    Investigation of extruded cereals enriched with plant by-products and their use in fermented beverage production

    ArticleThe aim of the study was to analyse the quality of extruded cereals enriched with plant by-products and to obtain fermented drinks from production rejects. Extrusion was performed with co-rotating twin-screw extruder (compression ratio 8:1) at MILZU Ltd. from rye and oat flour (80:20, control samples) with addition of apple (ABF), carrot (CBF) and pumpkin (PBF) by-product flour in various amounts (10%, 15% and 20%). Naturally fermented kvass production process was used for non-alcoholic fermented beverage production. Total dietary fibre (TDF), textural properties and sensory features of extruded products after addition of by-products (BP) were determined. Dry matter, active acidity and sensory properties were analysed in fermented beverages. The obtained results showed a 12-55% increase in TDF of extruded cereals (11.8 g 100 g -1 ) after addition of plant by-products. All extruded samples with BP showed lower hardness levels than control (35.55 ± 2.95 N); samples with PBF were the least hard (P < 0.05). Samples with the lowest bulk density were obtained by the addition of 10% and 15% PBF, and 15% CBF, whereas addition of apple by-product flour in all tested concentrations gave the samples a higher bulk density compared to control. Highest taste and aftertaste scores using 5-point hedonic scale were given to samples with addition of 15% and 20% ABF, which also showed high consumer acceptance. With regards to fermented drinks, the highest dry matter content was found in PBF and ABF drink, 8.1 ± 0.1 and 7.0 ± 0.1, respectively. Sensory evaluation of fermented beverages showed that the intensity of flavour, acidity and aroma was most pronounced in sample with ABF, whereas colour was most pronounced in sample with PBF.     Farmers’ management of functional biodiversity goes beyond pest management in organic European apple orchards

    Supporting functional biodiversity (FB), which provides natural pest regulation, is an environmentally sound and promising approach to reduce pesticide use in perennial cultures such as apple, especially in organic farming. However, little is known about farmers’ practices and motivations to implement techniques that favor FB, especially whether or not they really expect anything from FB in terms of pest regulation. In fact, FB-supporting techniques (FB-techniques) are massively questioned by practitioners due to inadequate information about their effectiveness. An interview survey was performed in eight European countries(i) to describe farmers’ practices and identify promising FB-techniques: (ii) to better understand their perceptions of and values associated with FB; and (iii) to identify potential drivers of (non-)adoption. Fifty-five advisors and 125 orchard managers with various degrees of experience and convictions about FB were interviewed and a total of 24 different FB-techniques which can be assigned to three different categories (ecological infrastructures, farming practices and redesign techniques) were described. Some were well-established measures (e.g., hedges and bird houses), while others were more marginal and more recent (e.g., animal introduction and compost). On average, farmers combined more than four techniques that had been implemented over a period of 13 years, especially during their establishment or conversion period. In general, it was difficult for farmers to evaluate the effectiveness of individual FB-techniques on pest regulation. They considered FB-techniques as a whole, targeting multiple species, and valued multiple ecosystem services in addition to pest regulation. The techniques implemented and their associated values differed among farmers who adopted various approaches towards FB. Three different approaches were defined: passive, active and integrated. Their appraisal of FB is even more complex because it may change with time and experience. These findings provide empirical evidence that the practical implementation of promising techniques remains a challenge, considering the diversity of situations and evaluation criteria. Increased cooperation between researchers, farmers and advisors should more effectively target research, advisory support and communication to meet farmers’ needs and perceptions

    Polymorphisms on PAI-1 and ACE genes in association with fibrinolytic bleeding after on-pump cardiac surgery

    Publisher Copyright: © 2015 Ozolina et al.Background: Carriers of plasminogen activator inhibitor -1 (PAI-1) -675 genotype 5G/5G may be associated with lower preoperative PAI-1 plasma levels and higher blood loss after heart surgery using cardiopulmonary bypass (CPB). We speculate if polymorphisms of PAI-1 -844 A/G and angiotensin converting enzyme (ACE) intron 16 I/D also might promote fibrinolysis and increase postoperative bleeding. Methods: We assessed PAI-1 -844 A/G, and ACE intron 16 I/D polymorphisms by polymerase chain reaction technique and direct sequencing of genomic DNA from 83 open heart surgery patients that we have presented earlier. As primary outcome, accumulated chest tube drainage (CTD) at 4 and 24 h were analyzed for association with genetic polymorphisms. As secondary outcome, differences in plasma levels of PAI-1, t-PA/PAI-1 complex and D-dimer were determined for each polymorphism. SPSS® was used for statistical evaluation. Results: The lowest preoperative PAI-1 plasma levels were associated with PAI-1 -844 genotype G/G, and higher CTD, as compared with genotype A/A at 4 and 24 h after surgery. Correspondingly, 4 h after the surgery CTD was higher in carriers of ACE intron 16 genotype I/I, as compared with genotype D/D. PAI-1 plasma levels and t-PA/PAI-1 complex reached nadir in carriers of ACE intron 16 genotype I/I, in whom we also noticed the highest D-dimer levels immediately after surgery. Notably, carriers of PAI-1 -844 genotype G/G displayed higher D-dimer levels at 24 h after surgery as compared with those of genotype A/G. Conclusions: Increased postoperative blood loss secondary to enhanced fibrinolysis was associated with carriers of PAI-1 -844 G/G and ACE Intron 16 I/I, suggesting that these genotypes might predict increased postoperative blood loss after cardiac surgery using CPB.publishersversionPeer reviewe

    The tree that hides the forest: Cryptic diversity and phylogenetic relationships in the Palaearctic vector Obsoletus/Scoticus Complex (Diptera: Ceratopogonidae) at the European level

    Background: Culicoides obsoletus is an abundant and widely distributed Holarctic biting midge species, involved in the transmission of bluetongue virus (BTV) and Schmallenberg virus (SBV) to wild and domestic ruminants. Females of this vector species are often reported jointly with two morphologically very close species, C. scoticus and C. montanus, forming the Obsoletus/Scoticus Complex. Recently, cryptic diversity within C. obsoletus was reported in geographically distant sites. Clear delineation of species and characterization of genetic variability is mandatory to revise their taxonomic status and assess the vector role of each taxonomic entity. Our objectives were to characterize and map the cryptic diversity within the Obsoletus/Scoticus Complex. Methods: Portion of the cox1 mitochondrial gene of 3763 individuals belonging to the Obsoletus/Scoticus Complex was sequenced. Populations from 20 countries along a Palaearctic Mediterranean transect covering Scandinavia to Canary islands (North to South) and Canary islands to Turkey (West to East) were included. Genetic diversity based on cox1 barcoding was supported by 16S rDNA mitochondrial gene sequences and a gene coding for ribosomal 28S rDNA. Species delimitation using a multi-marker methodology was used to revise the current taxonomic scheme of the Obsoletus/Scoticus Complex. Results: Our analysis showed the existence of three phylogenetic clades (C. obsoletus clade O2, C. obsoletus clade dark and one not yet named and identified) within C. obsoletus. These analyses also revealed two intra-specific clades within C. scoticus and raised questions about the taxonomic status of C. montanus. Conclusions: To our knowledge, our study provides the first genetic characterization of the Obsoletus/Scoticus Complex on a large geographical scale and allows a revision of the current taxonomic classification for an important group of vector species of livestock viruses in the Palaearctic region.[Figure not available: See fulltext.

    Towards combined multispectral, FLIM and Raman imaging for skin diagnostics

    To explore challenges for further improvement of diagnostic performance, a project aimed at development of technology for tri-modal skin imaging by combining multispectral, fluorescence lifetime and Raman band imaging was initiated. In this study, each of the mentioned imaging modalities has been preliminary tested and updated. Four different multispectral imaging devices were tested on color standards. Picosecond laser-excited fluorescence lifetime imaging equipment was examined on ex-vivo skin samples. Finally, a new Raman spectroscopy setup with 785 nm laser was launched and tested on cell cultures and ex-vivo skin.
