    Charge Photoinjection in Intercalated and Covalently Bound [Re(CO)_(3)(dppz)(py)]^(+)–DNA Constructs Monitored by Time-Resolved Visible and Infrared Spectroscopy

    The complex [Re(CO)_(3)(dppz)(py′-OR)]+ (dppz = dipyrido[3,2-a:2′,3′-c]phenazine; py′-OR = 4-functionalized pyridine) offers IR sensitivity and can oxidize DNA directly from the excited state, making it a promising probe for the study of DNA-mediated charge transport (CT). The behavior of several covalent and noncovalent Re–DNA constructs was monitored by time-resolved IR (TRIR) and UV/visible spectroscopies, as well as biochemical methods, confirming the long-range oxidation of DNA by the excited complex. Optical excitation of the complex leads to population of MLCT and at least two distinct intraligand states. Experimental observations that are consistent with charge injection from these excited states include similarity between long-time TRIR spectra and the reduced state spectrum observed by spectroelectrochemistry, the appearance of a guanine radical signal in TRIR spectra, and the eventual formation of permanent guanine oxidation products. The majority of reactivity occurs on the ultrafast time scale, although processes dependent on slower conformational motions of DNA, such as the accumulation of oxidative damage at guanine, are also observed. The ability to measure events on such disparate time scales, its superior selectivity in comparison to other spectroscopic techniques, and the ability to simultaneously monitor carbonyl ligand and DNA IR absorption bands make TRIR a valuable tool for the study of CT in DNA

    Ultrafast Excited-State Dynamics of Rhenium(I) Photosensitizers [Re(Cl)(CO)_(3)(N,N)] and [Re(imidazole)(CO)_(3)(N,N)]^+: Diimine Effects

    Femto- to picosecond excited-state dynamics of the complexes [Re(L)(CO)_(3)(N,N)]^n (N,N = bpy, phen, 4,7-dimethyl-phen (dmp); L = Cl, n = 0; L = imidazole, n = 1+) were investigated using fluorescence up-conversion, transient absorption in the 650−285 nm range (using broad-band UV probe pulses around 300 nm) and picosecond time-resolved IR (TRIR) spectroscopy in the region of CO stretching vibrations. Optically populated singlet charge-transfer (CT) state(s) undergo femtosecond intersystem crossing to at least two hot triplet states with a rate that is faster in Cl (~100 fs)^(−1) than in imidazole (~150 fs)^(−1) complexes but essentially independent of the N,N ligand. TRIR spectra indicate the presence of two long-lived triplet states that are populated simultaneously and equilibrate in a few picoseconds. The minor state accounts for less than 20% of the relaxed excited population. UV−vis transient spectra were assigned using open-shell time-dependent density functional theory calculations on the lowest triplet CT state. Visible excited-state absorption originates mostly from mixed L;N,N^(•−) → Re^(II) ligand-to-metal CT transitions. Excited bpy complexes show the characteristic sharp near-UV band (Cl, 373 nm; imH, 365 nm) due to two predominantly ππ*(bpy^(•−)) transitions. For phen and dmp, the UV excited-state absorption occurs at 305 nm, originating from a series of mixed ππ* and Re → CO;N,N•− MLCT transitions. UV−vis transient absorption features exhibit small intensity- and band-shape changes occurring with several lifetimes in the 1−5 ps range, while TRIR bands show small intensity changes (≤5 ps) and shifts (~1 and 6−10 ps) to higher wavenumbers. These spectral changes are attributable to convoluted electronic and vibrational relaxation steps and equilibration between the two lowest triplets. Still slower changes (≥15 ps), manifested mostly by the excited-state UV band, probably involve local-solvent restructuring. Implications of the observed excited-state behavior for the development and use of Re-based sensitizers and probes are discussed

    Relación de la calidad de vida profesional y el burnout en médicos de atención primaria

    ObjetivoConocer la relación, si es que la hay, entre la calidad de vida profesional (CVP) y burnout de facultativos de atención primaria.DiseñoEstudio transversal mediante cuestionario CVP-35 y MBI (Maslach Burnout Inventory), por correo postal.ÁmbitoTres centros urbanos y 3 zonas de atención rural. Participó un total de 131 profesionalesMedicionesDel CVP-35 se obtienen puntuaciones que van de 0 (mínimo) a 10 (máximo). El cuestionario Maslach clasifica en nivel alto, medio y bajo de cada dimensión.ResultadosContestó a la encuesta el 71,7%. La edad media era de 44 años y la media de tarjetas sanitarias, 1.339. El número de consultas/día fue de 37; en cuanto las visitas domiciliarias, realizaban una diaria. El valor medio de la CVP fue de 4,48 (intervalo de confianza [IC] del 95%, 4,08-4,88). La percepción de la demanda tuvo una puntuación de 5,9 (IC del 95%, 5,6-6,1); la motivación intrínseca del profesional se situó en 6,5 (IC del 95%, 6,2-6,7) y el apoyo de los directivos en 3,8 (IC del 95%, 3,6-4,1). En cuanto al cuestionario MBI, se observó una despersonalización alta en el 54,3%, cansancio emocional alto en un 56,2% y realización personal alta en el 9,3%. La correlación mayor fue entre las demandas en el puesto y el cansancio emocional (r=0,6; p<0,0001), la motivación intrínseca y la realización personal (r=0,46; p<0,0001) y la calidad de vida profesional frente al cansancio emocional (r=−0,4; p<0,0005).ConclusionesLa CVP y el burnout miden dimensiones diferentes pero relacionadas. Por tanto, su utilidad es diferente en función de los objetivos. MBI: high depersonalization 54.3%, emotional exhaustion (r=0.6; P<.0001), intrinsic motivation, and personal accomplishment (r=0.46; P<.0001) and profesional quality of life front emotional fatigue (r= −0.4; P<.0005).ConclusionsCPL and burnout measure different but related dimensions.     Experiments on a lab scale TES unit using eutectic metal alloy as PCM

    The behavior of a magnesium and zinc eutectic metal alloy used as thermal energy storage (TES) material is tested in a laboratory scale TES unit. The TES unit consists of two concentric tubes with the central tube surrounded by 67 kg of the metal alloy and two caps at both ends of the tube through which the heat transfer fluid (HTF) flows. Charging (melting) and discharging (solidification) processes of the eutectic metal alloy are performed using synthetic oil as the HTF. The experimental results are used to test the validity of the model via simulations performed with a computational fluid dynamics tool.     Effects of dry needling on gait and muscle tone in Parkinson''s disease: a randomized clinical trial

    Background: Alterations in gait and muscular rigidity are common and disabling in persons with Parkinson''s disease (PD). Objective: The aim of this study was to determine whether a single dry needling (DN) session can promote changes in gait and muscle tone in the lower extremities as well as in the evolution of the disease in persons with PD. Methods: A randomized double-blind clinical trial was designed. Participants were randomly assigned to an intervention group (IG) that received a session of DN over the semitendinosus, medial gastrocnemius, soleus and rectus femoris muscles, or to a control group (CG) that received a session of sham DN in the same muscles. The effects of DN were assessed using the timed up and go test (TUG), 10 meter walk test (10MWT), 6 minute walk test (6MWT) and myotonometry before, immediately after, and 7 days after the intervention. Results: Thirty-three participants were analyzed aged 69.9 +/- 7.2 years (mean +/- SD; 39% female). There were no significant differences between the IG and CG for any outcomes. Significant differences were observed when comparing the Pre and Follow-up values in the IG for functional mobility of gait in the TUG (p = 0.049), gait speed in the 10MWT (p = 0.041) and muscle tone in the lower extremities by myotonometry (frequency (p = 0.027) and stiffness (p = 0.013)). By comparison, there were no significant within-group differences in the CG. Conclusion: A single session of DN had no measurable benefit compared to a single session of sham DN. Within-group changes in the IG suggested improvements in functional mobility of gait and gait speed, as well as changes in the muscle tone in the lower extremities of PD patients, which could be worthy of further exploration by future research

    Surface mesoscale pico–nanoplankton patterns at the main fronts of the Alboran Sea

    © 2014 Elsevier B.V. The mesoscale (10-100. km, days-weeks) plays a key role in the Ocean's ecosystem structure and dynamics. This work compares the pico-nanoplankton patterns observed in the Alboran Sea (Western Mediterranean) during three oceanographic cruises. We analyse its response to different expressions of mesoscale circulation associated with the three major hydrodynamic features in the basin; namely the Northwestern Alboran Front (NWAF, surveyed in OMEGA-1 cruise), the Almeria-Oran Front (AOF, surveyed in OMEGA-2 cruise) and the Western Alboran Gyre (WAG, surveyed in BIOMEGA cruise). The first two surveys were carried out under the most typical quasi-stationary twin gyre conditions of the Alboran Sea, whereas the third cruise was performed after an eastward migration of the WAG. The analysis of pico and nanoplankton populations was carried out using flow cytometry. The patchiness observed in the three cruises indicates an association of phytoplankton peaks with the main frontal structures: abundances were higher in the NWAF/upwelling area in OMEGA-1, at the Mediterranean side of the AOF in OMEGA-2, and at a tongue of recent Atlantic Water west of the WAG in BIOMEGA. However, a more detailed analysis reveals that different factors explain the origin of the phytoplankton biomass in each front/cruise. Mixing processes at the Strait of Gibraltar and the subsequent advection of water properties into the Western Alboran Sea were the mechanisms responsible for the abundances observed in the NWAF. The highest concentrations observed in the AOF were related to the intrusion of Mediterranean Surface Waters to the north of the front. During the migrating WAG the phytoplankton distribution was influenced by the formation of a new gyre. The relation between phytoplankton and mesoscale dynamics is further explored in terms of vertical velocity diagnosis. In all cases, intense vertical motion is negatively correlated with the abundance of phytoplankton populations. This resulted from the intense geostrophic background flow associated with large vertical velocities, which drove low residence times of water mass properties and hence a weak biological response. Fast-repeated surveys made during the OMEGA-1 and OMEGA-2 cruises reveal that the surveyed hydrographic features are subjected to significant temporal variability.     Efficiency of enzymes and benzalkonium chloride treatments against Listeria monocytogenes dual-species blofllms determined by fluorescence mlcroscopy and image analysis

    1 poster presented at the 24th International ICFMH Conference, Food Micro 2014, 1-4 Septembre, Nantes, FranceIn the present study, three different mixed·species Listeria monocytogenes-carrying blofilms previously isolated from surfaces of seafood, meat and dairies Industrial environments were selected in order to test the efficiency of cleanlng and disinfection procedures based on the combination of an enzymatic solution, followed by a treatment with benzalkonium chloride (BAC). Biofilm samples were cultured onto AISI-316 type stainless steel 1 cm2 coupons In a batch system at 25° C. The effect of sequential exposure to different concentrations of Pronase- BAC, Cellulase - BAC or DNAsel - BAC solutions was then assessed in mature biofilms after 168 hours using an orthogonal first order experimental design. Samples were stained using FilmTracerTM LIVE/DEAD Blofllm viability kit (lnvitrogen} and the efficiency of each treatment was then determined by epifluorescence mlcroscopy and subsequent image anatysls of 25·field mosaics representing a total surface of 1.92 mm2, so as to quantify the total area occupied by living and damaged cells attached to the surface. Significant empirical equations obtained reveals that synergistic effects between Pronase - BAC on biofilms coming from flsh and dairy industries obtaining almost a total cell detachment in the latter case. No significant effect was observed among the Cellulase - BAC procedures in the experimental DNAse - BAC against biofilms coming from fish and dairy industries compared with the other two combinations enzyme- benzalkoniom chloride was also demonstrated even with no effect against the consortium isolated from meat industry These results show how the usage of customised treatments combining commerclally available enzymes with classical chemlcally-based disinfection procedures may represent an attractive and efficient alternative against bacterial blofilms present in the food IndustryThis research was financially supported by the Spanish Ministerio de Economía y Competitividad (ENZYMONO, AGL2010-22212-C02-12).
