High-temperature and high-pressure behavior of carbonates in the ternary diagram CaCO3-MgCO3-FeCO3
We report the thermal expansion and the compressibility of carbonates in the ternary compositional diagram CaCO3-MgCO3-FeCO3, determined by in situ X-ray powder and single-crystal diffraction. High-temperature experiments were performed by high-resolution X-ray synchrotron powder diffraction from ambient to decarbonation temperatures (25-850 \ub0C). Single-crystal synchrotron X ray diffraction experiments were performed in a variable pressure range (0-100 GPa), depending on the stability field of the rhombohedral structure at ambient temperature, which is a function of the carbonate composition. The thermal expansion increases from calcite, CaCO3, \u3b10 = 4.10(7)
710-5 K-1, to magnesite, MgCO3, \u3b10 = 7.04(2)
710-5 K-1. In the magnesite-siderite (FeCO3) join, the thermal expansion decreases as iron content increases, with an experimental value of \u3b10 = 6.44(4)
710-5 K-1 for siderite. The compressibility in the ternary join is higher (i.e., lower bulk modulus) in calcite and Mg-calcite [K0 = 77(3) GPa for Ca0.91Mg0.06Fe0.03(CO3)] than in magnesite, K0 = 113(1) GPa, and siderite, K0 = 125(1) GPa. The analysis of thermal expansion and compressibility variation in calcite-magnesite and calcite-iron-magnesite joins clearly shows that the structural changes associated to the order-disorder transitions [i.e., R3c calcite-type structure vs. R3 CaMg(CO3)2 dolomite-type structure] do not affect significantly the thermal expansion and compressibility of carbonate. On the contrary, the chemical compositions of carbonates play a major role on their thermo-elastic properties. Finally, we use our P-V-T equation of state data to calculate the unit-cell volume of a natural ternary carbonate, and we compare the calculated volumes to experimental observations, measured in situ at elevated pressure and temperatures, using a multi-anvil device. The experimental and calculated data are in good agreement demonstrating that the equation of state here reported can describe the volume behavior with the accuracy needed, for example, for a direct chemical estimation of carbonates based on experimental unit-cell volume data of carbonates at high pressures and temperatures
Single cell analysis reveals satellite cell heterogeneity for proinflammatory chemokine expression
Background: The expression of proinflammatory signals at the site of muscle injury are essential for efficient tissue repair and their dysregulation can lead to inflammatory myopathies. Macrophages, neutrophils, and fibroadipogenic progenitor cells residing in the muscle are significant sources of proinflammatory cytokines and chemokines. However, the inducibility of the myogenic satellite cell population and their contribution to proinflammatory signaling is less understood.Methods: Mouse satellite cells were isolated and exposed to lipopolysaccharide (LPS) to mimic sterile skeletal muscle injury and changes in the expression of proinflammatory genes was examined by RT-qPCR and single cell RNA sequencing. Expression patterns were validated in skeletal muscle injured with cardiotoxin by RT-qPCR and immunofluorescence.Results: Satellite cells in culture were able to express Tnfa, Ccl2, and Il6, within 2Â h of treatment with LPS. Single cell RNA-Seq revealed seven cell clusters representing the continuum from activation to differentiation. LPS treatment led to a heterogeneous pattern of induction of C-C and C-X-C chemokines (e.g., Ccl2, Ccl5, and Cxcl0) and cytokines (e.g., Tgfb1, Bmp2, Il18, and Il33) associated with innate immune cell recruitment and satellite cell proliferation. One cell cluster was enriched for expression of the antiviral interferon pathway genes under control conditions and LPS treatment. Activation of this pathway in satellite cells was also detectable at the site of cardiotoxin induced muscle injury.Conclusion: These data demonstrate that satellite cells respond to inflammatory signals and secrete chemokines and cytokines. Further, we identified a previously unrecognized subset of satellite cells that may act as sensors for muscle infection or injury using the antiviral interferon pathway
Sensitivity and Specificity of Multiple Kato-Katz Thick Smears and a Circulating Cathodic Antigen Test for Schistosoma mansoni Diagnosis Pre- and Post-repeated-Praziquantel Treatment
Two Kato-Katz thick smears (Kato-Katzs) from a single stool are currently recommended for diagnosing Schistosoma mansoni infections to map areas for intervention. This âgold standardâ has low sensitivity at low infection intensities. The urine point-of-care circulating cathodic antigen test (POC-CCA) is potentially more sensitive but how accurately they detect S. mansoni after repeated praziquantel treatments, their suitability for measuring drug efficacy and their correlation with egg counts remain to be fully understood. We compared the accuracies of one to six Kato-Katzs and one POC-CCA for the diagnosis of S. mansoni in primary-school children who have received zero to ten praziquantel treatments. A nonsynonymous SNP within PCDH15 is associated with lipid traits in familial combined hyperlipidemia
Familial combined hyperlipidemia (FCHL) is a common lipid disorder characterized by the presence of multiple lipoprotein phenotypes that increase the risk of premature coronary heart disease. In a previous study, we identified an intragenic microsatellite marker within the protocadherin 15 (PCDH15) gene to be associated with high triglycerides (TGs) in Finnish dyslipidemic families. In this study we analyzed all four known nonsynonymous SNPs within PCDH15 in 1,268 individuals from Finnish and Dutch multigenerational families with FCHL. Association analyses of quantitative traits for SNPs were performed using the QTDT test. The nonsynonymous SNP rs10825269 resulted in a PÂ =Â 0.0006 for the quantitative TG trait. Additional evidence for association was observed with the same SNP for apolipoprotein B levels (apo-B) (PÂ =Â 0.0001) and total cholesterol (TC) levels (PÂ =Â 0.001). None of the other three SNPs tested showed a significant association with any lipid-related trait. We investigated the expression of PCDH15 in different human tissues and observed that PCDH15 is expressed in several tissues including liver and pancreas. In addition, we measured the plasma lipid levels in mice with loss-of-function mutations in Pcdh15 (Pcdh15av-Tg and Pcdh15av-3J) to investigate possible abnormalities in their lipid profile. We observed a significant difference in plasma TG and TC concentrations for the Pcdh15av-3J carriers when compared with the wild type (PÂ =Â 0.013 and PÂ =Â 0.044, respectively). Our study suggests that PCDH15 is associated with lipid abnormalities
Intrinsic phase separation in superconducting K0.8Fe1.6Se2 (Tc= 31.8 K) single crystals
Temperature dependent single-crystal x-ray diffraction (XRD) in transmission
mode probing the bulk of the newly discovered K0.8Fe1.6Se2 superconductor (Tc =
31.8 K) using synchrotron radiation is reported. A clear evidence of intrinsic
phase separation at 520 K between two competing phases, (i) a first majority
magnetic phase with a ThCr2Si2-type tetragonal lattice modulated by the iron
vacancy ordering and (ii) a minority non-magnetic phase having an in-plane
compressed lattice volume and a weak superstructure, is reported. The XRD peaks
due to the Fe vacancy ordering in the majority phase disappear by increasing
the temperature at 580 K, well above phase separation temperature confirming
the order-disorder phase transition. The intrinsic phase separation at 520K
between a competing first magnetic phase and a second non-magnetic phase in the
normal phase both having lattice superstructures (that imply different Fermi
surface topology reconstruction and charge density) is assigned to a
lattice-electronic instability of the K0.8Fe1.6Se2 system typical of a system
tuned at a Lifshitz critical point of an electronic topological transition that
gives a multi-gaps superconductor tuned a shape resonance.Comment: 10 pages, 4 figure
Criteria for the differentiation between young and old Onchocerca volvulus filariae
Drugs exist that show long-lasting inhibition of embryogenesis and microfilaria production or macrofilaricidal activity against Onchocerca volvulus. Therefore, the patients have to be followed-up for several years. Clinical drug trials have to be performed in areas with ongoing transmission to assess the efficacy on younger worms. In addition, future vaccine trials may also require demonstrating efficacy against establishment of new worms. For the evaluation of the efficacy, it is necessary to differentiate between older worms, which were exposed to the drug, and younger worms newly acquired after drug treatment or vaccination. Here, we describe criteria for the differentiation between young and old filariae based on histological studies of worms with a known age from travellers, or from children, or patients living in areas with interrupted transmission in Burkina Faso, Ghana or Uganda. Older worms were larger and presented degenerated tissues. Gomori's iron stain showed that the worms accumulated more iron with increasing age, first in the gut and later in other organs. Using an antibody against O. volvulus lysosomal aspartic protease, the gut of young worms was stained only weakly; whereas, it was stronger labelled in older worms, accompanied by additional staining of hypodermis and epithelia. Using morphological and immunohistological criteria, it was possible to differentiate young (1â3Â years old) from older females and to identify young males
Probing the angular and polarization reconstruction of the ARIANNA detector at the South Pole
The sources of ultra-high energy (UHE) cosmic rays, which can have energies
up to 10^20 eV, remain a mystery. UHE neutrinos may provide important clues to
understanding the nature of cosmic-ray sources. ARIANNA aims to detect UHE
neutrinos via radio (Askaryan) emission from particle showers when a neutrino
interacts with ice, which is an efficient method for neutrinos with energies
between 10^16 eV and 10^20 eV. The ARIANNA radio detectors are located in
Antarctic ice just beneath the surface. Neutrino observation requires that
radio pulses propagate to the antennas at the surface with minimum distortion
by the ice and firn medium. Using the residual hole from the South Pole Ice
Core Project, radio pulses were emitted from a transmitter located up to 1.7 km
below the snow surface. By measuring these signals with an ARIANNA surface
station, the angular and polarization reconstruction abilities are quantified,
which are required to measure the direction of the neutrino. After deconvolving
the raw signals for the detector response and attenuation from propagation
through the ice, the signal pulses show no significant distortion and agree
with a reference measurement of the emitter made in an anechoic chamber.
Furthermore, the signal pulses reveal no significant birefringence for our
tested geometry of mostly vertical ice propagation. The origin of the
transmitted radio pulse was measured with an angular resolution of 0.37 degrees
indicating that the neutrino direction can be determined with good precision if
the polarization of the radio-pulse can be well determined. In the present
study we obtained a resolution of the polarization vector of 2.7 degrees.
