    Potential of PM-selected components to induce oxidative stress and root system alteration in a plant model organism

    Over the last years, various acellular assays have been used for the evaluation of the oxidative potential (OP) of particular matter (PM) to predict PM capacity to generate reactive oxygen (ROS) and nitrogen (RNS) species in biological systems. However, relationships among OP and PM toxicological effects on living organisms are still largely unknown. This study aims to assess the effects of atmospheric PM-selected components (brake dust - BD, pellet ash - PA, road dust - RD, certified urban dust NIST1648a - NIST, soil dust - S, coke dust - C and Saharan dust - SD) on the model plant A. thaliana development, with emphasis on their capacity to induce oxidative stress and root morphology alteration. Before growing A. thaliana in the presence of the PM-selected components, each atmospheric dust has been chemically characterized and tested for the OP through dithiothreitol (DTT), ascorbic acid (AA) and 2′,7′-dichlorofluorescin (DCFH) assays. After the exposure, element bioaccumulation in the A. thaliana seedlings, i.e., in roots and shoots, was determined and both morphological and oxidative stress analyses were performed in roots. The results indicated that, except for SD and S, all the tested dusts affected A. thaliana root system morphology, with the strongest effects in the presence of the highest OPs dusts (BD, PA and NIST). Principal component analysis (PCA) revealed correlations among OPs of the dusts, element bioaccumulation and root morphology alteration, identifying the most responsible dust-associated elements affecting the plant. Lastly, histochemical analyses of NO and O2•− content and distribution confirmed that BD, PA and NIST induce oxidative stress in A. thaliana, reflecting the high OPs of these dusts and ultimately leading to cell membrane lipid peroxidation

Os registros mostraram que as três porções do músculo trapézio participam no movimento de circundução do braço girando a roda de ombro, sendo que a fase de elevação do braço mostrou-se mais ativa nos movimentos realizados no sentido horário (MHRT e MHR/2) e, a fase de abaixamento foi mais ativa nos movimentos realizados no sentido anti-horário (MAHRT e MAHR/2). Não foram encontradas diferenças significativas nos potenciais de ação para nenhuma das porções do músculo trapézio, quando se procedeu a variação do raio da roda de ombroAbstract: The participation of the upper, middle and power portions of the trapezius muscle during the circumduction movement of the arm rotating the shoulder wheel, was analyzed electromyographically.     In-vivo exposure of a plant model organism for the assessment of the ability of PM samples to induce oxidative stress

    This study aims to propose an innovative, simple, rapid, and cost-effective method to study oxidative stress induced by PM through in-vivo exposure of the plant model organism Arabidopsis thaliana. A. thaliana seedlings were exposed to urban dust certified for its elemental content and to PM2.5 samples collected in an urban-industrial area of Northern Italy. An innovative technique for the detachment and suspension in water of the whole intact dust from membrane filters was applied to expose the model organism to both the soluble and insoluble fractions of PM2.5, which were analyzed for 34 elements by ICP-MS. Oxidative stress induced by PM on A. thaliana was assessed by light microscopic localization and UV–Vis spectrophotometric determination of superoxide anion (O2−) content on the exposed seedlings by using the nitro blue tetrazole (NBT) assay. The results showed a good efficiency and sensitivity of the method for PM mass concentrations >20 μg m−3 and an increase in O2− content in all exposed seedlings, which mainly depends on the concentration, chemical composition, and sources of the PM administered to the model organism. Particles released by biomass burning appeared to contribute more to the overall toxicity of PM. This method was found to be cost-effective and easy to apply to PM collected on membrane filters in intensive monitoring campaigns in order to obtain valuable information on the ability of PM to generate oxidative stress in living organisms

    CMB Polarization Systematics, Cosmological Birefringence and the Gravitational Waves Background

    Cosmic Microwave Background experiments must achieve very accurate calibration of their polarization reference frame to avoid biasing the cosmological parameters. In particular, a wrong or inaccurate calibration might mimic the presence of a gravitational wave background, or a signal from cosmological birefringence, a phenomenon characteristic of several non-standard, symmetry breaking theories of electrodynamics that allow for \textit{in vacuo} rotation if the polarization direction of the photon. Noteworthly, several authors have claimed that the BOOMERanG 2003 (B2K) published polarized power spectra of the CMB may hint at cosmological birefringence. Such analyses, however, do not take into account the reported calibration uncertainties of the BOOMERanG focal plane. We develop a formalism to include this effect and apply it to the BOOMERanG dataset, finding a cosmological rotation angle α=4.3±4.1\alpha=-4.3^\circ\pm4.1^\circ. We also investigate the expected performances of future space borne experiment, finding that an overall miscalibration larger then 11^\circ for Planck and 0.20.2\circ for EPIC, if not properly taken into account, will produce a bias on the constraints on the cosmological parameters and could misleadingly suggest the presence of a GW background.Comment: 10 pages, 3 figure

    A polarization modulator unit for the mid- and high-frequency telescopes of the LiteBIRD mission

    The LiteBIRD mission is a JAXA strategic L-class mission for all sky CMB surveys which will be launched in the 2020s. The main target of the mission is the detection of primordial gravitational waves with a sensitivity of the tensor-to-scalar ratio δr<0.001. The polarization modulator unit (PMU) represents a critical and powerful component to suppress 1/f noise contribution and mitigate systematic uncertainties induced by detector gain drift, both for the high-frequency telescope (HFT) and for the mid-frequency telescope (MFT). Each PMU is based on a continuously-rotating transmissive half-wave plate (HWP) held by a superconducting magnetic bearing in a 5K environment. In this contribution we will present the design and expected performance of the LiteBIRD PMUs and testing performed on every PMU subsystem with a room-temperature rotating disk used as a stand-in for the cryogenic HWP rotor

    The “Special” crystal-Stellate System in Drosophila melanogaster Reveals Mechanisms Underlying piRNA Pathway-Mediated Canalization

    The Stellate-made crystals formation in spermatocytes is the phenotypic manifestation of a disrupted crystal-Stellate interaction in testes of Drosophila melanogaster. Stellate silencing is achieved by the piRNA pathway, but many features still remain unknown. Here we outline the important role of the crystal-Stellate modifiers. These have shed light on the piRNA pathways that defend genome integrity against transposons and other repetitive elements in the gonads. In particular, we illustrate the finding that HSP90 participates in the molecular pathways of piRNA production. This observation has relevance for the mechanisms underlying the evolutionary canalization process

    Consensus communication strategies to improve doctor-patient relationship in paediatric severe asthma

    Background: Asthma is a chronic inflammatory disease that is very common among youth worldwide. The burden of this illness is very high not only considering financial costs but also on emotional and social functioning. Guidelines and many researches recommend to develop a good communication between physicians and children/caregiver and their parents. Nevertheless, a previous Italian project showed some criticalities in paediatric severe asthma management. The consensus gathered together experts in paediatric asthma management, experts in narrative medicine and patient associations with the aim of identify simple recommendation to improve communication strategies. Methods: Participants to the consensus received the results of the project and a selection of narratives two weeks before the meeting. The meeting was structured in plenary session and in three working groups discussing respectively about communication strategies with children, adolescents and parents. The task of each working group was to identify the most effective (DO) and least effective practices (DON' T) for 5 phases of the visit: welcome, comprehension of the context, emotions management, duration and end of the visit and endurance of the relationship. Results: Participants agreed that good relationships translate into positive outcomes and reached consensus on communication strategies to implement in the different phase of relationships. Conclusions: The future challenges identified by the participants are the dissemination of this Consensus document and the implementation of effective communication strategies to improve the management of pediatric asthma
