method, encompassing both the sub-barrier
and above barrier regions, viz., = 7-24 MeV. Two separate experiments
were performed, one for the above barrier region (= 11-24 MeV) and
another for the below barrier region (= 7-10 MeV). The results were
compared with our previously measured fusion cross section for the
Li+Si system. We observed enhancement of fusion cross section at
sub-barrier regions for both Li and Li, but yield was substantially
larger for Li. However, for well above barrier regions, similar type of
suppression was identified for both the systems.Comment: 8 pages, 6 figures, as accepted for publication in Eur.Phys.J.
Activation of Serine One-Carbon Metabolism by Calcineurin A beta 1 Reduces Myocardial Hypertrophy and Improves Ventricular Function
Background In response to pressure overload, the heart develops ventricular hypertrophy that progressively decompensates and leads to heart failure. This pathological hypertrophy is mediated, among others, by the phosphatase calcineurin and is characterized by metabolic changes that impair energy production by mitochondria. Objectives The authors aimed to determine the role of the calcineurin splicing variant CnAβ1 in the context of cardiac hypertrophy and its mechanism of action. Methods Transgenic mice overexpressing CnAβ1 specifically in cardiomyocytes and mice lacking the unique C-terminal domain in CnAβ1 (CnAβ1Δi12 mice) were used. Pressure overload hypertrophy was induced by transaortic constriction. Cardiac function was measured by echocardiography. Mice were characterized using various molecular analyses. Results In contrast to other calcineurin isoforms, the authors show here that cardiac-specific overexpression of CnAβ1 in transgenic mice reduces cardiac hypertrophy and improves cardiac function. This effect is mediated by activation of serine and one-carbon metabolism, and the production of antioxidant mediators that prevent mitochondrial protein oxidation and preserve ATP production. The induction of enzymes involved in this metabolic pathway by CnAβ1 is dependent on mTOR activity. Inhibition of serine and one-carbon metabolism blocks the beneficial effects of CnAβ1. CnAβ1Δi12 mice show increased cardiac hypertrophy and declined contractility. Conclusions The metabolic reprogramming induced by CnAβ1 redefines the role of calcineurin in the heart and shows for the first time that activation of the serine and one-carbon pathway has beneficial effects on cardiac hypertrophy and function, paving the way for new therapeutic approaches
PIP5KIβ Selectively Modulates Apical Endocytosis in Polarized Renal Epithelial Cells
Localized synthesis of phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] at clathrin coated pits (CCPs) is crucial for the recruitment of adaptors and other components of the internalization machinery, as well as for regulating actin dynamics during endocytosis. PtdIns(4,5)P2 is synthesized from phosphatidylinositol 4-phosphate by any of three phosphatidylinositol 5-kinase type I (PIP5KI) isoforms (α, β or γ). PIP5KIβ localizes almost exclusively to the apical surface in polarized mouse cortical collecting duct cells, whereas the other isoforms have a less polarized membrane distribution. We therefore investigated the role of PIP5KI isoforms in endocytosis at the apical and basolateral domains. Endocytosis at the apical surface is known to occur more slowly than at the basolateral surface. Apical endocytosis was selectively stimulated by overexpression of PIP5KIβ whereas the other isoforms had no effect on either apical or basolateral internalization. We found no difference in the affinity for PtdIns(4,5)P2-containing liposomes of the PtdIns(4,5)P2 binding domains of epsin and Dab2, consistent with a generic effect of elevated PtdIns(4,5)P2 on apical endocytosis. Additionally, using apical total internal reflection fluorescence imaging and electron microscopy we found that cells overexpressing PIP5KIβ have fewer apical CCPs but more internalized coated structures than control cells, consistent with enhanced maturation of apical CCPs. Together, our results suggest that synthesis of PtdIns(4,5)P2 mediated by PIP5KIβ is rate limiting for apical but not basolateral endocytosis in polarized kidney cells. PtdIns(4,5)P2 may be required to overcome specific structural constraints that limit the efficiency of apical endocytosis. © 2013 Szalinski et al
Comprehensive study of reaction mechanisms for the Be9+Sm144 system at near- and sub-barrier energies
The delayed x-ray detection technique was used to measure complete and incomplete fusion cross sections for the reaction at sub- and near-barrier energies. Elastic and inelastic scattering for this system were also measured. Reaction cross sections were derived and the transfer cross sections of one neutron were calculated. The suppression of complete fusion above the barrier, of the order of 10%, is attributed to breakup and is considerably smaller than the value of 30% found for the system
