    Precursors of extreme increments

    We investigate precursors and predictability of extreme increments in a time series. The events we are focusing on consist in large increments within successive time steps. We are especially interested in understanding how the quality of the predictions depends on the strategy to choose precursors, on the size of the event and on the correlation strength. We study the prediction of extreme increments analytically in an AR(1) process, and numerically in wind speed recordings and long-range correlated ARMA data. We evaluate the success of predictions via receiver operator characteristics (ROC-curves). Furthermore, we observe an increase of the quality of predictions with increasing event size and with decreasing correlation in all examples. Both effects can be understood by using the likelihood ratio as a summary index for smooth ROC-curves

    Effect of mesograzer and nutrient levels on the induction of defenses in several Brazilian acroalgae

    Herbivory can greatly modify benthic community structure by affecting the distribution of algal species. To deter herbivores, algae have developed several mechanisms, including the induction of chemical and morphological defenses, which may be influenced by nutrient availability. We tested 4 red (Chondrophycus flagellifera, Cryptonemia seminervis, Osmundaria obtusiloba, Pterocladiella capillacea), 4 brown (Dictyota menstrualis, Lobophora variegata, Sargassum vulgare, Stypopodium zonale), and 1 green (Codium decorticatum) algae for inducible defenses following exposure to direct consumption by an amphipod community dominated by Elasmopus brasiliensis. In addition, the effects of water-borne cues from nearby grazed conspecifics and non-grazing consumers on the induction of defenses were examined in C. decorticatum under natural and enhanced (200% natural) nutrient levels. Induction of defense was assessed in choice-feeding assays, using live algae or artificial food containing non-polar extracts of amphipod-exposed (treated) and non-exposed (control) algae. Palatability levels, estimated as the relative difference in wet mass due to consumption in feeding assays between grazer-exposed and control plants, declined significantly in 3 species after the acclimatization period. Tissue from the directly consumed red alga P. capillacea (live alga) was significantly less palatable than tissue from the control plants. Likewise, a significant effect was observed in the brown alga L. variegata. Similar, although not statistically significant, trends were observed in 6 other species. For the green alga C. decorticatum, nutrient enrichment did not affect induction of defenses by herbivores, yet unfertilized plants were more palatable than fertilized conspecifics

    Multifrequency Strategies for the Identification of Gamma-Ray Sources

    More than half the sources in the Third EGRET (3EG) catalog have no firmly established counterparts at other wavelengths and are unidentified. Some of these unidentified sources have remained a mystery since the first surveys of the gamma-ray sky with the COS-B satellite. The unidentified sources generally have large error circles, and finding counterparts has often been a challenging job. A multiwavelength approach, using X-ray, optical, and radio data, is often needed to understand the nature of these sources. This chapter reviews the technique of identification of EGRET sources using multiwavelength studies of the gamma-ray fields.Comment: 35 pages, 22 figures.     Protein Structure along the Order–Disorder Continuum

    Thermal fluctuations cause proteins to adopt an ensemble of conformations wherein the relative stability of the different ensemble members is determined by the topography of the underlying energy landscape. “Folded” proteins have relatively homogeneous ensembles, while “unfolded” proteins have heterogeneous ensembles. Hence, the labels “folded” and “unfolded” represent attempts to provide a qualitative characterization of the extent of structural heterogeneity within the underlying ensemble. In this work, we introduce an information-theoretic order parameter to quantify this conformational heterogeneity. We demonstrate that this order parameter can be estimated in a straightforward manner from an ensemble and is applicable to both unfolded and folded proteins. In addition, a simple formula for approximating the order parameter directly from crystallographic B factors is presented. By applying these metrics to a large sample of proteins, we show that proteins span the full range of the order–disorder axis.National Institutes of Health (U.S.) (NIH Grant 5R21NS063185-02

    Intramolecular Cohesion of Coils Mediated by Phenylalanine–Glycine Motifs in the Natively Unfolded Domain of a Nucleoporin

    The nuclear pore complex (NPC) provides the sole aqueous conduit for macromolecular exchange between the nucleus and the cytoplasm of cells. Its diffusion conduit contains a size-selective gate formed by a family of NPC proteins that feature large, natively unfolded domains with phenylalanine–glycine repeats (FG domains). These domains of nucleoporins play key roles in establishing the NPC permeability barrier, but little is known about their dynamic structure. Here we used molecular modeling and biophysical techniques to characterize the dynamic ensemble of structures of a representative FG domain from the yeast nucleoporin Nup116. The results showed that its FG motifs function as intramolecular cohesion elements that impart order to the FG domain and compact its ensemble of structures into native premolten globular configurations. At the NPC, the FG motifs of nucleoporins may exert this cohesive effect intermolecularly as well as intramolecularly to form a malleable yet cohesive quaternary structure composed of highly flexible polypeptide chains. Dynamic shifts in the equilibrium or competition between intra- and intermolecular FG motif interactions could facilitate the rapid and reversible structural transitions at the NPC conduit needed to accommodate passing karyopherin–cargo complexes of various shapes and sizes while simultaneously maintaining a size-selective gate against protein diffusion
