    Modulation of the glutamate-evoked release of arachidonic acid from mouse cortical neurons: involvement of a pH-sensitive membrane phospholipase A2

    Excitatory synaptic transmission is associated with changes in both extracellular and intracellular pH. Using mouse cortical neurons in primary cultures, we studied the sensitivity of glutamate-evoked release of 3H-arachidonic acid (3H-AA) to changes in extracellular pH (pHo) and related intracellular pH (pHi). As pHo was shifted from 7.2 to 7.8, the glutamate-evoked release of 3H-AA was enhanced by approximately threefold. The effect of alkaline pHo on the glutamate response was rapid, becoming significant within 2 min. 3H-AA release, evoked by both NMDA and kainate, was also enhanced by pHo alkalinization. NMDA- and kainate-induced increase in free intracellular Ca2+ was unaffected by changing pHo from 7.2 to 7.8, indicating that the receptor-induced Ca2+ influx is not responsible for the pHo sensitivity of the glutamate-evoked release of 3H-AA. Alkalinization of pHi obtained by incubating neurons in the presence of HCO3- or NH4 enhanced the glutamate-evoked release of 3H-AA, while pHi acidification obtained by blockade of Na+/H+ and Cl-/HCO3- exchangers decreased the glutamate response. Membrane-bound phospholipase A2 (mPLA2) activity was stimulated by Ca2+ in a pH-dependent manner, increasing its activity as pH was shifted from 7.2 to 7.8. This pH profile corresponds to the pH profile of the glutamate-, NMDA- and kainate-evoked release of 3H-AA. Taken together, these results indicate that the glutamate-evoked release of 3H-AA may be mediated by the pH-sensitive mPLA2. Since excitatory neurotransmission mediated by glutamate results in both pHo and pHi changes and since AA enhances glutamatergic neurotransmission at both pre- and postsynaptic levels, the data reported here reveals a possible molecular mechanism whereby glutamate can modulate its own signalling efficacy in a pH-dependent manner by regulating the release of AA

    Solid State Nuclear Magnetic Resonance (NMR) experiments allow characterizing the local structure and dynamics of oxide glasses and melts. Thanks to the development of new experiments, it now becomes possible to evidence not only the details of the coordination state of the network formers of glasses but also to characterize the nature of polyatomic molecular motifs extending over several chemical bonds.     Physiological and clinical consequences of relief of right ventricular outflow tract obstruction late after repair of congenital heart defects.

    BACKGROUND: Right ventricular outflow tract obstruction (RVOTO) is a common problem after repair of congenital heart disease. Percutaneous pulmonary valve implantation (PPVI) can treat this condition without consequent pulmonary regurgitation or cardiopulmonary bypass. Our aim was to investigate the clinical and physiological response to relieving RVOTO. METHODS AND RESULTS: We studied 18 patients who underwent PPVI for RVOTO (72% male, median age 20 years) from a total of 93 who had this procedure for various indications. All had a right ventricular outflow tract (RVOT) gradient >50 mm Hg on echocardiography without important pulmonary regurgitation (less than mild or regurgitant fraction <10% on magnetic resonance imaging [MRI]). Cardiopulmonary exercise testing, tissue Doppler echocardiography, and MRI were performed before and within 50 days of PPVI. PPVI reduced RVOT gradient (51.4 to 21.7 mm Hg, P<0.001) and right ventricular systolic pressure (72.8 to 47.3 mm Hg, P<0.001) at catheterization. Symptoms and aerobic (25.7 to 28.9 mL.kg(-1).min(-1), P=0.002) and anaerobic (14.4 to 16.2 mL.kg(-1).min(-1), P=0.002) exercise capacity improved. Myocardial systolic velocity improved acutely (tricuspid 4.8 to 5.3 cm/s, P=0.05; mitral 4.7 to 5.5 cm/s, P=0.01), whereas isovolumic acceleration was unchanged. The tricuspid annular velocity was not maintained on intermediate follow-up. Right ventricular end-diastolic volume (99.9 to 89.7 mL/m2, P<0.001) fell, whereas effective stroke volume (43.7 to 48.3 mL/m2, P=0.06) and ejection fraction (48.0% to 56.8%, P=0.01) increased. Left ventricular end-diastolic volume (72.5 to 77.4 mL/m2, P=0.145), stroke volume (45.3 to 50.6 mL/m2, P=0.02), and ejection fraction (62.6% to 65.8%, P=0.03) increased. CONCLUSIONS: PPVI relieves RVOTO, which leads to an early improvement in biventricular performance. Furthermore, it reduces symptoms and improves exercise tolerance. These findings have important implications for the management of this increasingly common condition

    L'organisation tridimensionnelle d'une couverture pédologique, comprenant l'étude des relations géométriques entre horizons, et particulièrement des relations de concordance ou de discordance, permet de reconstituer certaines étapes de l'évolution du modelé. Le versant étudié, situé en zone tropicale humide (Brésil atlantique), présente à l'amont une couverture ferrallitique très épaisse (> 10 m) constituée d'horizons à limites subhorizontales. La partie supérieure de cette couverture est recoupée par une différenciation pédologique moins développée. Cette dernière est concordante à la topographie actuelle et s'est développée le long du versant à partir des horizons successifs de la couverture ferrallitique jusqu'au substratum. Lorsque ce substratum est atteint, des sols lithodépendants se forment à partir des grès, siltites et argilites de la "formation Tubarao" (permo-carbonifère). Cette discordance entre la différenciation superficielle et les horizons de la couverture ferrallitique implique que la première est plus récente que la seconde. Plus à l'aval, une couverture épaisse réapparaît qui a pu être raccordée à celle de l'amont par l'étude selon le plan horizontal. En considérant que la géométrie de la couverture ferrallitique (limites entre horizons et front de structuration pédologique) est conforme à la surface topographique contemporaine de sa formation, il est possible de reconstituer cette dernière. La topographie actuelle apparaît alors comme partiellement incisée dans cette topographie ancienne. (Résumé d'auteur

    Percutaneous pulmonary valve implantation in humans - Results in 59 consecutive patients

    Background - Right ventricular outflow tract (RVOT) reconstruction with valved conduits in infancy and childhood leads to reintervention for pulmonary regurgitation and stenosis in later life.Methods and Results - Patients with pulmonary regurgitation with or without stenosis after repair of congenital heart disease had percutaneous pulmonary valve implantation (PPVI). Mortality, hemodynamic improvement, freedom from explantation, and subjective and objective changes in exercise tolerance were end points. PPVI was performed successfully in 58 patients, 32 male, with a median age of 16 years and median weight of 56 kg. The majority had a variant of tetralogy of Fallot (n = 36), or transposition of the great arteries, ventricular septal defect with pulmonary stenosis (n = 8). The right ventricular (RV) pressure (64.4 +/- 17.2 to 50.4 +/- 14 mm Hg, P < 0.001), RVOT gradient (33 +/- 24.6 to 19.5 +/- 15.3, P < 0.001), and pulmonary regurgitation ( PR) (grade 2 of greater before, none greater than grade 2 after, P < 0.001) decreased significantly after PPVI. MRI showed significant reduction in PR fraction (21 +/- 13% versus 3 +/- 4%, P < 0.001) and in RV end-diastolic volume (EDV) (94 +/- 28 versus 82 +/- 24 mL (.) beat(-1) (.) m(-2), P < 0.001) and a significant increase in left ventricular EDV ( 64 +/- 12 versus 71 +/- 13 mL (.) beat(-1.) m(-2), P = 0.005) and effective RV stroke volume ( 37 +/- 7 versus 42 +/- 9 mL (.) beat(-1) (.) m(-2), P = 0.006) in 28 patients (age 19 +/- 8 years). A further 16 subjects, on metabolic exercise testing, showed significant improvement in V(O2)max (26 +/- 7 versus 29 +/- 6 mL (.) kg(-1) (.) min(-1), P < 0.001). There was no mortality.Conclusions - PPVI is feasible at low risk, with quantifiable improvement in MRI-defined ventricular parameters and pulmonary regurgitation, and results in subjective and objective improvement in exercise capacity

    Wavefunction tomography of topological dimer chains with long-range couplings

    The ability to tailor with a high accuracy the inter-site connectivity in a lattice is a crucial tool for realizing novel topological phases of matter. Here, we report the experimental realization of photonic dimer chains with long-range hopping terms of arbitrary strength and phase, providing a rich generalization of the celebrated Su-Schrieffer-Heeger model. Our experiment is based on a synthetic dimension scheme involving the frequency modes of an optical fiber loop platform. This setup provides direct access to both the band dispersion and the geometry of the Bloch wavefunctions throughout the entire Brillouin zone allowing us to extract the winding number for any possible configuration. Finally, we highlight a topological phase transition solely driven by a time-reversal-breaking synthetic gauge field associated with the phase of the long-range hopping, providing a route for engineering topological bands in photonic lattices belonging to the AIII symmetry class

    A compositional breakage equation for wheat milling

    The compositional breakage equation is derived, in which the distributions of botanical components following milling of wheat are defined in terms of compositional breakage functions and concentration functions. The forms of the underlying functions are determined using experimental data for Outer Pericarp, Intermediate Layer, Aleurone and Starchy Endosperm generated from spectroscopic analysis of milled fractions of a hard and a soft wheat milled under Sharp-to-Sharp (S-S) and Dull-to-Dull (D-D) dispositions. For the hard Mallacca wheat, the Outer Pericarp, Intermediate Layer and Aleurone compositions mostly varied with particle size in similar ways, consistent with these layers fusing together as “bran” and breaking together, although with possibly a subtle difference around the production of very fine particles under D-D milling. By contrast, for the soft Consort wheat, Outer Pericarp, Intermediate Layer and Aleurone were distributed in broken particles very differently, particularly under D-D milling, suggesting a different breakage mechanism associated with differences in the mechanical properties and adhesion of the bran layers. These new insights into the nature of wheat breakage and the contributions of the component tissues could have implications for wheat breeding and flour mill operation

    On the distribution of initial masses of stellar clusters inferred from synthesis models

    The fundamental properties of stellar clusters, such as the age or the total initial mass in stars, are often inferred from population synthesis models. The predicted properties are then used to constrain the physical mechanisms involved in the formation of such clusters in a variety of environments. Population synthesis models cannot, however, be applied blindy to such systems. We show that synthesis models cannot be used in the usual straightforward way to small-mass clusters (say, M < few times 10**4 Mo). The reason is that the basic hypothesis underlying population synthesis (a fixed proportionality between the number of stars in the different evolutionary phases) is not fulfilled in these clusters due to their small number of stars. This incomplete sampling of the stellar mass function results in a non-gaussian distribution of the mass-luminosity ratio for clusters that share the same evolutionary conditions (age, metallicity and initial stellar mass distribution function). We review some tests that can be carried out a priori to check whether a given cluster can be analysed with the fully-sampled standard population synthesis models, or, on the contrary, a probabilistic framework must be used.
