    This paper deals with the photosensitizing activity of FLQs towards two different biosubstrates, membrane and DNA. Thein vitrophototoxic activity of these drugsvs.DNA presents peculiar features with respect to thatvs.membranes, probably due to a specific binding of the drugs to the double helix and to the operativeness of different photosensitization mechanisms with the two types of biosubstrates. A description of the UVA photochemistry and the photosensitizing properties of two significant examples in the FLQ family is reported. The investigated compounds are Enoxacin, 1-ethyl-6-fluoro-1,4-dihydro-4- oxo-7-[1-piperazinyl]-1,8-naphtyridine-3-carboxilic acid and Rufloxacin, 9-fluoro-2,3-dihydro-10-4´-methyl- 1´-piperazinyl-7-oxo-7H-pyrido[1,2,3−de]-1,4-benzothiazine-6-carboxylic acid

    Transient absorption study on Red Vermilion darkening in presence of chlorine ions and after UV exposure

    The application of no destructive techniques in the field of Cultural Heritage is becoming fundamental to understanding degradation phenomena. In this study, Transient Absorption (TA) spectroscopy was exploited to explain the process which causes the darkening of Red Vermilion, a famous pigment known also as cinnabar. The optical properties involved in the process are studied in pure HgS and chlorine doped HgS samples, before and after exposure to UV light (365 nm). The study was carried out with particular attention on the ground state bleaching signals, directly connected to the formation of intra-gap trap levels responsible for the pigment degradation. First derivative reflectance spectra reveal the presence of these defectivities, while the analysis of Tauc plots from Kubelka Munk function confirms the reduction of energy band gap due to UV exposure. With the help of Density Functional calculations, we simulated the role of S vacancies in producing a defective alpha-phase, the consequent reduction of the energy band gap and, finally, the progressive phase transformation to the cubic metacinnabar. Transient Absorption turns out to be an important tool of diagnosis about the conservation state of pigments applied in the field of Cultural Heritage

    MBSE Certification-Driven Design of a UAV MALE Configuration in the AGILE 4.0 Design Environment

    This paper presents a certification-driven design process for an Unmanned Medium-Altitude- Long-Endurance (UAV MALE) air vehicle, including on-board system design and placements, electro-magnetic compatibility analysis, and thermal risk assessments. In literature, the preliminary aircraft design phase is mainly driven by mission performances and structural integrity aspects. However, the inclusion of other disciplines, like on-board system design or electro-magnetic compatibility, or thermal analysis, can lead to more efficient and cost- effective solutions and becomes paramount for non-conventional configurations like unmanned vehicles or highly electrified platforms. In the EC-funded AGILE 4.0 project (2019-2022), the traditional scope of the preliminary aircraft design is extended by including domains that are usually considered only in later design phases, such as certification, production and maintenance. In this paper, the AGILE 4.0 design environment supports the definition and execution of a certification-driven design process of a UAV MALE configuration, using a Model-Based Systems Engineering (MBSE) approach

    Sam68 splicing regulation contributes to motor unit establishment in the postnatal skeletal muscle

    RNA-binding proteins orchestrate the composite life of RNA molecules and impact most physiological processes, thus underlying complex phenotypes. The RNA-binding protein Sam68 regulates differentiation processes by modulating splicing, polyadenylation, and stability of select transcripts. Herein, we found that Sam68-/- mice display altered regulation of alternative splicing in the spinal cord of key target genes involved in synaptic functions. Analysis of the motor units revealed that Sam68 ablation impairs the establishment of neuromuscular junctions and causes progressive loss of motor neurons in the spinal cord. Importantly, alterations of neuromuscular junction morphology and properties in Sam68-/- mice correlate with defects in muscle and motor unit integrity. Sam68-/- muscles display defects in postnatal development, with manifest signs of atrophy. Furthermore, fast-twitch muscles in Sam68-/- mice show structural features typical of slow-twitch muscles, suggesting alterations in the metabolic and functional properties of myofibers. Collectively, our data identify a key role for Sam68 in muscle development and suggest that proper establishment of motor units requires timely expression of synaptic splice variants

    Patient-reported outcome measures of the impact of cancer on patient’s everyday lives: a systematic review

    Purpose: Patients with advanced disease are living longer and commonly used patient-reported outcome measures (PROMs) may miss relevant elements of the quality of extended survival. This systematic review examines the measures used to capture aspects of the quality of survival including impact on patients’ everyday lives such as finances, work and family roles. Methods: Searches were conducted in MEDLINE, EMBASE, CINAHL and PsycINFO restricted to English language articles. Information on study characteristics, instruments and outcomes was systematically extracted and synthesised. A predefined set of criteria was used to rate the quality of studies. Results: From 2761 potentially relevant articles, 22 met all inclusion criteria, including 10 concerning financial distress, 3 on roles and responsibilities and 9 on multiple aspects of social well-being. Generally, studies were not of high quality; many lacked bias free participant selection, had confounding factors and had not accounted for all participants. High levels of financial distress were reported and were associated with multiple demographic factors such as age and income. There were few reports concerned with impacts on patients’ roles/responsibilities in everyday life although practical and emotional struggles with parenting were identified. Social difficulties were common and associated with multiple factors including being a caregiver. Many studies were single time-point surveys and used non-validated measures. Exceptions were employment of the COST and Social Difficulties Inventory (SDI), validated measures of financial and social distress respectively. Conclusions: Impact on some important parts of patients’ everyday lives is insufficiently and inconsistently captured. Further PROM development focussing on roles and responsibilities, including work and caring for dependents, is warranted. Implications for Cancer Survivors: Factors such as finances, employment and responsibility for caring for dependents (e.g. children and elderly relatives) can affect the well-being of cancer survivors. There is a need to ensure that any instruments used to assess patients’ social well-being are broad enough to include these areas so that any difficulties arising can be better understood and appropriately supported

    "Delirium Day": A nationwide point prevalence study of delirium in older hospitalized patients using an easy standardized diagnostic tool

    Background: To date, delirium prevalence in adult acute hospital populations has been estimated generally from pooled findings of single-center studies and/or among specific patient populations. Furthermore, the number of participants in these studies has not exceeded a few hundred. To overcome these limitations, we have determined, in a multicenter study, the prevalence of delirium over a single day among a large population of patients admitted to acute and rehabilitation hospital wards in Italy. Methods: This is a point prevalence study (called "Delirium Day") including 1867 older patients (aged 65 years or more) across 108 acute and 12 rehabilitation wards in Italian hospitals. Delirium was assessed on the same day in all patients using the 4AT, a validated and briefly administered tool which does not require training. We also collected data regarding motoric subtypes of delirium, functional and nutritional status, dementia, comorbidity, medications, feeding tubes, peripheral venous and urinary catheters, and physical restraints. Results: The mean sample age was 82.0 \ub1 7.5 years (58 % female). Overall, 429 patients (22.9 %) had delirium. Hypoactive was the commonest subtype (132/344 patients, 38.5 %), followed by mixed, hyperactive, and nonmotoric delirium. The prevalence was highest in Neurology (28.5 %) and Geriatrics (24.7 %), lowest in Rehabilitation (14.0 %), and intermediate in Orthopedic (20.6 %) and Internal Medicine wards (21.4 %). In a multivariable logistic regression, age (odds ratio [OR] 1.03, 95 % confidence interval [CI] 1.01-1.05), Activities of Daily Living dependence (OR 1.19, 95 % CI 1.12-1.27), dementia (OR 3.25, 95 % CI 2.41-4.38), malnutrition (OR 2.01, 95 % CI 1.29-3.14), and use of antipsychotics (OR 2.03, 95 % CI 1.45-2.82), feeding tubes (OR 2.51, 95 % CI 1.11-5.66), peripheral venous catheters (OR 1.41, 95 % CI 1.06-1.87), urinary catheters (OR 1.73, 95 % CI 1.30-2.29), and physical restraints (OR 1.84, 95 % CI 1.40-2.40) were associated with delirium. Admission to Neurology wards was also associated with delirium (OR 2.00, 95 % CI 1.29-3.14), while admission to other settings was not. Conclusions: Delirium occurred in more than one out of five patients in acute and rehabilitation hospital wards. Prevalence was highest in Neurology and lowest in Rehabilitation divisions. The "Delirium Day" project might become a useful method to assess delirium across hospital settings and a benchmarking platform for future surveys

    Prenatal Hypoxic-Ischemic Insult Changes the Distribution and Number of NADPH-Diaphorase Cells in the Cerebellum

    Astrogliosis, oligodendroglial death and motor deficits have been observed in the offspring of female rats that had their uterine arteries clamped at the 18th gestational day. Since nitric oxide has important roles in several inflammatory and developmental events, here we evaluated NADPH-diaphorase (NADPH-d) distribution in the cerebellum of rats submitted to this hypoxia-ischemia (HI) model. At postnatal (P) day 9, Purkinje cells of SHAM and non-manipulated (NM) animals showed NADPH-d+ labeling both in the cell body and dendritic arborization in folia 1 to 8, while HI animals presented a weaker labeling in both cellular structures. NADPH-d+ labeling in the molecular (ML), and in both the external and internal granular layer, was unaffected by HI at this age. At P23, labeling in Purkinje cells was absent in all three groups. Ectopic NADPH-d+ cells in the ML of folia 1 to 4 and folium 10 were present exclusively in HI animals. This labeling pattern was maintained up to P90 in folium 10. In the cerebellar white matter (WM), at P9 and P23, microglial (ED1+) NADPH-d+ cells, were observed in all groups. At P23, only HI animals presented NADPH-d labeling in the cell body and processes of reactive astrocytes (GFAP+). At P9 and P23, the number of NADPH-d+ cells in the WM was higher in HI animals than in SHAM and NM ones. At P45 and at P90 no NADPH-d+ cells were observed in the WM of the three groups. Our results indicate that HI insults lead to long-lasting alterations in nitric oxide synthase expression in the cerebellum. Such alterations in cerebellar differentiation might explain, at least in part, the motor deficits that are commonly observed in this model
