This eukaryote feeds as single cells but starvation triggers the formation of a multicellular organism in response to extracellular pulses of cAMP and the eventual generation of spores. Strains in which the gene encoding Cdk8 have been disrupted fail to form multicellular aggregates unless supplied with exogenous pulses of cAMP and later in development, cdk8- cells show a defect in spore production. RESULTS: Microarray analysis revealed that the cdk8- strain previously described (cdk8-HL) contained genome duplications. Regeneration of the strain in a background lacking detectable gene duplication generated strains (cdk8-2) with identical defects in growth and early development, but a milder defect in spore generation, suggesting that the severity of this defect depends on the genetic background. The failure of cdk8- cells to aggregate unless rescued by exogenous pulses of cAMP is consistent with a failure to express the catalytic subunit of protein kinase A. However, overexpression of the gene encoding this protein was not sufficient to rescue the defect, suggesting that this is not the only important target for Cdk8 at this stage of development. Proteomic analysis revealed two potential targets for Cdk8 regulation, one regulated post-transcriptionally (4-hydroxyphenylpyruvate dioxygenase (HPD)) and one transcriptionally (short chain dehydrogenase/reductase (SDR1)). CONCLUSIONS: This analysis has confirmed the importance of Cdk8 at multiple stages of Dictyostelium development, although the severity of the defect in spore production depends on the genetic background. Potential targets of Cdk8-mediated gene regulation have been identified in Dictyostelium which will allow the mechanism of Cdk8 action and its role in development to be determined.Published versio

Assessment of novel gear designs to reduce interactions between species

This project tested modified gillnets designed by commercial net fishers in the Queensland East Coast Inshore Finfish Fishery (ECIFF) to try and identify gears that would mitigate and/or improve interactions between fishing nets and Species of Conservation Interest (SOCI). The study also documents previously unrecognised initiatives by pro-active commercial net fishers that reflect a conservation-minded approach to their fishing practices, which is the opposite of what is perceived publicly. Between 2011 and 2014, scientists from James Cook University and the Queensland Department of Agriculture and Fisheries teamed with commercial fishers representing the Queensland Seafood Industry Association and the Moreton Bay Seafood Industry Association to conduct field trials of various modified net designs under normal fishery conditions. Trials were conducted in Moreton Bay (southern part of the fishery) and Bowling Green Bay (northern) and tested different net designs developed by fishers to improve the nature of interactions between net fishing gear and SOCI

Effectiveness of immersive virtual reality in surgical training—A randomized control trial

Purpose: Surgical training methods are evolving with the technological advancements, including the application of virtual reality (VR) and augmented reality. However, 28 to 40% of novice residents are not confident in performing a major surgical procedure. VR surgery, an immersive VR (iVR) experience, was developed using Oculus Rift and Leap Motion devices (Leap Motion, Inc, San Francisco, CA) to address this challenge. Our iVR is a multisensory, holistic surgical training application that demonstrates a maxillofacial surgical technique, the Le Fort I osteotomy. The main objective of the present study was to evaluate the effect of using VR surgery on the self-confidence and knowledge of surgical residents. Materials and Methods: A multisite, single-blind, parallel, randomized controlled trial (RCT) was performed. The participants were novice surgical residents with limited experience in performing the Le Fort I osteotomy. The primary outcome measures were the self-assessment scores of trainee confidence using a Likert scale and an objective assessment of the cognitive skills. Ninety-five residents from 7 dental schools were included in the RCT. The participants were randomly divided into a study group of 51 residents and a control group of 44. Participants in the study group used the VR surgery application on an Oculus Rift with Leap Motion device. The control group participants used similar content in a standard PowerPoint presentation on a laptop. Repeated measures multivariate analysis of variance was applied to the data to assess the overall effect of the intervention on the confidence of the residents. Results: The study group participants showed significantly greater perceived self-confidence levels compared with those in the control group (P = .034; α = 0.05). Novices in the first year of their training showed the greatest improvement in their confidence compared with those in their second and third year. Conclusions: iVR experiences improve the knowledge and self-confidence of the surgical residents

Parental Monitoring of Children’s Media Consumption: The Long-term Influences on Body Mass Index in Children

IMPORTANCE: Although children’s media consumption has been one of the most robust risk factors for childhood obesity, effects of specific parenting influences, such as parental media monitoring, have not been effectively investigated. OBJECTIVES: This study examined the potential influences of maternal and paternal monitoring of child media exposure and children’s general activities on children’s BMI in middle childhood. DESIGN: A longitudinal study, taken from a subsample of the Three Generational Study, with assessments at children’s ages of 5, 7, and/or 9 years collected from 1998-2012. SETTING: The Three Generational Study, a predominantly Caucasian, Pacific-Northwest U.S. community sample (overall participation rate 90%). PARTICIPANTS: Analyses included 112 mothers, 103 fathers and their 213 children (55% girls) at ages 5, 7, and/or 9 years. Participation rates ranged from 67% to 72% of all eligible Three Generational Study children across the three assessments. MAIN EXPOSURES: Parents reported on their general monitoring of their children (whereabouts and activities), specific monitoring of child media exposure, children’s participation in sports and recreational activities, children’s media time (hours/week), annual income, and education level. Parental BMI was recorded. MAIN OUTCOME MEASURE: Predictions to level and change in child BMI z scores were tested. RESULTS: Linear mixed-effects modeling indicated that more maternal, but not paternal, monitoring of child media exposure predicted lower child BMI z scores at age 7 years (95% CI, -.39 – -.07) and less steeply increasing child BMI z scores from ages 5-9 years (95% CI, -.11 – -.01). These effects held when controlling for more general parental monitoring, and parent BMI, income and education. Results supported that the significant negative effect of maternal media monitoring on children’s BMI z scores at age 7 years was marginally accounted for by the effect of child media time. The maternal media monitoring effect on children’s BMI z score slopes remained significant once adjusting for children’s media time, and sports and recreational activity. CONCLUSIONS: This study suggests that parental behaviors related to children’s media consumption may have long-term impacts on children’s BMI in middle childhood. The results underscore the importance of targeting parental media monitoring in efforts to prevent childhood obesity
