    Botanicals in Functional Foods and Food Supplements: Tradition, Efficacy and Regulatory Aspects

    In recent decades, the interest in products containing botanicals and claiming \u201cfunctional\u201d properties has increased exponentially. Functional foods, novel foods and food supplements have a special impact on the consumers, who show significant expectation for their well-being. Food supplements with botanical ingredients are the food area that has witnessed the greatest development, in terms of the number of available products, budget, and consumer acceptability. This review refers to and discusses some open points, such as: 1) the definitions and regulation of products containing botanicals; 2) the difficulty in obtaining nutritional and functional claims (botanical ingredients obtaining claims in the EU are listed and summarized); 3) the safety aspects of these products; and 4) the poor harmonization between international legislations. The availability of these "new" products can positively influence the well-being of the population, but it is essential to provide the consumers with the necessary recommendations to guide them in their purchase and use

    K+ accumulation and clearance in the calyx synaptic cleft of type I mouse vestibular hair cells

    Vestibular organs of Amniotes contain two types of sensory cells, named Type I and Type II hair cells. While Type II hair cells are contacted by several small bouton nerve terminals, Type I hair cells receive a giant terminal, called a calyx, which encloses their basolateral membrane almost completely. Both hair cell types release glutamate, which depolarizes the afferent terminal by binding to AMPA post-synaptic receptors. However, there is evidence that non-vesicular signal transmission also occurs at the Type I hair cell-calyx synapse, possibly involving direct depolarization of the calyx by K+ exiting the hair cell. To better investigate this aspect, we performed whole-cell patch-clamp recordings from mouse Type I hair cells or their associated calyx. We found that [K+] in the calyceal synaptic cleft is elevated at rest relative to the interstitial (extracellular) solution and can increase or decrease during hair cell depolarization or repolarization, respectively. The change in [K+] was primarily driven by GK,L, the low-voltage-activated, non-inactivating K+ conductance specifically expressed by Type I hair cells. Simple diffusion of K+ between the cleft and the extracellular compartment appeared substantially restricted by the calyx inner membrane, with the ion channels and active transporters playing a crucial role in regulating intercellular [K+]. Calyx recordings were consistent with K+ leaving the synaptic cleft through postsynaptic voltage-gated K+ channels involving KV1 and KV7 subunits. The above scenario is consistent with direct depolarization and hyperpolarization of the calyx membrane potential by intercellular K+

    Phenolic profile and biological activity of table grapes (Vitis vinifera L.)

    Table grapes are largely produced and consumed in the Mediterranean area. Furthermore, in the last years, the commercial interest in table grapes and other non-alcoholic grape products has notably increased worldwide. In addition to the nutritional aspects, polyphenol compounds in fresh grapes could exert positive effects on human health. The aim of this study was the characterization of the phenolic pattern of sixteen grape varieties and the evaluation of the associated antioxidant capacity and anti-inflammatory activity at gastric level. The methods used were: 1) Folin-Cocalteau\u2019s assay for the quantification of total polyphenol content; 2) High-Performance Liquid Chromatography (HPLC) coupled with Diode-Array Detector (DAD) to perform the quantitative analysis of grapes phenolic fraction 3) DPPH (1,1,-diphenil-2-picrylhydrazyl) spectrophotometric assay for the assessment of radical scavenging activity; 4) assessment of IL-8 release from human gastric epithelial cells to evaluate the anti-inflammatory activity of grape extracts. Some grapes, including seedless varieties, showed that the phenolic pattern was highly correlated with the biological activities, and, in particular, with peel and seeds portion. These results suggest that selected grape varieties could represent, also for consumers who do not drink wine, a source of healthy compounds potentially able to counteract oxidative stress and gastric inflammation

    The Role of Wine in Modulating Inflammatory Processes: A Review

    Several epidemiological studies associated the consumption of wine with the reduction of the risk factors for cardiovascular disease and certain cancers, as well as for diabetes. These conditions are characterized by inflammatory mechanisms in addition to other biological mechanisms. Acute and chronic inflammation is mediated by a plethora of biomarkers production and pathway activation. Since the health promoting properties of wine in different pathological conditions may include the reduction of inflammation, the aim of this paper was to collect and review the in vitro, in vivo, and human studies performed to evaluate the effects of wine on different models of inflammation. Although great variability in wine intake, period of consumption, and content of phenolic compounds was observed, data from both human and animal studies showed a positive modulation of inflammatory biomarkers (cytokines, coagulation parameters) and oxidative stress (mainly malondialdehyde) involved in cardiovascular function. In addition, some convincing evidence was obtained in different models suggesting a positive modulation of risk factors for gastric and intestinal inflammation. Contradictory results were obtained for metabolic syndrome and type 2 diabetes. To date, no significant paper has been published in the area of immune function.     2D seismic tomography of Somma-Vesuvius: Description of the experiment and preliminary results

    A multidisciplinary project for the investigation of Mt. Vesuvius structure was started in 1993. The core of the project is represented by a high resolution seismic tomography study by using controlled and natural sources. The main research objective is to investigate the feeding system of the volcano and to retrieve details of the upper crustal structure in the area. A first 2D active seismic experiment was performed in May 1994, with the aim of studing the feasibility of using tomographic techniques for exploring the volcano interiors. Particularly, this experiment was designed to obtain information on the optimal sources-receivers configuration and on the depth extension of the volume sampled by shot-generated seismic waves. 66 three-component seismic stations and 16 single-component analogue instruments were installed by several Italian and French groups to record signals generated by three on-land, underground explosions. Sources and geophones were deployed along a 30-km NW-SE profile passing through the volcano crater. Receivers were placed at an average spacing of 250 m in the middle of the recording line and at 500 m outside. The arrival time data base was complemented by first P and S readings of microearthquakes which occurred in the recent past within the volcano. The first arrival data set was preliminarily used to determine the shallow structure of the volcano by applying Thurber's (1983) tomographic inversion technique. This analysis shows evidence for a high-velocity body which extends vertically from about 400 m below the crater down to at least 3000 m and for a shallow 300-500 m thick low-velocity cover which borders the edifice. Data from the distant shot show evidence for arrivals of deep reflected/converted phases and provide information on the deeper structure under the volcano. The results from the interpretation of 2D data are used for planning a 3D tomographic survey which will be carried out in 1996

    Further experimental data supporting the safety of potassium polyaspartate used as a food additive in wine stabilization

    Potassium polyaspartate (KPPA) is a food additive used for wine stabilization. KPAA was included in the list of additives allowed in EU, with the Commission Regulation (EU) 2017/1399, having received a positive opinion by EFSA (European Food Safety Authority) in 2016. KPAA is rich in negative charges, which are essential for its enological function consisting in binding positive ions in order to reduce wine instability. Concerns were raised on the fact that the same binding properties could be responsible for a reduction of microelement bioavailability in humans. For this reason and for the protection of consumers' health, the binding properties of potassium polyaspartate versus three minerals (calcium, iron, magnesium) was assayed using the Size-Exclusion Chromatography (SEC). Aliquots obtained by SEC were analysed for their content in polyaspartate (by microbiuret method) and bound minerals (by ICP-OES method). The results obtained by this research shows that, when potassium polyaspartate is added to wine, the negative charges of the additive are saturated, as expected by the specific role of KPAA in tartaric stabilization. In conclusions, the effect on mineral bioavailability must be considered negligible

    Out-of-equilibrium physics in driven dissipative coupled resonator arrays

    Coupled resonator arrays have been shown to exhibit interesting many- body physics including Mott and Fractional Hall states of photons. One of the main differences between these photonic quantum simulators and their cold atoms coun- terparts is in the dissipative nature of their photonic excitations. The natural equi- librium state is where there are no photons left in the cavity. Pumping the system with external drives is therefore necessary to compensate for the losses and realise non-trivial states. The external driving here can easily be tuned to be incoherent, coherent or fully quantum, opening the road for exploration of many body regimes beyond the reach of other approaches. In this chapter, we review some of the physics arising in driven dissipative coupled resonator arrays including photon fermionisa- tion, crystallisation, as well as photonic quantum Hall physics out of equilibrium. We start by briefly describing possible experimental candidates to realise coupled resonator arrays along with the two theoretical models that capture their physics, the Jaynes-Cummings-Hubbard and Bose-Hubbard Hamiltonians.
