Forelimb muscle and joint actions in Archosauria: insights from Crocodylus johnstoni (Pseudosuchia) and Mussaurus patagonicus (Sauropodomorpha)
Many of the major locomotor transitions during the evolution of Archosauria, the lineage including crocodiles and birds as well as extinct Dinosauria, were shifts from quadrupedalism to bipedalism (and vice versa). Those occurred within a continuum between more sprawling and erect modes of locomotion and involved drastic changes of limb anatomy and function in several lineages, including sauropodomorph dinosaurs. We present biomechanical computer models of two locomotor extremes within Archosauria in an analysis of joint ranges of motion and the moment arms of the major forelimb muscles in order to quantify biomechanical differences between more sprawling, pseudosuchian (represented the crocodile Crocodylus johnstoni) and more erect, dinosaurian (represented by the sauropodomorph Mussaurus patagonicus) modes of forelimb function. We compare these two locomotor extremes in terms of the reconstructed musculoskeletal anatomy, ranges of motion of the forelimb joints and the moment arm patterns of muscles across those ranges of joint motion. We reconstructed the three-dimensional paths of 30 muscles acting around the shoulder, elbow and wrist joints. We explicitly evaluate how forelimb joint mobility and muscle actions may have changed with postural and anatomical alterations from basal archosaurs to early sauropodomorphs. We thus evaluate in which ways forelimb posture was correlated with muscle leverage, and how such differences fit into a broader evolutionary context (i.e. transition from sprawling quadrupedalism to erect bipedalism and then shifting to graviportal quadrupedalism). Our analysis reveals major differences of muscle actions between the more sprawling and erect models at the shoulder joint. These differences are related not only to the articular surfaces but also to the orientation of the scapula, in which extension/flexion movements in Crocodylus (e.g. protraction of the humerus) correspond to elevation/depression in Mussaurus. Muscle action is highly influenced by limb posture, more so than morphology. Habitual quadrupedalism in Mussaurus is not supported by our analysis of joint range of motion, which indicates that glenohumeral protraction was severely restricted. Additionally, some active pronation of the manus may have been possible in Mussaurus, allowing semi-pronation by a rearranging of the whole antebrachium (not the radius against the ulna, as previously thought) via long-axis rotation at the elbow joint. However, the muscles acting around this joint to actively pronate it may have been too weak to drive or maintain such orientations as opposed to a neutral position in between pronation and supination. Regardless, the origin of quadrupedalism in Sauropoda is not only linked to manus pronation but also to multiple shifts of forelimb morphology, allowing greater flexion movements of the glenohumeral joint and a more columnar forelimb posture
Analytic approximation of solutions of parabolic partial differential equations with variable coefficients
A complete family of solutions for the one-dimensional reaction-diffusion
equation with a coefficient
depending on is constructed. The solutions represent the images of the heat
polynomials under the action of a transmutation operator. Their use allows one
to obtain an explicit solution of the noncharacteristic Cauchy problem for the
considered equation with sufficiently regular Cauchy data as well as to solve
numerically initial boundary value problems. In the paper the Dirichlet
boundary conditions are considered however the proposed method can be easily
extended onto other standard boundary conditions. The proposed numerical method

Authentic science activities in the primary level classroom: Investigating the effects of a data collection and analysis interface on primary level students’ scientific literacy
This study explores the use of technology in instruction in a primary level classroom. Though many researchers and educators advocate for the incorporation of technology into science instruction, little is known about the effects of the implementation of various types of educational technology. Here, we investigate the effects of the incorporation of a data collection and analysis interface, the SparkTM, on the scientific literacy of a group of second grade students in the United States. Treatment and control group comparisons indicate that the use of this technology during a lesson designed to investigate the relationships between physical variables increases the likelihood that students will represent their ideas graphically
Role of critical spin fluctuations in ultrafast demagnetization of transition-metal rare-earth alloys
Ultrafast magnetization dynamics induced by femtosecond laser pulses have
been measured in ferrimagnetic Co0.8Gd0.2, Co.74Tb.26 and Co.86Tb.14 alloys.
Using element sensitivity of X-ray magnetic circular dichroism at the Co L3, Tb
M5 and Gd M5 edges we evidence that the demagnetization dynamics is element
dependent. We show that a thermalization time as fast as 280 fs is observed for
the rare-earth in the alloy, when the laser excited state temperature is below
the compensation temperature. It is limited to 500 fs when the laser excited
state temperature is below the Curie temperature (Tc). We propose critical spin
fluctuations in the vicinity of TC as the mechanism which reduces the
demagnetization rates of the 4f electrons in transition-metal rare-earth alloys
whereas at any different temperature the limited demagnetization rates could be

Temporal and phylogenetic evolution of the sauropod dinosaur body plan
The colossal size and body plan of sauropod dinosaurs are unparalleled in terrestrial vertebrates. However, to date, there have been only limited attempts to examine temporal and phylogenetic patterns in the sauropod bauplan. Here, we combine three-dimensional computational models with phylogenetic reconstructions to quantify the evolution of whole-body shape and body segment properties across the sauropod radiation. Limitations associated with the absence of soft tissue preservation in fossils result in large error bars about mean absolute body shape predictions. However, applying any consistent skeleton : body volume ratio to all taxa does yield changes in body shape that appear concurrent with major macroevolutionary events in sauropod history. A caudad shift in centre-of-mass (CoM) in Middle Triassic Saurischia, associated with the evolution of bipedalism in various dinosaur lineages, was reversed in Late Triassic sauropodomorphs. A craniad CoM shift coincided with the evolution of quadrupedalism in the Late Triassic, followed by a more striking craniad shift in Late Jurassic–Cretaceous titanosauriforms, which included the largest sauropods. These craniad CoM shifts are strongly correlated with neck enlargement, a key innovation in sauropod evolution and pivotal to their gigantism. By creating a much larger feeding envelope, neck elongation is thought to have increased feeding efficiency and opened up trophic niches that were inaccessible to other herbivores. However, we find that relative neck size and CoM position are not strongly correlated with inferred feeding habits. Instead the craniad CoM positions of titanosauriforms appear closely linked with locomotion and environmental distributions, potentially contributing to the continued success of this group until the end-Cretaceous, with all other sauropods having gone extinct by the early Late Cretaceous
Onco-miR-155 targets SHIP1 to promote TNFalpha-dependent growth of B cell lymphomas.
Non-coding microRNAs (miRs) are a vital component of post-transcriptional modulation of protein expression and, like coding mRNAs harbour oncogenic properties. However, the mechanisms governing miR expression and the identity of the affected transcripts remain poorly understood. Here we identify the inositol phosphatase SHIP1 as a bonafide target of the oncogenic miR-155. We demonstrate that in diffuse large B cell lymphoma (DLBCL) elevated levels of miR-155, and consequent diminished SHIP1 expression are the result of autocrine stimulation by the pro-inflammatory cytokine tumour necrosis factor a (TNFalpha). Anti-TNFalpha regimen such as eternacept or infliximab were sufficient to reduce miR-155 levels and restored SHIP1 expression in DLBCL cells with an accompanying reduction in cell proliferation. Furthermore, we observed a substantial decrease in tumour burden in DLBCL xenografts in response to eternacept. These findings strongly support the concept that cytokine-regulated miRs can function as a crucial link between inflammation and cancer, and illustrate the feasibility of anti-TNFalpha therapy as a novel and immediately accessible (co)treatment for DLBCL
Thioredoxin and glutathione systems differ in parasitic and free-living platyhelminths
<p>Abstract</p> <p>Background</p> <p>The thioredoxin and/or glutathione pathways occur in all organisms. They provide electrons for deoxyribonucleotide synthesis, function as antioxidant defenses, in detoxification, Fe/S biogenesis and participate in a variety of cellular processes. In contrast to their mammalian hosts, platyhelminth (flatworm) parasites studied so far, lack conventional thioredoxin and glutathione systems. Instead, they possess a linked thioredoxin-glutathione system with the selenocysteine-containing enzyme thioredoxin glutathione reductase (TGR) as the single redox hub that controls the overall redox homeostasis. TGR has been recently validated as a drug target for schistosomiasis and new drug leads targeting TGR have recently been identified for these platyhelminth infections that affect more than 200 million people and for which a single drug is currently available. Little is known regarding the genomic structure of flatworm TGRs, the expression of TGR variants and whether the absence of conventional thioredoxin and glutathione systems is a signature of the entire platyhelminth phylum.</p> <p>Results</p> <p>We examine platyhelminth genomes and transcriptomes and find that all platyhelminth parasites (from classes Cestoda and Trematoda) conform to a biochemical scenario involving, exclusively, a selenium-dependent linked thioredoxin-glutathione system having TGR as a central redox hub. In contrast, the free-living platyhelminth <it>Schmidtea mediterranea </it>(Class Turbellaria) possesses conventional and linked thioredoxin and glutathione systems. We identify TGR variants in <it>Schistosoma </it>spp. derived from a single gene, and demonstrate their expression. We also provide experimental evidence that alternative initiation of transcription and alternative transcript processing contribute to the generation of TGR variants in platyhelminth parasites.</p> <p>Conclusions</p> <p>Our results indicate that thioredoxin and glutathione pathways differ in parasitic and free-living flatworms and that canonical enzymes were specifically lost in the parasitic lineage. Platyhelminth parasites possess a unique and simplified redox system for diverse essential processes, and thus TGR is an excellent drug target for platyhelminth infections. Inhibition of the central redox wire hub would lead to overall disruption of redox homeostasis and disable DNA synthesis.</p
Variations in AXIN2 predict risk and prognosis of colorectal cancer
Objective: Colorectal cancer (CRC) and hypodontia are frequent and different diseases with common genes are involved in their etiology. The objective of this study was to identify the association between AXIN2 rs2240308 with hypodontia and CRC. Patients and methods: This study consisted of 50 individuals with hypodontia, 50 individuals with CRC, and 155 healthy individuals from Colombia. SNP genotyping assays of rs2240308 were performed and family history of cancer in individuals with hypodontia was documented. In silico analysis was implemented to define the genomic profile of the AXIN2 gene associated with CRC. Multivariate analysis, chi square, odd ratio tests, and R software were used for statistical analysis. Results: AXIN2 rs2240308 showed association with CRC (OR = 5.4 CI: 2.7–10.4; p < 0.001) and with other familial cancer in individuals with hypodontia (p < 0.005 OR = 1.75, 95% CI: 1.22–6.91). In silico analysis showed that variations in AXIN2 found in CRC patients, were more frequently in earlier stages of tumor and patients who carry variations in the AXIN2 gene have a worse prognosis (p < 0.05). The association between AXIN2 rs2240308 with hypodontia was not significant. Conclusions: These results suggest that AXIN2 rs2240308 polymorphism is associated with CRC and AXIN2 could be a risk marker for predisposition and prognosis of CRC.Fil: Otero, L.. Desenvolvimento urbano contemporâneo em cidades médias do interior paulista sob o estatuto da cidade: uma análise comparada de Piracicaba, Bauru e Rio Claro
O artigo traça um quadro do desenvolvimento urbano contemporâneo em cidades médias do interior do
estado de São Paulo (Brasil), sob três dimensões complementares: econômica, urbanÃstica e polÃticoinstitucional.
Para tanto, indica os processos interdependentes que estruturam o desenvolvimento urbano
com base na análise comparativa desses municÃpios (Piracicaba, Bauru e Rio Claro). Nesses termos,
identifica como essas cidades se inseriram nos processos de transformação da economia nacional nas
décadas de 1970-90, e como essa trajetória impactou suas dinâmicas urbano-regionais. Observa-se de que
maneira as determinantes econômicas – tanto as vinculadas às mudanças no padrão locacional das
indústrias quanto outras associadas à dinâmica imobiliária – impactaram o ordenamento urbanÃstico e como
ocorreram tais processos decisórios. A análise se faz no contexto institucional da promulgação e vigência
do Estatuto da Cidade, no qual os municÃpios refizeram seus Planos Diretores e passaram a contar com
novos meios de regular a produção do espaço urbano.The article focuses on contemporary urban development in medium-sized cities in the state of São Paulo
(Brazil), under three complementary dimensions: economic, urban and political-institutional. Thus, it indicates
the interdependent processes that shape urban development based on comparative analysis of these
municipalities (Piracicaba, Bauru and Rio Claro). In these terms it identifies how these cities were inserted in
the processes of transformation of the national economy in the decades of 1970-90, and how that path
impacted their urban-regional dynamics. We observe how the economic determinants – changes in the
industry standard locational and dynamic real estate – impacted the urban planning and decision-making
processes. The analysis is done in the context of institutional effectiveness and promulgation enactment of
the Statute of the City, when municipalities remade their Master Plans and have acquired new ways to
