Efficiency of application original preservatives for conservation leukocytes at -40 °C

    Get PDF
When frozen leukocytes exposed to harmful factors of the complex, due to their complex cellular structure and high metabolism. Cryopreservatives allow to avoid damages, but most of which are toxic. The aim of the present was to compare the efficacy of application of two non-toxic solutions for conservation of leukocytes at -40°С for 1 day. It was show that solution containing cryoprotector mixed action (a derivative of urea) and antihypoxant (sodium fumarate) is most effective in preserving the functional activity of leukocytes

Association Between Long-Term Cognitive Decline in Vietnam Veterans With TBI and Caregiver Attachment Style.

OBJECTIVE: To examine whether a caregiver's attachment style is associated with patient cognitive trajectory after traumatic brain injury (TBI). SETTING: National Institute of Neurological Disorders and Stroke, National Institutes of Health, Bethesda, Maryland. PARTICIPANTS: Forty Vietnam War veterans with TBI and their caregivers. MAIN OUTCOME MEASURE: Cognitive performance, measured by the Armed Forces Qualification Test percentile score, completed at 2 time points: preinjury and 40 years postinjury. DESIGN: On the basis of caregivers' attachment style (secure, fearful, preoccupied, dismissing), participants with TBI were grouped into a high or low group. To examine the association between cognitive trajectory of participants with TBI and caregivers' attachment style, we ran four 2 × 2 analysis of covariance on cognitive performances. RESULTS: After controlling for other factors, cognitive decline was more pronounced in participants with TBI with a high fearful caregiver than among those with a low fearful caregiver. Other attachment styles were not associated with decline. CONCLUSION AND IMPLICATION: Caregiver fearful attachment style is associated with a significant decline in cognitive status after TBI. We interpret this result in the context of the neural plasticity and cognitive reserve literatures. Finally, we discuss its impact on patient demand for healthcare services and potential interventions

Extraction of the resonance parameters at finite times

In this paper we propose a model-independent method to extract the resonance parameters on the lattice directly from the Euclidean 2-point correlation functions of the field operators at finite times. The method is tested in case of the two-point function of the Delta-resonance, calculated at one loop in Small Scale Expansion. Further, the method is applied to a 1+1-dimensional model with two coupled Ising spins and the results are compared with earlier ones obtained by using Luescher's approach.Comment: 20 pages, 6 figure

    Production of α1,3-galactosyltransferase-deficient pigs

The enzyme α1,3-galactosyltransferase (α1,3GT or GGTA1) synthesizes α1,3galactose (α1,3Gal) epitopes (Galα1,3Galβ1,4GlcNAc-R), which are the major xenoantigens causing hyperacute rejection in pig-to-human xenotransplantation. Complete removal of α1,3Gal from pig organs is the critical step toward the success of xenotransplantation. We reported earlier the targeted disruption of one allele of the α1,3GT gene in cloned pigs. A selection procedure based on a bacteria[toxin was used to select for cells in which the second allele of the gene was knocked out. Sequencing analysis demonstrated that knockout of the second allele of the α1,3GT gene was caused by a T-to-G single point mutation at the second base of exon 9, which resulted in inactivation of the α1,3GT protein. Four healthy α1,3GT double-knockout female piglets were produced by three consecutive rounds of cloning. The piglets carrying a point mutation in the α1,3GT gene hold significant value, as they would allow production of α1,3Gal-deficient pigs free of antibiotic-resistance genes and thus have the potential to make a safer product for human use

Here, we report the 10,807-nucleotide-long consensus RNA genome sequences of three spatiotemporally distinct and genetically divergent Zika virus strains, with the functionality of their genomic sequences substantiated by reverse genetics: MR-766 (African lineage, Uganda, 1947), P6-740 (Asian lineage, Malaysia, 1966), and PRVABC-59 (Asian lineage-derived American strain, Puerto Rico, 2015)

Comparison of Meat Composition from Offspring of Cloned and Conventionally Produced Boars

This study compares the meat composition of the offspring from boars produced by somatic cell nuclear transfer (n = 4) to that of the offspring from conventionally produced boars (n = 3). In total, 89 commercial gilts were artificially inseminated and 61 progressed to term and farrowed. All of the resulting piglets were housed and raised identically under standard commercial settings and slaughtered upon reaching market weight. Loin samples were taken from each slaughtered animal and shipped offsite for meat composition analysis. In total, loin samples from 404 animals (242 from offspring of clones and 162 from controls) were analyzed for 58 different parameters generating 14,036 and 9396 data points from offspring of clones and the controls, respectively. Values for controls were used to establish a range for each parameter. Ten percent was then added to the maximum and subtracted from the minimum of the control range, and all results within this range were considered clinically irrelevant. Of the 14,036 data points from the offspring of clones, only three points were found outside the clinically irrelevant range, two of which were within the range established by the USDA National Nutrient Database for Standard Reference, Release 18, 2005. The only outlier was the presence of Eicosadienoic acid (C20:2) in one sample which is typically present in minute quantities in pork; no reference data were found regarding this fatty acid in the USDA National Nutrient Database. In conclusion, these data indicated that meat from the offspring of clones was not chemically different than meat from controls and therefore supported the case for the safety of meat from the offspring of clones

Types of life strategy of people with disabilities in the perspective of labor employment

The article analyzes the concept of "life strategy", identifies the features and factors of life strategies of people with disabilities. Based on the results of the empirical research carried out by the authors, a typology of life strategies of disabled people – self-realization, adaptation, survival. Each of the selected strategies is characterized by a certain combination of components of the strategy, including the ability to solve life problems, social activity, self-esteem, value orientations of people with disabilities. The predominant strategies of working and non-working disabled people are shown.
