Protease activity in the medium of larch (Larix spec.) embryogenic suspension cultures and medium-protein stabilization by compatible solutes

The stability of recombinant, secreted protein in the medium of transgenic plant cell cultures heavily determines the resulting protein yield, which is a crucial factor for every production system. In order to gain more knowledge about the feasability of rapidly growing larch (Larix sp.) embrogenic cell cultures as a possible expression system for recombinant proteins, spent cell culture medium was characterized in this study. An accumulation of endogenous proteins could be observed in the medium of larch embrogenic suspension cultures which reached up to 1750 μg per g fresh weight. In contrast, low protease activity accumulated within a typical 14-day culture period in the medium. This activity was up to 20 times lower than the protease activity in two callus-derived suspension cultures of tobacco (genotype R1 and BY-2) which were measured in parallel. To asses the stability of foreign proteins, medium aliquots were spiked with Immunoglobulin G (IgG) and the amount of protein degradation was determined after 23 h of incubation by SDS-PAGE. The loss of IgG was comparable in three different larch genotypes, resulting in a mean loss of 18 % during the incubation time. This loss could remarkably be diminished by the addition of ectoin derivatives, known to be protein-protective „compatible solutes“ of bacterial origin. The most effective one was hydroxyectoin which resulted in a 76 % reduction of the observed IgG degradation. The stabilization of proteins in plant cell culture medium by compatible solutes is shown here for the first time. The possible mechanism of the stabilizing effect is discussed

Evaluation of a semi-automated von willebrand factor multimer assay, the Hydragel 5 von Willebrand multimer, by two European Centers

Background The phenotypic diagnosis of von Willebrand disease (VWD) is a multistep process with classification dependent on the quantification of von Willebrand factor (VWF) multimeric structure. VWF multimer analysis is a technically challenging, lengthy and non‐standardised assay, usually performed in specialist laboratories. Recently, a new semi‐automated multimer assay, the Hydragel 5 von Willebrand multimers (H5VWM) has become available. Objectives This study, performed in two European centres, compared existing in‐house multimer assays to the H5VWM in individuals with and without VWD. Results Overall agreement of 91.1% was observed in 74 individuals with normal VWF levels, 57 patients grouped as type 1 VWD, 33 type 2A, 16 type 2B, 28 type 2M, 11 type 2N. Patients tested following Desmopressin or VWF concentrate, with thrombotic thrombocytopenic purpura and acquired von Willebrand syndrome were also evaluated. Many of the discrepancies between methods were in patients with genetic mutations linked to more than one type of VWD including p.R1374C/H and p.R1315C. Quantifiable multimer results were available within one working day. Densitometry improved the interpretation of the multimers with slight structural variations that were not apparent by visual inspection of the in‐house method. Conclusions 5VWM was a rapid, sensitive, standardised assay which used existing technology and could be included as an initial screen of VWF multimers in a VWD diagnostic algorithm in conjunction with traditional multimer analysis

Математическое моделирование процессов тепло- и массопереноса, протекающих совместно, в условиях плавления больших массивов льда при движении криобота под действием силы тяжести. В работе варьировались параметры криобота, а также температура льда. По результатам численного моделирования было установлено значительное охлаждение поверхности криобота. Prevalence and psychopathological characteristics of depression in consecutive otorhinolaryngologic inpatients

<p>Abstract</p> <p>Background</p> <p>High prevalence of depression has been reported in otorhinolaryngologic patients (ORL). However, studies using a semi-structured interview to determine the prevalence of depression in ORL are lacking. Therefore the present study sought to determine the depression prevalence in ORL applying a semi-structured diagnostic interview and to further characterize the pathopsychological and demographic characteristics of depression in these patients.</p> <p>Methods</p> <p>One-hundred inpatients of the otorhinolaryngologic department of a German university hospital participated voluntarily (age M = 38.8 years, SD = 13.9; 38.0% female). Depression was assessed using a clinical interview in which the International Diagnostic Checklist for depression (IDCL) was applied. Patients completed the Brief Symptom Inventory (BSI) which constitutes three composite scores and nine symptom scales and the Beck Depression Inventory (BDI). Multivariate analyses of variance, correlations and effect sizes were conducted.</p> <p>Results</p> <p>A prevalence of depression of 21.0% was determined, 38.0% of the depressed patients were female. Depressed patients showed higher scores on the BSI-scales "interpersonal sensitivity", "depression", "anxiety", "phobic anxiety" and "psychoticism" with medium effect sizes.</p> <p>Conclusions</p> <p>High prevalence of depression was found which is in accordance with results of prior studies. Depressed patients showed higher psychological distress as compared to non-depressed patients. The results call for carrying on in engaging in depression research and routine depression screening in ORL.</p
