Eucalyptus globulus Labill. decoction extract inhibits the growth of NCI-H460 cells by increasing the p53 levels and altering the cell cycle profile
Eucalyptus globulus Labill. is a widespread evergreen plant belonging to the Myrtaceae family. Several
species of Eucalyptus are known to have a plethora of medicinal properties, particularly anti-tumor
activity, which prompts the study of the chemical composition and bioactivity of extracts from this plant.
Hereby, the main aims of this work were to (i) profile the phenolic compounds in E. globulus extracts prepared
by decoction and infusion; (ii) test the cell growth inhibitory activity of E. globulus decoction and
infusion, in three human tumor cell line models: colorectal, pancreatic and non-small cell lung cancer
(HCT-15, PANC-1 and NCI-H460, respectively); and (iii) study the mechanism of action of the most
potent extract in the most sensitive cell line. Our work demonstrated that both the decoction and infusion
preparations revealed the presence of phenolic acids, flavonoids and gallotannins, the last group being
the most abundant polyphenols found, especially two digalloyl-glucosides. Both extracts inhibited the
growth of all the tumor cell lines tested. The decoction extract was the most potent in inhibiting the
NCI-H460 cell growth (lower GI50 determined by sulforhodamine B assay), which could be due to its
higher content of phenolic compounds. Hence, the effect of the decoction extract on the NCI-H460
cells was further investigated. For this, cell viability (by Trypan blue exclusion assay), the cell cycle profile
and apoptosis (by flow cytometry), cell proliferation (by bromodeoxyuridine assay) and protein expression
(by western blot) were analyzed. Two different concentrations of the extract (52 μg mL−
1 and 104
μg mL−1, corresponding to GI50 and 2 × GI50 concentration) were tested in these studies. Remarkably,
the E. globulus decoction extract caused a dose-dependent decrease in the NCI-H460 cell number,
which was correlated with a cell cycle arrest in the G0/G1 phase, a decrease in cell proliferation and an
increase in the expression of p53, p21 and cyclin D1 proteins. Interestingly, no differences were found in
the levels of ds-DNA damage and in the levels of apoptosis. This work highlights the relevance of the

Chronic kidney disease has a graded association with death and cardiovascular outcomes in stable coronary artery disease : An analysis of 21,911 patients from the CLARIFY registry
Chronic kidney disease (CKD) is associated with an increased cardiovascular risk in a broad spectrum of populations. However, the risk associated with a reduced estimated glomerular filtration rate (eGFR) in patients with stable coronary artery disease receiving standard care in the modern era, independently of baseline cardiovascular disease, risk factors, and comorbidities, remains unclear. We analyzed data from 21,911 patients with stable coronary artery disease, enrolled in 45 countries between November 2009 and July 2010 in the CLARIFY registry. Patients with abnormal renal function were older, with more comorbidities, and received slightly lower-although overall high-rates of evidence-based secondary prevention therapies than patients with normal renal function. The event rate of patients with CKD stage 3b or more (eGFR <45 mL/min/1.73 m) was much higher than that associated with any comorbid condition. In a multivariable adjusted Cox proportional hazards model, lower eGFR was independently associated with a graded increased risk of cardiovascular mortality, with adjusted HRs (95% CI) of 0.98 (0.81-1.18), 1.31 (1.05-1.63), 1.77 (1.38-2.27), and 3.12 (2.25-4.33) for eGFR 60-89, 45-59, 30-44, and <30 mL/min/1.73 m, compared with eGFR ≥90 mL/min/1.73 m. A strong graded independent relationship exists between the degree of CKD and cardiovascular mortality in this large cohort of patients with chronic coronary artery disease, despite high rates of secondary prevention therapies. Among clinical risk factors and comorbid conditions, CKD stage 3b or more is associated with the highest cardiovascular mortality

Comb, cloaca and feet scores and testis morphometry in male broiler breeders at two different ages
ABSTRACT In broiler breeder roosters, the weight of the testes is positively associated with daily sperm production and fertility. In birds, the testes are located in the coelomic cavity, not being accessible to reproductive evaluation as mammalian testes. The reproductive evaluation of roosters is based on phenotypic traits. Any changes on testes will be reflected on fertility levels. The aim of this paper was to evaluate the potential of association of phenotypic traits as body weight (BW), comb scores (COS), cloaca scores (CLS) and feet scores (FS) with testicular morphometric parameters in adult Cobb® Mx roosters in two ages (25 and 45 weeks of age). The low BW roosters had lower testes weight (TW), smaller seminiferous tubule diameter (STD) and lower comb and cloaca scores than the medium and high BW roosters (P≤0.05). Heavy roosters had the highest COS and bigger seminiferous epithelium height (SEH). There was no statistical difference between the BW categories in FS and volumetric proportion of seminiferous tubule (ST) and interstitial tissue (IT). Considering the age effect, roosters with 45 weeks of age had lower TW and ST than 25-week roosters. Positive, moderate and significant correlation was found between testes weight and phenotypic characteristics as BW, COS and CLS in 45week roosters. A positive, moderate and significant correlation was found between body weight and histologic testicular characteristics as STD and SHE in both evaluated ages. In conclusion, there is a regression in the testes weight with age and it is more intense in Cobb® Mx roosters with lower BW. Furthermore, it is concluded that the BW positively influences the testes weight and histological quality of the testes. COS and CLS can be used with moderate potential prediction to identify roosters with low testes weight at 45 weeks of age.</jats:p
Scalar field "mini--MACHOs": a new explanation for galactic dark matter
We examine the possibility that galactic halos are collisionless ensembles of
scalar field ``massive compact halo objects'' (MACHOs). Using mass constraints
from MACHO microlensing and from theoretical arguments on halos made up of
massive black holes, as well as demanding also that scalar MACHO ensambles of
all scales do not exhibit gravothermal instability (as required by consistency
with observations of LSB galaxies), we obtain the range: m\alt 10^{-7}
M_\odot or 30 M_\odot\alt m\alt 100 M_\odot. The rather narrow mass range of
large MACHOs seems to indicate that the ensambles we are suggesting should be
probably made up of scalar MACHOs in the low mass range (``mini--MACHOs''). The
proposed model allows one to consider a non--baryonic and non--thermal
fundamental nature of dark matter, while at the same time keeping the same
