Testicular degeneration in Bclw-deficient mice
To identify genes required for mammalian spermatogenesis, we screened lines of mutant mice created using a retroviral gene-trap system for male infertility. Homozygous ROSA41 male mice exhibit sterility associated with progressive testicular degeneration. Germ-cell defects are first observed at 19 days post-natal (p19). Spermatogenesis is blocked during late spermiogenesis in young adults. Gradual depletion of all stages of germ cells results in a Sertoli-cell-only phenotype by approximately six months of age. Subsequently, almost all Sertoli cells are lost from the seminiferous tubules and the Leydig cell population is reduced. Molecular analysis indicates that the gene mutated is Bclw, a death-protecting member of the Bcl2 family. The mutant allele of Bclw in ROSA41 does not produce a Bclw polypeptide. Expression of Bclw in the testis appears to be restricted to elongating spermatids and Sertoli cells. Potential roles for Bclw in testicular function are discussed

Effects of Combined Long-Term Treatment with a Growth Hormone-Releasing Hormone Analogue and a Growth Hormone Secretagogue in the Growth Hormone-Releasing Hormone Knock Out Mouse
GH secretagogues (GHS) are synthetic ghrelin receptor agonists that stimulate GH secretion. It is not clear whether they act predominantly by stimulating the secretion of hypothalamic growth hormone-releasing hormone (GHRH), or directly on the somatotrope cells. In addition, it is not known whether combined treatment with GHRH and GHS has synergistic effects on growth. To address these questions, we used the GH-deficient GHRH knock out (GHRHKO) mouse model, which has severe somatotrope cell hypoplasia. We treated GHRHKO mice for 5 weeks (from week 1 to week 6 of age) with the GHRH analogue JI-38 alone, or in combination with a GHS (GHRP-2), and at the end of the treatment we examined their response to an acute stimulus with GHRP-2 or GHRP-2 plus JI-38. We used placebo-treated GHRHKO mice and animals heterozygous for the GHRHKO allele as controls. Animals treated with JI-38+GHRP-2 reached higher body length and weight than animals treated with JI-38 alone. All the animals receiving JI-38 (with or without GHRP-2) showed similar correction of somatotrope cell hypoplasia. None of the GHRHKO animals showed a serum GH response to the acute stimulation with GHRP-2 alone, while both treated groups responded to the combined test with JI-38 + GHRP-2. These data demonstrate that in GHRHKO mice, GHRP-2 has a growth-stimulating effect that augments the response induced by JI-38. Effetto di una dieta carente di iodio sulla funzione tiroidea in topi knock out per il gene della pendrina
La sindrome di Pendred \ue8 un disordine genetico recessivo caratterizzato da sordit\ue0 neurosensoriale, gozzo e difetti nell\u2019organificazione dello iodio. Tale sindrome \ue8 causata da mutazioni del gene PDS che codifica per un trasportatore anionico (Pendrina) espresso nella tiroide, nell\u2019orecchio interno e nel rene. Nella tiroide la Pendrina si localizza sulla superficie apicale delle cellule follicolari, dove regola il trasporto di iodio dal citoplasma alla colloide. La Sindrome di Pendred presenta un\u2019 estrema variabilit\ue0 inter- ed intrafamiliare in termini di gravit\ue0 dei sintomi ed et\ue0 di insorgenza. In particolare le disfunzioni tiroidee sono presenti solo nel 50% circa dei pazienti e variano da un leggero aumento del volume della ghiandola alla presenza di gozzo di notevoli dimensioni. Tale variabilit\ue0 \ue8 probabilmente dovuta all\u2019interazione tra fattori genetici e ambientali, quali l\u2019apporto di iodio. In questo studio abbiamo valutato l\u2019effetto di una dieta carente di iodio sul fenotipo tiroideo di topi knock-out per la Pendrina (PDS -/-). I topi PDS -/- sviluppano nel corso della prime settimane di vita alterazioni a carico dell\u2019orecchio interno tipiche della sindrome di Pendred , ma la funzione tiroidea \ue8 riportata essere nella norma. Tuttavia \ue8 da considerare il fatto che tali studi sono stati condotti su animali alimentati con un mangime ad alto contenuto di iodio mentre non \ue8 stata finora valutata la funzionalit\ue0 tiroidea in presenza di insufficiente apporto di iodio. Per chiarire questo aspetto abbiamo mantenuto una colonia di topi con diverso genotipo (PDS +/+; PDS +/-; PDS -/-) suddivisa in due gruppi di trattamento: gruppo sperimentale (S, n=21) e gruppo controllo (C, n=22). Il gruppo S \ue8 stato sottoposto ad alimentazione controllata con mangime semisintetico a basso contenuto di iodio e l\u2019aggiunta di una dose minima di ioduro nell\u2019acqua (KI 2ng/ml). Il gruppo C ha ricevuto invece una dieta normale, tipicamente ad elevato contenuto di iodio. Tutti gli animali sono stati genotipizzati ed \ue8 stato eseguito un prelievo di sangue a 6 mesi per la valutazione della funzionalit\ue0 tiroidea tramite dosaggio di TSH e TT4 mediante RIA. I valori di TSH non rivelano differenze significative tra i vari gruppi. In particolare non sembrano influenzati n\ue9 dal genotipo, n\ue9 dall\u2019apporto di iodio (PDS -/- gruppo S: 203,7 +/- 14,09 ng/ml; PDS -/- gruppo C: 217,1 +/- 18,86 ng/ml). Non sono state osservate differenze significative anche per quanto riguarda i valori di TT4 (PDS -/- gruppo S: 55,57 +/- 12,75 nmol/L; PDS-/- gruppo C : 74,03 +/- 7,54 nmol/L). Sulla base di questi dati \ue8 possibile concludere che una dieta carente di iodio non sembra influenzare in maniera rilevante il fenotipo tiroideo di topi PDS -/-. \uc8 pertanto ipotizzabile che nel topo esistano altri sistemi di trasporto in grado di compensare la mancanza della Pendrina. Studi futuri sono necessari per identificare ulteriori meccanismi di trasporto apicale dello iodio e chiarire le eventuali differenze presenti tra la patologia umana e il modello murino
Overexpression of the HMGA2 gene in transgenic mice leads to the onset of pituitary adenomas.
Overexpression of the HMGA2 gene is a common feature of neoplastic cells both in experimental and human models. Intragenic and extragenic HMGA2 rearrangements responsible for HMGA2 gene overexpression have been frequently detected in human benign tumours of mesenchymal origin. To better understand the role of HMGA2 overexpression in human tumorigenesis, we have generated transgenic mice carrying the HMGA2 gene under the transcriptional control of the cytomegalovirus promoter. High expression of the transgene was demonstrated in all the mouse tissues analysed, whereas no expression of the endogenous HMGA2 gene was detected in the same tissues from wild-type mice. In this study, two independent lines of transgenic mice have been generated. By 6 months of age, 85% of female animals of both transgenic lines developed pituitary adenomas secreting prolactin and growth hormone. The transgenic males developed the same phenotype with a lower penetrance (40%) and a longer latency period (about 18 months). Therefore, these data demonstrate that the overexpression of HMGA2 leads to the onset of mixed growth hormone/prolactin cell pituitary adenomas. These transgenic mice may represent an important tool for the study of this kind of neoplasia

Extensively unilateral recruitment for daily activities may determine performance asymmetries in favor of the preferred side eliciting functional adaptation. Our study evaluated asymmetries in elbow torque output between preferred and non-preferred limbs. Eighteen subjects performed maximal elbow flexor and extensor isometric contractions at five different elbow joint angles (0º, 30º, 60º, 90º, 120º) and five different angular velocities (60, 120, 180, 240, 300º.s-1) on an isokinetic dynamometer. Higher flexor torque in favor of preferred arm was observed at 90º of flexion (p<0.05), which also corresponded to the highest torque produced (p<0.05). The fact that joint angle influenced torque asymmetries, whereas angular velocity did not, suggest that the observed asymmetry is likely related to preferential recruitment of elbow flexors at a 90º joint angle for daily tasks requiring high levels of force production. Muscle functional adaptation to frequent stimuli at this joint angle in healthy subjects may explain these results.<br>O frequente recrutamento unilateral de membros superiores pode determinar assimetrias de desempenho em favor do lado preferido, resultando em adaptação funcional. Assimetrias no torque gerado pelos músculos do cotovelo entre o membro preferido e não-preferido foram avaliadas. Dezoito sujeitos realizaram contrações máximas de flexo-extensão do cotovelo em cinco ângulos articulares (0º, 30º, 60º, 90º, 120º) e cinco velocidades angulares (60, 120, 180, 240, 300º.s-1) em um dinamômetro isocinético. Torque flexor mais elevado em favor do lado preferido foi encontrado no ângulo de 90º (p<0,05), que também correspondeu ao ângulo de maior torque (p<0,05). O fato de o ângulo articular determinar assimetrias no torque (enquanto a velocidade angular não) sugere que o recrutamento preferencial dos flexores do cotovelo em um ângulo de 90º nas tarefas da vida diária que requerem força elevada é responsável pela assimetria. Adaptação funcional a estÃmulos frequentes nesse ângulo articular pode explicar esses resultados em sujeitos saudáveis
